      • Tic21 is an essential translocon component for protein translocation across the chloroplast inner envelope membrane.

        <P>An Arabidopsis thaliana mutant defective in chloroplast protein import was isolated and the mutant locus, cia5, identified by map-based cloning. CIA5 is a 21-kD integral membrane protein in the chloroplast inner envelope membrane with four predicted transmembrane domains, similar to another potential chloroplast inner membrane protein-conducting channel, At Tic20, and the mitochondrial inner membrane counterparts Tim17, Tim22, and Tim23. cia5 null mutants were albino and accumulated unprocessed precursor proteins. cia5 mutant chloroplasts were normal in targeting and binding of precursors to the chloroplast surface but were defective in protein translocation across the inner envelope membrane. Expression levels of CIA5 were comparable to those of major translocon components, such as At Tic110 and At Toc75, except during germination, at which stage At Tic20 was expressed at its highest level. A double mutant of cia5 At tic20-I had the same phenotype as the At tic20-I single mutant, suggesting that CIA5 and At Tic20 function similarly in chloroplast biogenesis, with At Tic20 functioning earlier in development. We renamed CIA5 as Arabidopsis Tic21 (At Tic21) and propose that it functions as part of the inner membrane protein-conducting channel and may be more important for later stages of leaf development.</P>

        Hypolipidemic and Antioxidative Effects of Glossogyne tenuifolia in Hamsters Fed an Atherogenic Diet

        Glossogyne tenuifolia (GT) Cassini is a special herbal tea in the Penghu Islands, Taiwan, and has a long history of being used as an antipyretic, detoxifying, and anti-inflammatory remedy in folk medicine among local residents. The aim of this study was to investigate the effect of hot water extracts from GT on oxidative stress and lipid metabolism in animals. Five- to 6-week-old male Syrian hamsters were divided into four groups (n = 14) for different treatments, that is: control group (C), high-fat/cholesterol (HF) group, HF diet containing 0.5% (GT0.5) and 1.5% (GT1.5) GT extracts for 4 weeks. Hamsters fed with 0.5% GT powder as well as 1.5% GT powder exhibited reduced serum total cholesterol (TC), conjugated diene of low-density lipoprotein (LDL), and increased serum antioxidant capacity, but 1.5% GT powder was more potent at loweringserum LDL cholesterol and thiobarbituric acid reactive substance concentrations than 0.5% GT. GT extracts significantly lowered liver triacylglycerol (TG) concentration by diminishing activities of fatty acid synthase (FAS) and glucose-6-phosphate dehydrogenase (G-6-PDH). In addition, fecal excretion of cholesterol and bile acids were increased in GT extract groups. In conclusion, GT extracts increase the antioxidative capacity, decrease serum TC, inhibit the activities of FAS and G-6-PDH, and further reduce liver TG accumulation in hamster fed on atherogenic diets.

      • Overexpression and Clinicopathological Contribution of DcR3 in Bladder Urothelial Carcinoma Tissues

        Background: To explore the expression of DcR3 protein and its clinicopathological significance in bladder urothelial carcinomas (BUC). Materials and Methods: Immunohistochemistry was performed to detect the expression of DcR3, caspase-3, Bcl-2, VEGF, Ki-67, PCNA and P53 in 166 BUC and 56 normal bladder tissues. Western blotting was used to detect the expression of DcR3 in the supernatants of cultured BUC cells. Results: Overexpression of DcR3 was found in BUC tissues and cell lines, with significant elevation as compared to normal bladder tissues (p<0.0001). Higher DcR3 expression was related to the status of invasion, lymph node metastasis and recurrence. Furthermore, DcR3 expression was negatively correlated with caspase-3 and positively associated with Bcl-2, VEGF, Ki-67 labeling index (LI), PCNA LI and P53 (all p<0.0001), respectively. Conclusions: DcR3 may play a crucial role as an oncogene in tumorigenesis, deterioration and progress of BUC via influencing related pathways of apoptosis, proliferation and angiogenesis. The detection of DcR3 protein in the formalinfixed and paraffin-embedded samples could assist to predict in prognosis of BUC patients.

