Screening for Angiotensin 1-Converting Enzyme Inhibitory Activity of Ecklonia cava

Seven brown algal species (Ecklonia cava, Ishige okamurae, Sargassum fulvellum, Sargassum horneri, Sargassum coreanum, Sargassum thunbergii and Scytosiphon lomentaria) were hydrolyzed using five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase) and screened for angiotensin 1-converting enzyme (ACE) inhibitory activities. Most algal species examined showed good ACE inhibitory activities after the enzymatic hydrolysis. However, E. cava was the most potent ACE inhibitor of the seven species. Flavourzyme digest of E. cava exhibited an $IC_{50}$ of around $0.3\;{\mu}g/mL$ for ACE; captopril has an $IC_{50}$ of $\~0.05\;{\mu}g/mL$. The Flavourzyme digest was separated to three fractions by an ultrafiltration membrane (5, 10, 30 kDa MWCO) system according to the molecular weights. The active components were mainly concentrated in >30 kD fraction which are composed of the highest protein content $(27\%)$ and phenolic content (261 mg/100 mL) compared to the other two smaller molecular weight fractions. Therefore, the active compounds appear to be relatively high molecular weight complex molecules associated with protein (glycoprotein) and polyphenols. Therefore, E. cave is a potential source of antihypertensive compound.

Characterization of Algarose Product from Agar Using DMSO

Agar was extracted from Gelidium amansii, which was harvested at the shores of Jeju Island in South Korea. As a unique solvent, the ability of dimethyl sulfoxide (DMSO) was used to separate agarose from agar by removing agaropectine and quality of the resultant agarose was characterized for chromatography purposes. Agar sample was agitated by motor-driven stirrer with DMSO in a water bath (at 70°C for 2 h) and centrifuged (3,000 rpm for 20 min). Resultant upper agarose layer was gelled, washed, dried and milled. The quality of agarose was evaluated by the analysis of proximate chemical composition, sulfate content, gelling strength and DNA migration. In this study, the separated agarose showed low sulfate amount (0.28%) and showed high gel strength (1190 g · cm–2). The resolution power and the ligase activities gave clear picture about the suitability of the present agarose for practical purposes.
Antioxidant Activity of Sulfated Polysaccharides Isolated from Sargassum fulvellum

Sargassum fulvellum, a marine brown alga, is a popular low priced edible plant in Korean markets. The polysaccharide fraction of the alga was separated and investigated for its radical scavenging activities and the results compared with those of commercial fucoidans (Fucus vesiculosus and Undaria pinnatifida), BHA and ${\alpha}$-tocopherol. The polysaccharide fraction of S. fulvellum showed a promising DPPH radical scavenging activity than did other fucoidans. Moreover, the sample exhibited a dose-dependent activity on hydrogen peroxide scavenging activity in the V79-4 cell line. Interestingly, all the tested polysaccharide counterparts were more potent NO. scavengers than were the commercial antioxidants, BHA and ${\alpha}$-tocopherol. The sulfated polysaccharide of S. fulvellum had an approximate molecular weight of 529 kDa and mainly consisted of fucose and galactose, and minor amounts of mannose, rhamnose and xylose.
