An improved method for the preparation of monomethine cyanine dyes with quinoline nucleus by one-pot three-component using 1-methyl-2-quinolinethione, quaternized 2- or 4-methylheterocyclic compounds and methyl p-toluenesulfonate as starting materials was described. Compared with the traditional methods, the new synthetic method reduced the reaction steps, shortened the reaction time, avoided the separation and purification of the intermediate and reduced cost. The dyes absorbed in the region 478.0-563.0 nm and had molar extinction coefficients of $1.3{\times}10^4-9.4 {\times}10^4L\;mol^{-1}\;cm^{-1}$. Their fluorescence maxima and Stokes shifts were in the range of 525.2-594.4 nm and 16.2-80.6 nm in different solvents, respectively. From the spectral properties of the dyes in different solvents, it could be found that the ${\lambda}_{max}$ of the dyes were shorter in protonic solvents, and showed hypsochromic shifts with the increase of polarity of the solvents.

Production of recombinant proteins by excretory expression has many advantages over intracellular expression in Escherichia coli. Hyperexpression of a secretory exoglucanase, Exg, of Cellulomonas fimi was previously shown to saturate the SecYEG pathway and result in dramatic cell death of E. coli. In this study, we demonstrated that overexpression of the PspA in the JM101(pM1VegGcexL-pspA) strain enhanced excretion of Exg to 1.65 U/ml using shake-flask cultivation, which was 80% higher than the highest yield previously obtained from the optimized JM101(pM1VegGcexL) strain. A much higher excreted Exg activity of 4.5 U/ml was further achieved with high cell density cultivation using rich media. Furthermore, we showed that the PspA overexpression strain enjoyed an elevated critical value (CV), which was defined as the largest quotient between the intracellular unprocessed precursor and its secreted mature counterpart that was still tolerable by the host cells prior to the onset of cell death, improving from the previously determined CV of 20/80 to the currently achieved CV of 45/55 for Exg. The results suggested that the PspA overexpression strain might tolerate a higher level of precursor Exg making use of the SecYEG pathway for secretion. The reduced lethal effect might be attributable to the overexpressed PspA, which was postulated to be able to reduce membrane depolarization and damage. Our findings introduce a novel strategy of the combined application of metabolic engineering and construct optimization to the attainment of the best possible E. coli producers for secretory/excretory production of recombinant proteins, using Exg as the model protein.
The Synthesis and Spectral Properties of Some New Monomethine Cyanine Dyes for Live Cell Imaging

Four monomethine cyanine dyes were synthesized and their structures were confirmed by 1H NMR, IR, UV–Vis, and HRMS. Their spectral properties were studied. It could be found that the λmax of the dyes was shorter in the protonic solvents compared with that in the non-protonic solvents. Moreover, the λmax shows a hypsochromic shift with the increasing polarity of the solvents. The interactions of four dyes with DNA were also studied. The results showed that the fluorescence intensities of the dyes 3a and 3b took place a qualitative change in the presence of DNA compared with the free dyes. The dyes used as cellular imaging agents were researched. The results indicated that the dye 3b could penetrate the live cell membrane.

We have performed a comparative investigation of the series compounds (InSb)nCum to assess the roles of Cu addition on the thermoelectric properties and nanostructuring in bulk InSb. Detailed temperature dependent transport properties including electrical conductivity, the Seebeck coefficient, and thermal conductivity are presented. The Seebeck coefficients of In20Sb20Cu (m:n ¼ 1:20) are increased by 13percent in magnitude if compared to those of InSb, which is responsible for the 22 percent enhancement in the highest ZT value at 687 K. Although the magnitudes of kL are larger than those of InSb over the entire temperature range, a remarkable reduction in lattice thermal conductivities (kL) was observed with measuring temperature elevation. Such changes are mainly due to the precipitation of a large number of Cu9In4 nanoparticles with the size of smaller than 5 nm, dispersed in the matrix observed using high resolution transmission electron microscopy (HRTEM) images.

High-energy synchrotron X-ray diffraction technique was used to in-situ characterize microstructure, lattice strain, and phase transition behavior of a Ni<SUB>47</SUB>Ti<SUB>44</SUB>Nb<SUB>9</SUB> shape memory alloy. Phase transformation kinetics and deformation mechanisms were studied under a uniaxial tension at three testing temperatures, i.e., -70<SUP>o</SUP>C, 25<SUP>o</SUP>C, and 150<SUP>o</SUP>C. At a testing temperature of -70<SUP>o</SUP>C, a complicated phase transformation with four stages of micromechanical deformation was identified which is associated with changes of martensite substructures. At room temperature of 25<SUP>o</SUP>C, there was no stress-induced selection process of variants of B19' phases observed. Whereas at a testing temperature of 150<SUP>o</SUP>C, there was no any phase transformation observed. It is verified that β-Nb phase, an effective stabilizer for the austenite, delays the process of martensitic transformation and relaxes the strain energy without strengthening the matrix. This new finding is important to understand the relationship between the micromechanical deformation behavior and phase transformations in the Ni<SUB>47</SUB>Ti<SUB>44</SUB>Nb<SUB>9</SUB> SMA.
100KW DC Arc Plasma of CVD System for Low Cost Large Area Diamond Film Deposition

In the present paper, a new type of DC arc plasma torch is disclosed. The principles of the new magnetic and fluid dynamic controlled large orifice long discharge tunnel plasma torch is discussed. Two series of DC Plasma Jet diamond film deposition equipment have been developed. The 20kW Jet equipped with a $\Phi$70 mm orifice torch is capable of deposition diamond films at a growth rate as high as 40$\mu\textrm{m}$/h over a substrate area of $\Phi$65 mm. The 100kW high power Jet which is newly developed based on the experience of the low power model is equipped with a $\Phi$120 mm orifice torch, and is capable of depositing diamond films over a substrate area of $\Phi$110 mm at growth rate as high as 40 $\mu\textrm{m}$/h, and can be operated at gas recycling mode, which allows 95% of the gases be recycled. It is demonstrated that the new type DC plasma torch can be easily scaled up to even higher power Jet. It is estimated that even by the 100kW Jet, the cost for tool grade diamond films can be as low as less than $4/carat.

Located at the important tumor suppressor locus, 3p22, PLCD1 encodes an enzyme that mediates regulatory signaling of energy metabolism, calcium homeostasis and intracellular movements. We identified PLCD1 as a downregulated gene in aerodigestive carcinomas through expression profiling and epigenetic characterization. We found that PLCD1 was expressed in all normal adult tissues but low or silenced in 84% (16/19) gastric cancer cell lines, well correlated with its CpG island (CGI) methylation status. Methylation was further detected in 62% (61/98) gastric primary tumors, but none of normal gastric mucosa tissues. PLCD1 methylation was significantly correlated with tumor high stage. Detailed methylation analysis of 37 CpG sites at the PLCD1 CGI by bisulfite genomic sequencing confirmed its methylation. PLCD1 silencing could be reversed by pharmacological demethylation with 5-aza-2′-deoxycytidine, indicating a direct epigenetic silencing. Ectopic expression of PLCD1 in silenced gastric tumor cells dramatically inhibited their clonogenicity and migration, possibly through downregulating MMP7 expression and hampering the reorganization of cytoskeleton through cofilin inactivation by phosphorylation. Thus, epigenetic inactivation of PLCD1 is common and tumor-specific in gastric cancer, and PLCD1 acts as a functional tumor suppressor involved in gastric carcinogenesis.Oncogene (2009) 28, 2466–2475; doi:10.1038/onc.2009.92; published online 18 May 2009
