      • A Feasibility Research on the Mechanism of Cold Chain Business in Deppon Logistics

        Cold chain distribution is always well-known as the “Mount Everest” in international logistics industry, while the rapid development in recent years of domestic meat, frozen food, cold drinks industry and dairy industry brought a leapfrog growth of cold chain logistics, which respectively stimulated the need for logistics outsourcing service from Third Party Logistics. Those companies demanding for exploiting cold chain business on one hand requires a complete set of refrigerated logistics chains, on the other hand also requires enterprises to establish a good and professional transport system with refrigerated operation equipment, which are precisely the weakness of manufacturing enterprises. Thus, outsourcing of cold chain logistics to a third party is the best choice for companies to focus on its core business. By analyzing the bulletins of International Cold Chain Logistics Summits, this paper shows the problems of cold chain industry in China and points out the potentials for its future development. It further analyzes the feasibility of launching cold chain business of Deppon Logistics. The purpose is to offer some suggestions to work out the barriers and set up a modern operation system under the big data age in Deppon.

        The Up-Regulation of miR-199b-5p in Erythroid Differentiation Is Associated with GATA-1 and NF-E2

        MicroRNAs (miRNAs) represent a class of small non-coding regulatory RNAs that play important roles in normal hematopoiesis, including erythropoiesis. Although studies have identified several miRNAs that regulate erythroid commitment and differentiation, we do not understand the mechanism by which the crucial erythroid transcription factors, GATA-1and NF-E2 directly regulate and control differentiation via miRNA pathways. In this study, we identified miR-199b-5p as a key regulator of human erythropoiesis, and its expression was up-regulated during the erythroid differentiation of K562 cells. Furthermore, the increase of miR-199b-5p in erythroid cells occurred in a GATA-1- and NF-E2-dependent manner during erythrocyte maturation. Both GATA-1 and NF-E2 bound upstream of the miR-199b gene locus and activated its transcription. Forced expression of miRNA-199b-5p in K562 cells affected erythroid cell proliferation and maturation. Moreover, we identified c-Kit as a direct target of miR-199b-5p in erythroid cells. Taken together, our results establish a functional link among the erythroid transcription factors GATA-1/NF-E2, miR-199b-5p and c-Kit, and provide new insights into the coupling of transcription and post-transcription regulation in erythroid differentiation.

        3D-quantitative structure–activity relationship and antiviral effects of curcumin derivatives as potent inhibitors of infl uenza H1N1 neuraminidase

        Curcumin derivatives have been shown to inhibitreplication of human infl uenza A viruses (IAVs). However, itis not clear whether curcumin and its derivatives can inhibitneuraminidase (NA) of influenza virus. In this study, ameaningful 3D quantitative structure–activity relationshipmodel (comparative molecular fi eld analysis R 2 = 0.997,q 2 = 0.527, s = 0.064, F = 282.663) was built to understandthe chemical–biological interactions between their activitiesand neuraminidase. Molecular docking was used to predictbinding models between curcumin derivatives and neuraminidase. Real-time polymerase chain reactions showed thatthe fi ve active curcumin derivatives might have direct eff ectson viral particle infectivity in H1N1-infected lung epithelial(MDCK) cells. Neuraminidase activation assay showed thatfi ve active curcumin derivatives decreased H1N1-inducedneuraminidase activation in MDCK cells. Indirect immunofluorescence assay indicated that two active curcuminderivatives (tetramethylcurcumin and curcumin) down-regulatedthe nucleoprotein expression. Curcumin inhibited IAVin vivo. The therapeutic mechanism of curcumin in the treatmentof infl uenza viral pneumonia is related to improvingthe immune function of infected mice and regulating secretionof tumor necrosis-α, interleukin-6, and interferon-γ. These results indicate that curcumin derivatives inhibit IAVby blocking neuraminidase in the cellular model and curcuminalso has anti-IAV activity in the animal model.

        Protective Effects of Polysaccharide Extracted from Portulacae oleracea L. on Colitis Induced by Dextran Sulfate Sodium

        Polysaccharide from Ma-chi-xian (Portulacae oleracea L., POLP) was prepared and the therapeutic effect on dextran sodium sulfate-induced colitis mice was investigated in this study. The results of clinical activity score and H&E staining confirmed the therapeutic effect of POLP. POLP could diminished the symptoms of colitis and improve colon histopathological structure of the colitis mice. The expression levels of four cytokines were determined. The concentrations of PGE2 and IL-6 were downregulated by POLP treatment. The COX-2 protein expression levels and the STAT3 phosphorylation levels were detected. The results showed that these two protein levels were all increased in colitis and decreased after POLP treatment, indicating that these two proteins were closely related with the protective effect of POLP. Because the synthesis of PGE2 is catalyzed by COX-2 and phosphorylation of STAT3 can induce the expression of COX-2, it was concluded that STAT3 was a key protein related to the POLP exerting its activity in colitis.

