표고버섯 균사체 배양기간에 따른 PDB 배지 내 유리 아미노산 변화

This study was carried out to investigate the changes in the amount of 20 free amino acids in PDB (Potato Dextrose Broth) medium during the incubation period while cultivating Lentinula edodes. The total incubation period was 90 days, and the total amount of free amino acids was confirmed every 45 days. Among the 18 kinds of amino acids whose increase and decrease patterns were confirmed, 10 amino acids were increased compared to that in the control, among which cysteine increased from its initial value of 9,889 ± 3 μg/L to 12,909 ± 2 μg/L at 45 days and 29,256 ± 4 μg/L at 90 days. Six amino acids with decreased expression patterns were identified. Arginine decreased to 83,751 ± 2 μg/L after 45 days from its initial value of 161,787 ± 1 μg/L and to 79,055 ± 7 μg/L at 90 days.
전로슬래그 및 페로니켈슬래그를 혼입한 모르타르의 기초물성 연구

본 연구에서는 건설 산업의 고도화에 따라 대량 발생하는 철강 슬래그 및 페로니켈슬래그의 활용을 위해 모르타르 배합에 혼입하여 실험적 연구를 진행하였다. 물의 흡수율이 낮은 BOF와 표면이 매끄러운 성질을 가진 FNS를 시멘트에 치환하면, 희석효과(dilution effect) 작용으로 플로 값과 응결시간이 증가하였다. 다만, BOF를 표준사 대비 10% 초과하여 혼입할 경우 재료 분리 현상(segregation)이 발생하였고, 이에 다량의 혼입 배합은 실험에서 제외하였다. BOF 잔골재와 FNS 혼입 모르타르는 응결 지연으로 인한 수화열 감소로 종결이 완료된 후 길이변화가 발생하지 않았다. BOF 잔골재를 혼입한 모르타르의 압축강도는 표준사와 시멘트만을 혼입한 모르타르 강도 보다 감소되었지만 FNS와 함께 혼입한 배합의 경우 양생 일이 증가함에 따라 압축강도도 증가하였다. BOF 잔골재를 혼입한 B10F0 및 B10F20 모르타르에서는 수화가 진행되어 BOF 원재료 XRD에서 관찰할 수 있었던 larnite, mayenite, wuestite 클링커는 거의 관찰되지 않았지만, FNS의 낮은 수화 반응성으로 FNS의 클링커는 관찰되었다. 주사전자현미경 분석 결과 수화결정체로 존재하지 않고 수화가 진행 중인 FNS를 확인할 수 있었으며 이를 통해 FNS의 잠재수경성을 확인하였다. FNS를 첨가하지 않은 시편의 경우 BOF 골재가 겔이나 침상결정이 아닌 전체가 괴상으로 존재하였으며, 인산화칼슘(calcium phosphate) 형성을 확인하였다. 다만, 전로슬래그를 혼입할 경우 내부 공극은 밀실함이 다소 저하되었으며, 추후 BOF를 잔골재 또는 건설 재료로 활용할 경우 적정 배합비 선정이 필요할 것으로 판단된다. Converter steel slag(BOF slag) is a vast amount of solid waste generated in the steelmaking process which has very low utilization rate in Korea. Due to the presence of free CaO which can derive bad volume stability in BOF slag, it usually land filled. For recycling BOF and identify its applicability as fine aggregate, this study investigates the fundamental characteristics of mortar with cement replaced ferronickel slag(FNS), which has the potential to be used as a binder. The results suggest that the mineral phases of BOF slag mainly include larnite(CaSiO₄), mayenite(Ca12Al14O33) and wuestite(FeO) while olivine crystallines are shown in FNS. The results of flow and setting time reveals that the flowability and process of hardening increased when the amount of FNS and BOF slag incorporated was increased. The length change shows that the amount of change in the length of the mortar was almost constant regardless of mix proportion while compressive strength was reduced. Micro structure test results revealed that FNS or/and BOF slag mix took a long time to react in the cement matrix to form a complete hydration products. To achieve the efficient utilization of BOF slag as construction materials, proper replacement rate is necessary.

