<P>Basal type breast cancer is the most aggressive and has mesenchymal features with a high metastatic ability. However, the signaling node that determines the basal type features in breast cancer remains obscure. Here, we report that FYN among SRC family kinases is required for the maintenance of basal type breast cancer subtype. Importantly, FYN enhanced NOTCH2 activation in basal type breast cancer cells through STAT5-mediated upregulation of Jagged-1 and DLL4 NOTCH ligands, thereby contributed to mesenchymal phenotypes. In addition, we found that high levels of FYN persist in basal type breast cancer cells by a positive feedback loop between FYN and STAT5. FYN interacted directly with STAT5 and increased p-STAT5 that further acts as a transcription factor for FYN. Taken together, our findings demonstrate a pivotal role of FYN and its downstream effectors in maintaining the basal type features in breast cancer.</P>

Background: Although the pulse pressure (PP) is an independent predictor of coronary heart disease (CHD), many physicians disregard its importance and there have been no recent studies on Korean elderly. We investigated the association between PP and CHD in Korean elderly through the nationwide, observational study. Methods: This study included 4100 subjects aged ≥65 years from the 7th Korean National Health and Nutrition Examination Survey (2016-2018). Pulse pressure was calculated as the difference between the systolic blood pressure (SBP) and the diastolic blood pressure (DBP), and high PP was defined as ≥60 mmHg. Subjects diagnosed with angina pectoris or myocardial infarction were classified into the CHD group. Multivariate logistic regression analysis was performed to evaluate the association between PP and CHD. Results: Among the subjects, the incidence of high PP was higher in women, while CHD was higher in men. Subjects with high PP were older, had higher SBP, lower DBP and higher FBG compared to subjects with normal PP. After adjustment for multiple covariates, participants with high PP had a higher risk for incidence of CHD with an OR of 1.361 (95% of CI: 1.077-1.719). Conclusions: In this study, we found the significant association between PP and CHD in Korean elderly.

<P>Cells have evolved sophisticated mechanisms to maintain genomic integrity in response to DNA damage. Ionizing radiation (IR)–induced DNA damage results in the formation of IR-induced foci (iRIF) in the nucleus. The iRIF formation is part of the DNA damage response (DDR), which is an essential signaling cascade that must be strictly regulated because either the loss of or an augmented DDR leads to loss of genome integrity. Accordingly, negative regulation of the DDR is as critical as its activation. In this study, we have identified ring finger protein 126 (RNF126) as a negative regulator of the DDR from a screen of iRIF containing 53BP1. RNF126 overexpression abolishes not only the formation of 53BP1 iRIF but also of RNF168, FK2, RAP80, and BRCA1. However, the iRIF formation of γH2AX, MDC1, and RNF8 is maintained, indicating that RNF126 acts between RNF8 and RNF168 during the DDR. In addition, RNF126 overexpression consistently results in the loss of RNF168-mediated H2A monoubiquitination at lysine 13/15 and inhibition of the non-homologous end joining capability. Taken together, our findings reveal that RNF126 is a novel factor involved in the negative regulation of DDR, which is important for sustaining genomic integrity.</P>

<P>Metabolite profiling of four different-sized Aloe vera plants was performed using gas chromatography–ion trap–mass spectrometry (GC-IT-MS) and ultra performance liquid chromatography–quadrupole–time of flight–mass spectrometry (UPLC-Q-TOF-MS) with multivariate analysis. Amino acids, sugars, and organic acids related to growth and development were identified by sizes. In particular, the relative contents of glucose, fructose, alanine, valine, and aspartic acid increased gradually as the size of the aloe increased. Anthraquinone derivatives such as 7-hydroxy-8-<I>O</I>-methylaloin, 7-hydroxyaloin A, and 6′-malonylnataloins A and B increased gradually, whereas chromone derivatives decreased continuously as the size of the aloe increased. The A30 aloe (size = 20–30 cm) with relatively high contents of aloins A and B, was suggested to have antioxidant components showing the highest antioxidant activity among the four different sizes of aloe. Objective The primary objective of this study was to investigate the effect of methylphenidate (MPH) on height, weight, and body mass index (BMI) in drug-naive children and adolescents with attention deficit hyperactivity disorder (ADHD) over 24 months. The secondary objective was to investigate whether the age of MPH initiation and sex act as risk factors for growth retardation.Methods A total of 82 patients with ADHD were included. Weight, height, and BMI were measured at baseline and every 6 months up to 24 months. Weight, height, and BMI data were converted to z-scores and analyzed using two-way repeated-measures ANOVA and multiple linear regression.Results The z-score of height, weight and BMI decreased from the baseline values. The z-scores of height were at baseline 0.002; 6 months -0.100; 12 months -0.159; 18 months -0.159; 24 months -0.186. The z-scores of weight were at baseline 0.104; 6 months -0.155; 12 months -0.256; 18 months -0.278; 24 months -0.301. Here were no age and sex differences of height, weight, and BMI.Conclusion The use of MPH was associated with attenuation of weight and height gain rates in children and adolescents with ADHD.

A wiper is a safety device removing rain and debris from windshield and ensuring visibility of drivers. If contact pressure distribution between rubber of the blade and the windshield is unbalanced, unwanted noise, vibration, and abrasion of the blade can occur and sometimes fatal accidents could occur. To improve the safety of the wiper, there have been many researches on the contact pressure analysis of the wiper, but the analysis results were not converged or require much computational time due to material nonlinearity of the rubber and contact conditions between the blade rubber and the windshield. In this research, a simple model with 1D beam and 2D shell elements was used for the contact pressure analysis instead of the 3D blade model. The simplified model saved computational time of the analysis and resolved convergence problems. The accuracy of the analysis results was verified by comparing them with experimental results for different rail spring curvatures.

고압연료펌프는 GDI 엔진의 핵심 구성요소로써, 엔진출력 및 연료 효율을 향상시키기 위해서는 고압연료펌프의 유동특성을 연구하는 것이 필요하다. 본 연구에서는 유압해석툴인 AMEsim을 이용하여 고압연료펌프의 통합 모델을 생성하여 유동해석을 수행하였다. 하지만, AMEsim은 시스템 해석을 위한 1차원 모델이므로 복잡한 유동현상이 발생하는 부근에서의 해석 결과는 정확하지 않은 단점이 있으므로 본 연구에서는 전산해석프로그램인 Fluent를 이용하여 난류유동이 발생하는 체크밸브의 흡입부와 토출부에서 유량과 알짜힘을 계산하였다. 다양한 압력조건과 밸브 간극변화에 따른 CFD 해석 결과는 AMEsim모델에 대한 룩업테이블로 사용되어 AMEsim의 결과를 보완함으로써 고압연료펌프에 대한 성능분석결과의 정확성을 향상시키는 결과를 얻을 수 있었다. A high-pressure fuel pump is a key component in a gasoline direct injection (GDI) engine; thus, understanding its flow characteristics is essential for improving the engine power and fuel efficiency. In this study, AMESim, which is a hydraulic analysis program, was used to analyze the performance of the high-pressure fuel pump. However, since AMESim uses a one-dimensional model for the system analysis, it does not accurately analyze the complicated flow characteristics. Thus, Fluent, computational fluid dynamics (CFD) software, was used to calculate the flow rates and net forces at the intake and discharge ports of the high-pressure fuel pump where turbulent flow occurs. The CFD analysis results for various pressure conditions and valve lifts were used as look-up tables for the AMEsim model. The CFD analysis results complemented the AMEsim results, and thus, improved the accuracy of the performance analysis results for the high-pressure fuel pump.
