        C-Reactive Protein Gene Variants in Depressive Symptoms & Antidepressants Efficacy

        Objective Although the pathogenesis of depression remains unclear, C-reactive protein (CRP) levels are commonly elevated in depressed patients. Thus, CRP single-nucleotide polymorphisms (SNPs) that influence CRP levels may be associated with depression. In the present study, we explored whether CRP SNPs are related to depressive symptoms and antidepressants efficacy in Han Chinese patients. Methods We analyzed data from 440 patients with first-episode depression. We obtained genome CRP SNPs, scores of the 17-item Hamilton Rating Scale for Depression 17 (HAMD17) and its four-factor at baseline and after 6 weeks. Quantitative trait analysis was performed using UNPHASED software and curative effects were analyzed using SPSS software. Results Male patients with SNP rs1800947G exhibited lower insomnia scores and rs2794521CC exhibited lower scores of anxiety/ physical symptoms, total HAMD17 score. Female patients with rs2794521TT exhibited higher scores of insomnia and lower antidepressants efficacy. Conclusion CRP SNPs rs1800947 and rs2794521 may be associated with depressive symptoms in patients with depression in a sex-specific fashion. Furthermore, rs2794521 may be a predictor of the efficacy of antidepressants in female patients.

        Complex Current Controller Design Based on Multiple Equations Construction of Three-Phase Inverter

        This paper proposes a complex PI current controller design method of three-phase inverter based on multiple equations construction. The mathematical model of three-phase inverter system plays an important role in the controller design process. In the synchronous reference frame, the inverter model introduces coupling elements, which is diff erent from the model in the stationary reference frame and brings diffi culty for current controller design. Complex vector model (CVM) is introduced in the current loop, which is established in the synchronous reference frame and takes part in the current controller design. Multiple equations construction is established to solve the parameters of the complex PI current controller. Because the controller satisfi es the restrictions of the multiple equations, it can realize good dynamic performance and the coupling characteristic of the three-phase system. In this paper, the CVM is established. The process of construct and solve the multiple equations is analyzed in detail. The accuracy of the inverter system is validated. Finally the simulation and experimental results are given to verify the theoretical analysis.

        Crosstalk Between Apoptosis and Autophagy Is Regulated by the Arginylated BiP/Beclin-1/p62 Complex

        <P>Emerging evidence demonstrates that autophagy and apoptosis are interconnected and their interplay greatly affects cell death. However, the key regulators in this crosstalk remain elusive. Therefore, the role of N-terminal arginylated BiP (R-BiP)/Beclin-1/p62 complex was examined in the crosstalk between apoptosis and autophagy during combination chemotherapy with mitomycin C and bortezomib using immunoblot, immunoprecipitation, and cellular imaging assays in wild-type (WT) and genetically engineered colorectal cancer cells. In addition, the tumoricidal efficacy of the combinatorial treatment in a nude mouse tumor xenograft model of colorectal cancer was assessed. Bortezomib combined with mitomycin C synergistically induced cytotoxicity and apoptosis rather than autophagy. Mechanistically, this combination inactivated Akt and subsequently induced Beclin-1 (<I>BECN1</I>) dephosphorylation at Ser 234/295. Dephosphorylation of Beclin-1 resulted in increased cleavage of Beclin-1 and disruption of the R-BiP/Beclin-1/p62 complex, which led to switching autophagy to the synergistic induction of apoptosis. Importantly, the combination significantly suppressed LS174T intraperitoneal xenograft tumor growth, induced Akt inactivation and Beclin-1 cleavage, and decreased autophagy <I>in vivo</I>. Moreover, the tumoricidal efficacy of the combinatorial treatment was less effective, <I>in vitro</I> and <I>in vivo</I>, in HCT116 tumors harboring a Beclin-1 caspase 8 cleavage site mutant knock-in.</P><P><B>Implications:</B> This study uncovers that the R-BiP/Beclin-1/p62 complex has an important role in the crosstalk between apoptosis and autophagy.         Epidemiological Characteristics of Norovirus Outbreaks in Shenyang from 2017 to 2021