Korean bell is famous for its peculiar acoustic property, beat that indicates a periodic repetition of strong and weak sound. Beat occurs when an axisymmetric structure has a slight asymmetry. In this study, we investigate the effect of an artificial dumshoi on the mode pair and beat property. The artificial dumshoi means a thick local element attached on the bell inside. The size of the artificial dumshoi is determined to overwhelm other asymmetry of a Korean bell. By attaching the artificial dumshoi on 22.5 degree from the striking point, best mode pair position is obtained for clear beat. As a result, mode pair equally generates by striking and strong beat is available. The proposed method will be applied to a large size Korean bell in order to make a strong beat.
Dual functions of <i>Nicotiana benthamiana</i> Rae1 in interphase and mitosis

<P>Summary</P><P>Rae1 performs multiple functions in animal systems, acting in interphase as an mRNA export factor and during mitosis as a mitotic checkpoint and spindle assembly regulator. In this study we characterized multiple functions of Rae1 in plants. Virus-induced gene silencing of <I>Nicotiana benthamiana Rae1</I>, <I>NbRae1</I>, which encodes a protein with four WD40 repeats, resulted in growth arrest and abnormal leaf development. NbRae1 was mainly associated with the nuclear envelope during interphase, and NbRae1 deficiency caused accumulation of poly(A) RNA in the nuclei of leaf cells, suggesting defective mRNA export. In the shoot apex, depletion of NbRae1 led to reduced mitotic activities, accompanied by reduced cyclin-dependent kinase (CDK) activity and decreased expression of cyclin B1, CDKB1-1, and histones H3 and H4. The secondary growth of stem vasculature was also inhibited, indicating reduced cambial activities. Differentiated leaf cells of <I>NbRae1</I>-silenced plants exhibited elevated ploidy levels. Immunolabeling in BY-2 cells showed that NbRae1 protein localized to mitotic microtubules and the cell plate-forming zone during mitosis, and recombinant NbRae1 directly bound to microtubules <I>in vitro</I>. Inhibition of <I>NbRae1</I> expression in BY-2 cells using a &bgr;-estradiol-inducible RNAi system resulted in severe defects in spindle organization and chromosome alignment and segregation, which correlated with delays in cell cycle progression. Together, these results suggest that NbRae1 plays a dual role in mRNA export in interphase and in spindle assembly in mitosis.</P>

Increase of the interior noise in the tunnel deteriorates the ride quality and increases the discomfort of the passenger. The increase of the noise level and frequency characteristics depends on the sound insulation performance, train speed and tunnel acoustic environment. For a high speed train with a reasonable sound insulation performance, it is reported that the noise level in the tunnel is 5dB~7dB higher than in the tunnel.(open land) This study compares the noise level and characteristics of ‘Gyeongchun’ electric line in the open land and tunnel. Measurement and frequency analysis on the interior noise is performed in the real operating situation, in order to provide the basic data for the quieter and more comfortable electric line.

We investigated the coupling of the M2 muscarinic acetylcholine receptors expressed in Chinese hamster ovary cells to activation of neuronal nitric oxide(NO) synthase. Stimulation of guanylate cyclase activity in detector neuroblastoma cells was used as an indirect measure of the generation of NO in Chinese hamster ovary cells. The muscarinic agonist carbachol induced marked time-and concentration-dependent enhancement of the activity of NO synthase. Activation of neuronal NO synthase by M2 muscarinic receptors was associated with a small increase in the concentration of intracellular Ca^2+. These data suggest the presence of alternate mechanisms of activation of neuronal NO synthase which might be operative in the absence of large changes in the concentration of cellular Ca^2+. These findings help to understand the mechanisms of activation of NO synthase.