        Patient preferences and shared decision making for the traditional Chinese medicine treatment of lung cancer: A discrete choice experiment study

        Background: Traditional Chinese Medicine (TCM), integrating patient preferences into decision-making process, has been widely used in the multimodality therapy of lung cancer. This study aimed to estimate patient preferences for treatment and shared decision-making (SDM) modes concerning inpatient TCM treatment of lung cancer in Shanghai in order to provide a basis for clinical decision-making process on TCM therapy for lung cancer. Methods: This study was conducted among patients (n = 347) from nine tertiary hospitals in Shanghai by discrete-choice experiment (DCE) survey and Shared Decision-Making Questionnaire-patient version (SDM-Q-9) survey. The DCE was developed with the inclusion of the most relevant attributes at appropriate levels for the TCM treatment of lung cancer. The empirical data analyses of patients were performed using mixed logit models. Additionally, subgroup analysis was conducted. Results: In total, 347 respondents completed the questionnaire. All attributes’ coefficients were statistically significant for patients’ preferences. Patients showed strong preferences for increasing disease control rate, relieving nausea and vomiting, reducing the risk of side effects, and were concerned about monthly out-of-pocket costs. Subgroup analysis indicated that patients with a lower SDM-Q-9 score and those who were satisfied with medical services emphasized more importance of higher disease control rate. Furthermore, most of the patients (90.20%) self-reported a high willingness to use SDM during the decision-making process. Conclusion: In Shanghai, patients mainly preferred SDM and considered disease control rate as the most essential attribute in the TCM treatment of lung cancer. The study findings could underscore the importance of considering patients’ preferences and promote SDM.

        Diabetes and Risk of Hepatocellular Carcinoma in Cirrhosis Patients with Nonalcoholic Fatty Liver Disease

        Nonalcoholic fatty liver disease (NAFLD) is the most common cause of chronic liver disease in the world. NAFLD is a hepatic manifestation of insulin resistance, the core pathophysiology of diabetes. Multiple clinical studies show that diabetes increases the risk of liver disease progression and cirrhosis development in patients with NAFLD. Diabetes has causal associations with many different cancers, including hepatocellular carcinoma (HCC). More recent studies demonstrate that diabetes increases the risk of HCC in patients with underlying NAFLD cirrhosis, confirming the direct hepatocarcinogenic effect of diabetes among cirrhosis patients. Diabetes promotes hepatocarcinogenesis via the activation of inflammatory cascades producing reactive oxygen species and proinflammatory cytokines, leading to genomic instability, cellular proliferation, and inhibition of apoptosis. Given the global increase in the burden of NAFLD and HCC, high-risk patients such as older diabetic individuals should be carefully monitored for HCC development. Future larger studies should explore whether the effect of diabetes on HCC risk in NAFLD cirrhosis is modifiable by the type of antidiabetic medication and the effectiveness of diabetes control.

        Development of a performance booster for the evaporator of window-type air conditioners

        This study investigated the effect of a performance booster (PB) on the performance of a window-type air conditioner (WAC). The PB was fabricated using perforated aluminum plates with different pore size configurations according to the return air distribution of evaporator. The WAC was tested under various outdoor air conditions, and the performance of the WAC before and after PB installation was compared. Results showed that the PB effectively improved the performance of the WAC. The PB with three pore sizes improved the WAC performance more than the PB with two pore sizes. The maximum improvements in the WAC’s cooling capacity, dehumidification capacity, and energy efficiency ratio (EER) under the configuration with three pore sizes were 13.70 %, 59.63 % and 14.37 %, respectively, compared with the original WAC at an outdoor dry-bulb temperature of 30 °C.

      • Study on Rotating Compression Forming of Double-layer Clad Ring Considering Coulomb Friction

        This study uses two FEM simulations based on deform 3D and superform commercial software assuming coulomb friction to explore the plastic behavior in rotating compression forming of double-layer bounded clad ring. The rotating compression forming can reduce the compressive force and make the deformation uniform. Effects of the frictional coefficient, shear yield stress ratio, rotating angular velocity, frictional coefficient ratio, clad ring height ratio etc on compression characteristics including the effective stress, the effective strain, the velocity field, the shear strain, the compressive force etc. are investigated effectively. Moreover, the realistic rotating compression experiments of clad ring have been carried out to verify the acceptance of both FEM simulation models.