        Norovirus is one of the leading causes of acute gastroenteritis outbreaks worldwide. This study aimed to identify the epidemiological characteristics of norovirus outbreaks and to provide evidence for public health entities. Specimens and epidemiological survey data were collected to determine if there were differences in the attack rate of norovirus in terms of the year, season, transmission route, exposure setting, and region and to determine whether there were relationships between the reporting interval, the number of illnesses in a single outbreak and the duration of the outbreak. Norovirus outbreaks were reported throughout the year, with seasonal characteristics (i.e., high rates in spring and winter). Among all regions in Shenyang with the exception of Huanggu and Liaozhong, norovirus outbreaks had been reported, and the primary genotype was GII.2[P16]. Vomiting was the most common symptom. The main places of occurrence were childcare institutions and schools. The person-to-person route was the main transmission route. The median duration of norovirus was 3 days (IQR [interquartile range]: 2–6 days), the median reporting interval was 2 days (IQR: 1–4 days), the median number of illnesses in a single outbreak was 16 (IQR: 10–25); there was a positive correlation between these parameters. Norovirus surveillance and genotyping studies still need to be further strengthened to increase knowledge regarding the pathogens and their variant characteristics, to better characterize the patterns of norovirus outbreaks and to provide information for outbreak prevention. Norovirus outbreaks should be detected, reported and handled early. Public health entities and the government should develop corresponding measures for different seasons, transmission routes, exposure settings, and regions.

      • A Big-Data-Based Urban Flood Defense Decision Support System

        As cities in developing countries are expanding rapidly in recent years, flood has an increasing impact on urban management. In this paper, we present the design and implementation of an urban flood defense decision support system based on big data. The system connects real-time sensor to collect streaming data, and uses a data-driven method that considers temporal and spatial factors to forecast water level in the next 6 hours. Thus, it can provide enough time for the authorities to take pertinent flood protection measures such as evacuation. Our predictive model is a hybrid of linear regression and artificial neural network, and can give early warning of potential flood using the forecast results. The system is implemented on Java EE platform, and integrated with Baidu Maps API to provide a user-friendly interface.

      • An Intelligent Analysis and Mining System for Urban Lighting Information

        Urban lighting information plays an important role in digital urban management. In this paper, an IAMSULI (Intelligent Analysis and Mining System for Urban Lighting Information) is proposed. Using data analysis technology, case-based reasoning technology and data mining technology comprehensively, the system aims at providing better decision support for lighting management. IAMSULI is composed of system management, geographical information management, lighting information management, lighting data analysis, comprehensive classification statistics and event management. The system can help us quickly find lighting problems, provide early warning alarm of the problems, and also supply constructive information for the urban development.

        Insertion Mutation in HMG-CoA Lyase Increases the Production Yield of MPA through Agrobacterium tumefaciens-Mediated Transformation

        Mycophenolic acid (MPA) is an antibiotic produced by Penicillium brevicompactum. MPA has antifungal, antineoplastic, and immunosuppressive functions, among others. β-Hydroxy-β-methylglutaryl-CoA (HMG-CoA) lyase is a key enzyme in the bypass metabolic pathway. The inhibitory activity of HMG-CoA lyase increases the MPA biosynthetic flux by reducing the generation of by-products. In this study, we cloned the P. brevicompactum HMG-CoA lyase gene using the thermal asymmetric interlaced polymerase chain reaction and gene walking technology. Agrobacterium tumefaciens-mediated transformation (ATMT) was used to insert a mutated HMG-CoA lyase gene into P. brevicompactum. Successful insertion of the HMG-CoA lyase gene was confirmed by hygromycin screening, PCR, Southern blot analysis, and enzyme content assay. The maximum MPA production by transformants was 2.94 g/l. This was 71% higher than wild-type ATCC 16024. Our results demonstrate that ATMT may be an alternative practical genetic tool for directional transformation of P. brevicompactum.

        Characterization of high-alumina coal fly ash based silicate material and its adsorption performance on volatile organic compound elimination

        A highly stable silicate material from high-alumina coal fly ash was prepared and characterized using X-raydiffraction, scanning electron microscopy (SEM), Fourier transform infrared (FTIR) spectroscopy, thermal analysis, Xrayphotoelectron spectroscopy, and elemental analysis. The spectral results show that the silicate material was mainlycomposed of six elements, C, Ca, O, Si, Mg, and Al, in the form of Ca2+, Mg2+, Al3+, SiO32−, and CO32− ions. Some adsorbedwater and/or water of crystallization was also observed. The silicate material showed exceptionally high capabilityto adsorb volatile organic compounds (VOCs). The results of dynamic adsorption behavior show that the silicatematerial presents similar properties with commercial activated carbon and stronger absorption properties than commercialdiatomite for the adsorption of VOCs. The FTIR spectral results show weak hydrogen bonding interactions ofthe silicate material with three VOCs.