        External Counterpulsation Reduces Beat-to-Beat Blood Pressure Variability When Augmenting Blood Pressure and Cerebral Blood Flow in Ischemic Stroke

        Background and PurposezzExternal counterpulsation (ECP) is a noninvasive method used to enhance cerebral perfusion by elevating the blood pressure in ischemic stroke. However, the response of the beat-to-beat blood pressure variability (BPV) in ischemic stroke patients during ECP remains unknown. MethodszzWe enrolled recent ischemic stroke patients and healthy controls. Changes in the blood fow velocities in bilateral middle cerebral arteries and the continuous beat-to-beat blood pressure before, during, and afer ECP were monitored. Power spectral analysis revealed that the BPV included oscillations at very low frequency (VLF; <0.04 Hz), low frequency (LF; 0.04– 0.15 Hz), and high frequency (HF; 0.15–0.40 Hz), and the total power spectral density (TP; <0.40 Hz) and LF/HF ratio were calculated. ResultszzWe found that ECP signifcantly increased the systolic and diastolic blood pressures in both stroke patients and controls. ECP decreased markedly the systolic and diastolic BPVs at VLF and LF and the TP, and the diastolic BPV at HF when compared with baseline. Te decreases in diastolic and systolic BPV reached 37.56% and 23.20%, respectively, at VLF, 21.15% and 12.19% at LF, 8.76% and 16.59% at HF, and 31.92% and 23.62% for the total TP in stroke patients, which did not difer from those in healthy controls. Te change in fow velocity on the contralateral side was positively correlated with the total TP systolic BPV change induced by ECP (r=0.312, p=0.035). ConclusionszzECP reduces the beat-to-beat BPV when increasing the blood pressure and cerebral blood fow velocity in ischemic stroke patients. ECP might be able to improve the clinical outcome by decreasing the beat-to-beat BPV in stroke patients, and this should be explored further in future studies.

        An Energy Harvesting Aware Routing Algorithm for Hierarchical Clustering Wireless Sensor Networks

        Recently, energy harvesting technology has been integrated into wireless sensor networks to ameliorate the nodes` energy limitation problem. In theory, the wireless sensor node equipped with an energy harvesting module can work permanently until hardware failures happen. However, due to the change of power supply, the traditional hierarchical network routing protocol can not be effectively adopted in energy harvesting wireless sensor networks. In this paper, we improve the Low-Energy Adaptive Clustering Hierarchy (LEACH) protocol to make it suitable for the energy harvesting wireless sensor networks. Specifically, the cluster heads are selected according to the estimation of nodes` harvested energy and consumed energy. Preference is given to the nodes with high harvested energy while taking the energy consumption rate into account. The utilization of harvested energy is mathematically formulated as a max-min optimization problem which maximizes the minimum energy conservation of each node. We have proved that maximizing the minimum energy conservation is an NP-hard problem theoretically. Thus, a polynomial time algorithm has been proposed to derive the near-optimal performance. Extensive simulation results show that our proposed routing scheme outperforms previous works in terms of energy conservation and balanced distribution.

        Characterization of a Strain of Malva Vein Clearing Virus in Alcea rosea via Deep Sequencing

        Malva vein clearing virus (MVCV) is a member of the Potyvirus species, and has a negative impact on the aesthetic development of Alcea rosea. It was first reported in Germany in 1957, but its complete genome sequence data are still scarce. In the present work, A. rosea leaves with vein-clearing and mosaic symptoms were sampled and analyzed with small RNA deep sequencing. By denovo assembly the raw sequences of virus-derived small interfering RNAs (vsiRs) and whole genome amplification of malva vein cleaning virus SX strain (MVCVSX) by specific primers targeting identified contig gaps, the full-length genome sequences (9,645 nucleotides) of MVCV-SX were characterized, constituting of an open reading frame that is long enough to encode 3,096 amino acids. Phylogenetic analysis showed that MVCVSX was clustered with euphorbia ringspot virus and yam mosaic virus. Further analyses of the vsiR profiles revealed that the most abundant MVCV-vsiRs were between 21 and 22 nucleotides in length and a strong bias was found for “A” and “U” at the 5′-terminal residue. The results of polarity assessment indicated that the amount of sense strand was almost equal to that of the antisense strand in MVCV-vsiRs, and the main hot-spot region in MVCV-SX genome was found at cylindrical inclusion. In conclusion, our findings could provide new insights into the RNA silencing-mediated host defence mechanism in A. rosea infected with MVCV-SX, and offer a basis for the prevention and treatment of this virus disease.

        A Combination of Baicalin and Berberine Hydrochloride Ameliorates Dextran Sulfate Sodium-Induced Colitis by Modulating Colon Gut Microbiota

        Baicalin and berberine hydrochloride are the main chemical compositions of Scutellariae Radix and Coptidis Rhizoma, respectively. S. Radix and C. Rhizoma are two traditional Chinese herbs that are commonly used together in compounded formulations to treat colitis. Therefore, the combination of Baicalin and berberine hydrochloride (BBH) to treat colitis was studied. The results of pharmacological evaluations demonstrated the excellent protective effects of BBH on colitis induced by dextran sulfate sodium (DSS). BBH could improve the morphological condition of colitis in mice and maintain the balance of proinflammation cytokines (IL-6, IL-8, IL-1β, and TNF-α) and anti-inflammation cytokines (IL-4 and IL-10). The 16s rDNA sequencing revealed that BBH was able to modulate the composition of intestinal microflora, especially the abundances of Eubacterium_brachy_group, Holdemania, Erysipelotrichaceae_UCG_003, Christensenellaceae_R-7_group, and Sellimonas. The results of PICRUSt indicated that the therapeutic effects of BBH were tightly connected with DNA synthesis, replication and repair of gut microbiota. In summary, it was concluded that BBH could protect mice against DSS-induced colitis, and the protective effects were tightly correlated with gut microbiota.