      • Clinical Prognostic Factors and Survival Outcome in Renal Cell Carcinoma Patients - A Malaysian Single Centre Perspective

        Background: This study concerns clinical characteristics and survival of renal cell carcinoma (RCC) patients in University Malaya Medical Centre (UMMC), as well as the prognostic significance of presenting symptoms. Materials and Methods: The clinical characteristics, presenting symptoms and survival of RCC patients (n=151) treated at UMMC from 2003-2012 were analysed. Symptoms evaluated were macrohaematuria, flank pain, palpable abdominal mass, fever, lethargy, loss of weight, anaemia, elevated ALP, hypoalbuminemia and thrombocytosis. Univariate and multivariate Cox regression analyses were performed to determine the prognostic significance of these presenting symptoms. Kaplan Meier and log rank tests were employed for survival analysis. Results: The 2002 TNM staging was a prognostic factor (p<0.001) but Fuhrman grading was not significantly correlated with survival (p=0.088). At presentation, 76.8% of the patients were symptomatic. Generally, symptomatic tumours had a worse survival prognosis compared to asymptomatic cases (p=0.009; HR 4.74). All symptoms significantly affect disease specific survival except frank haematuria and loin pain on univariate Cox regression analysis. On multivariate analysis adjusted for stage, only clinically palpable abdominal mass remained statistically significant (p=0.027). The mean tumour size of palpable abdominal masses, $9.5{\pm}4.3cm$, was larger than non palpable masses, $5.3{\pm}2.7cm$ (p<0.001). Conclusions: This is the first report which includes survival information of RCC patients from Malaysia. Here the TNM stage and a palpable abdominal mass were independent predictors for survival. Further investigations using a multicentre cohort to analyse mortality and survival rates may aid in improving management of these patients.

        오메가-3 불포화지방산이 노인여성의 심혈관 반응 및 노르에피네프린에 미치는 효과

        It is generally known that Omega-3 polyunsaturated fatty acids (n-3 PUFAs) supplementations decrease risks of sudden death and cardiovascular diseases. A previous study showed that n-3 PUFAs supplements improved stroke volume and peripheral vasoconstriction in the young. On the other hand, in the elderly, systolic blood pressure increases because of arterial stiffness produced by structural alterations of arterial wall occurring with aging. However, the effect of n-3 PUFAs has not been revealed in older humans. Therefore, the purpose of this study was to investigate the effects of hemodynamic responses with n-3 PUFAs supplement at rest and during exercise in elderly women. Subjects (>60 yrs) were divided into only n-3 group (OG), n-3+Placebo group (CG), and placebo group (PG). All subjects completed submaximal exercise testing for 5-7 minutes (60% VO2peak) before and after the nutritional supplementation. Systolic blood pressure (SBP), diastolic blood pressure (DBP), heart rate (HR), stroke volume (SV) were continuously measured from rest through exercise. Blood samples were collected at rest and during exercise. There were no differences in HR and SBP at rest across groups. SBP was significantly decreased in OG (p<.05) and CG (p<.001) during exercise after the n-3 supplement. DBP and mean arterial pressure (MAP) were not significantly different at rest and during exercise across groups (p<.05). There were no differences in SV, cardiac output (CO), and total vascular conductance (TVC) at rest across groups. Both SV and CO were significantly decreased in PG (p<.05), and TVC were significantly decreased in PG (p<.05). Both SV and TVC tended to be increased during exercise in OG and CG. There was no difference in norepinephrine (NE) concentration at rest across groups, while NE tended to be decreased in OG and CG. These results suggest that n-3 PUFAs supplement could improve SV and blood flow to active skeletal muscle. Thus, n-3 PUFAs supplement may prevent hypertension and enhance exercise performance in elderly women.
