Cloning and characterization of the cardiac-specific Lrrc10 promoter

Leucine-rich repeat containing protein 10 (LRRC10) is characterized as a cardiac-specific gene, suggesting a role in heart development and disease. A severe cardiac morphogenic defect in zebrafish morphants was recently reported but a contradictory result was found in mice, suggesting a more complicated molecular mechanism exists during mouse embryonic development. To elucidate how LRRC10 is regulated, we analyzed the 5`enhancer region approximately 3 kilo bases (kb) upstream of the Lrrc10 start site using luciferase reporter gene assays. Our characterization of the Lrrc10 promoter indicates it possesses complicated cis-and trans-acting elements. We show that GATA4 and MEF2C could both increase transcriptional activity of Lrrc10 promoter individually but that they do not act synergistically, suggesting that there exists a more complex regulation pattern. Surprisingly, knockout of Gata4 and Mef2c binding sites in the 5`enhancer region (-2,894/-2,889) didn`t change the transcriptional activity of the Lrrc10 promoter and the likely GATA4 binding site identified was located in a region only 100 base pair (bp) upstream of the promoter. Our data provides insight into the molecular regulation of Lrrc10 expression, which probably also contributes to its tissue-specific expression. [BMB reports 2011; 44(2): 123-128]
Geft is dispensable for the development of the second heart field

Geft is a guanine nucleotide exchange factor, which can specifically activate Rho family of small GTPase by catalyzing the exchange of bound GDP for GTP. Geft is highly expressed in the excitable tissue as heart and skeletal muscle and plays important roles in many cellular processes, such as cell proliferation, migration, and cell fate decision. However, the in vivo role of Geft remains unknown. Here, we generated a Geft conditional knockout mouse by flanking exons 5-17 of Geft with loxP sites. Cre-mediated deletion of the Geft gene in heart using Mef2c-Cre transgenic mice resulted in a dramatic decrease of Geft expression. Geft knockout mice develop normally and exhibit no discernable phenotype, suggesting Geft is dispensable for the development of the second heart field in mouse. The Geft conditional knockout mouse will be a valuable genetic tool for uncovering the in vivo roles of Geft during development and in adult homeostasis. (BMB reports 2012; 45(3): 153-158)
DP-LinkNet: A convolutional network for historical document image binarization

Document image binarization is an important pre-processing step in document analysis and archiving. The state-of-the-art models for document image binarization are variants of encoder-decoder architectures, such as FCN (fully convolutional network) and U-Net. Despite their success, they still suffer from three limitations: (1) reduced feature map resolution due to consecutive strided pooling or convolutions, (2) multiple scales of target objects, and (3) reduced localization accuracy due to the built-in invariance of deep convolutional neural networks (DCNNs). To overcome these three challenges, we propose an improved semantic segmentation model, referred to as DP-LinkNet, which adopts the D-LinkNet architecture as its backbone, with the proposed hybrid dilated convolution (HDC) and spatial pyramid pooling (SPP) modules between the encoder and the decoder. Extensive experiments are conducted on recent document image binarization competition (DIBCO) and handwritten document image binarization competition (H-DIBCO) benchmark datasets. Results show that our proposed DP-LinkNet outperforms other state-of-the-art techniques by a large margin. Design of Switchable and Reconfigurable Semi-lumped Wideband Bandpass Filter

A switchable single-wideband (SWB)-to-dual-wideband (DWB) bandpass filter (BPF), which is realized by using lumped switches, is presented in this paper. By alternating the operation modes—ON and OFF—in which the ON mode is achieved by placing the capacitors at the switching spots and the OFF mode is achieved by replacing the capacitors with inductors, DWB-to-SWB BPF can be achieved on the same device. In addition, by changing the capacitor values, the center frequency (CF) of the lower passband of DWB BPF can be easily tuned from 1.69 GHz to 2.22 GHz, while the higher passband stays almost unchanged. As an example, an SWB-to-DWB BPF is designed, fabricated, and measured. This BPF exhibits good performance including wideband, high isolation, compact size, and ability to switch.

Hierarchical carbon foams (HCFs) with micro-, meso-, and macropores were successfully synthesized via a two-step process: (1) polymerization in oil-in-water (O/W) emulsions without any hard templates and (2) carbonization at 850°C. With the aim of both enhancing the stability of the emulsion and forming a micro- and mesoporous structure during the carbonization process, potassium citrate was introduced in an aqueous solution of resorcinol and formaldehyde. A series of HCFs were fabricated by changing the mass ratio of potassium citrate to total carbon sources from 0.25 to 1.5. The effect of potassium citrate on the porous structure of HCFs was investigated through nitrogen sorption tests. The prepared HCFs exhibited well-developed porous structures of micro-, meso- and macropores and high surface areas. The structural characteristics of the HCFs, including pore size distribution, surface area, and porosity, were significantly dependent on the amount of potassium citrate. It was concluded that potassium citrate greatly contributed to the formation of carbon foams with nano-sized pore structures and high porosity. Interestingly, it was found that when the mass ratio of potassium citrate to total carbon sources was 0.5, the HCFs showed the highest specific surface area (~1360 m2/g). Furthermore, the capacitive performances of the HCFs were evaluated in a 6.0 M KOH aqueous solution using typical electrochemical methods such as cyclic voltammetry and galvanostatic charge/discharge tests. The capacitance of the HCFs tended to increase with the increase in surface area. In particular, the HCFs with the highest surface area also exhibited excellent electrochemical properties (high capacitance of 224 F/g at 1.0 A/g, high rate capability of 191 F/g at 10.0 A/g). These features may be attributed to both the resulting interconnected pore structure that is easily accessible to ions and the high surface area. We believe that this synthesis strategy can be easily extended to the preparation of hierarchical porous carbon materials with a desirable pore structure by tuning the organic monomer.
Inhibition of cancer cell growth and migration by dihydroxynaphthyl aryl ketones

Dihydroxynaphthyl aryl ketones 1-5 exhibit activity as tubulin polymerization inhibitors by targeting the colchicine binding site of microtubules making them potential anticancer drugs. Therefore, analogues 1-5 have been evaluated for their cytotoxic activity against the cancer cell lines DU-145 (prostate), T24 (bladder) and MCF-7 (breast). notable differences in biological activity were observed for compounds 1-5, most likely related to the nature of the aryl substituent bonded to the carbonyl group. among the tested compounds, only compound 5 showed selectivity for cancer cells over healthy, non-transformed cells. T24 cancer cells treated with compound 5 presented a concentration-dependent decrease in cell proliferation and a loss of migration ability. The cytotoxicity of compounds 1-5 on the selected cell-based assays is discussed in terms of it lipophilicity and polarizability parameters.
Study on Dynamic Guidance Obstacle Avoidance for UAV

With the flight environment becoming more and more complex, how to avoid the moving threat for the UAV under the dynamic environment and complete the mission become a key problem. With the idea of guidance, using the method of dimension reduction, the dynamic guidance obstacle avoidance method in three dimensional spaces is given. Considering the collision avoidance completion time and the UAV’s maneuverability, the parameters range of obstacle avoidance navigation law are deduced. Simulation results show the validity of the algorithm.

With the extensive use of UAV, how to make the UAV avoid the obstacle and arrive at the destination has become a hot issue. To make the angle shift minimum, a method on dynamic guidance obstacle avoidance of UAV based on the minimum angle shift is proposed, considering the constraints such as the completion time of obstacle, the motor performance, the direction of velocity after obstacle avoidance and so on, determining the range of guidance parameters. In the meanwhile, the obstacle method is extended to 3D-space based on 2D. Moreover, numerical simulations are carried out to verify the effectiveness of the method.

An LC-MS/MS method was established for simultaneous identification of levels of bilpaterol, cimbuterol, clenproperol, and bambuterol in goat and beef muscle. Meat samples were subjected to extraction using an aqueous acidic solution and cleansed using MCX solid phase extraction. β2-Agonists residues were extracted using an aqueous acidic solution. Proteins in extraction liquids was removed based on adjusting pH using perchloric acid and a sodium hydroxide solution, followed by SPE for clean-up. Linearity values of the method based on R2 values were 0.9976-0.9997 with 70.1-108.8% recovery and relative standard deviation values of 3.5-13.3%. Low limits of detection (LOD) and quantitation (LOQ) of 0.01-0.02 and 0.02-0.08 μg/kg, respectively, were achieved for spiked goat and spiked beef muscle. The method was sensitive and specific and is an improvement over other currently available technologies.
