        Microvascular Myocardial Ischemia in Patients With Diabetes Without Obstructive Coronary Stenosis and Its Association With Angina

        Objective: To investigate the incidence of microvascular myocardial ischemia in diabetic patients without obstructive coronary artery disease (CAD) and its relationship with angina. Materials and Methods: Diabetic patients and an intermediate-to-high pretest probability of CAD were prospectively enrolled. Non-diabetic patients but with an intermediate-to-high pretest probability of CAD were retrospectively included as controls. The patients underwent dynamic computed tomography-myocardial perfusion imaging (CT-MPI) and coronary computed tomography angiography (CCTA) to quantify coronary stenosis, myocardial blood flow (MBF), and extracellular volume (ECV). The proportion of patients with microvascular myocardial ischemia, defined as any myocardial segment with a mean MBF ≤ of 100 mL/min/100 mL, in patients without obstructive CAD (Coronary Artery Disease–Reporting and Data System [CAD-RADS] grade 0–2 on CCTA) was determined. Various quantitative parameters of the patients with and without diabetes without obstructive CAD were compared. Multivariable analysis was used to determine the association between microvascular myocardial ischemia and angina symptoms in diabetic patients without obstructive CAD. Results: One hundred and fifty-two diabetic patients (mean age: 59.7 ± 10.7; 77 males) and 266 non-diabetic patients (62.0 ± 12.3; 167 males) were enrolled; CCTA revealed 113 and 155 patients without obstructive CAD, respectively. For patients without obstructive CAD, the mean global MBF was significantly lower for those with diabetes than for those without (152.8 mL/min/100 mL vs. 170.4 mL/min/100 mL, P < 0.001). The mean ECV was significantly higher for diabetic patients (27.2% vs. 25.8%, P = 0.009). Among the patients without obstructive CAD, the incidence of microvascular myocardial ischemia (36.3% [41/113] vs. 10.3% [16/155], P < 0.001) and interstitial fibrosis (69.9% [79/113] vs. 33.3% [8/24], P = 0.001) were significantly higher in diabetic patients than in the controls. The presence of microvascular myocardial ischemia was independently associated with angina symptoms (adjusted odds ratio = 3.439, P = 0.037) in diabetic patients but without obstructive CAD. Conclusion: Dynamic CT-MPI + CCTA revealed a high incidence of microvascular myocardial ischemia in diabetic patients without obstructive CAD. Microvascular myocardial ischemia is strongly associated with angina.

        Compression Property of Curved Three-dimensional Flat-knitted Spacer Fabric Composites under Quasi-static Loading

        In order to study the compression property of curved three-dimensional(3D) flat-knitted spacer fabric compositesand to provide experimental data for the study of subsequent low speed impact and residual strength, 3D flat-knitted spacerfabrics were knitted into flat and curvilinear shapes, then 3D flat-knitted spacer fabric composites curved in different angleswere developed. Compression tests on various curvilinear composites were carried out using the universal material testingmachine after which stress-strain curves are compared. The results revealed that when other experimental conditions are thesame, the coursewise compression peak was significantly higher than waleswise, and compression peak also graduallyincreased with the increase of curvature, no matter the surface of 3D spacer fabric reinforced composites is flat or curvilinear. This research will lay the foundation for the further mechanical property study of curved 3D spacer fabric composites.

        Health oil preparation from gardenia seeds by aqueous enzymatic extraction combined with puffing pre-treatment and its properties analysis

        Gardenia jasminoides Ellis, a representative for “homology of medicine and food”, can be used to produce pigment and edible oil. Here, aqueous enzymatic extraction (AEE) combined with puffing pre-treatment was explored to prepare oil from gardenia seeds. Both wet-heating puffing (WP) at 90 °C and dry-heating puffing (DP) at 1.0 MPa facilitated the release of free oil by AEE, resulting in the highest free oil yields (FOY) of 21.8% and 23.2% within 3 h, much higher than that of un-puffed group. Additionally, active crocin and geniposide were also completely released. The FOY obtained was much higher than mechanical pressing method (10.44%), and close to solvent extraction (25.45%). Microstructure analysis indicated that gardenia seeds expanded by dry-heating puffing (1.0 MPa) had a larger, rougher surface and porous structure than other groups. Overall, AEE coupled with puffing pre-treatment developed is an eco-friendly extraction technology with high efficiency that can be employed to oil preparation.

        Characteristics analysis of rotor-rolling bearing coupled system with fit looseness fault and its verification

        Outer ring of bearing fit looseness fault is a common fault. Scratch often appears in the inner surface of pedestal. The fit looseness fault mechanism is not clear. For rotor-rolling bearing system with fit looseness fault between rotor-bearing outer ring and pedestal, a rotor coupling dynamic model that the interaction of bearing outer ring and pedestal are considered. This model is different from the universal rubbing model, where the directions of relative motion between rotor and stator are not considered. Numerical integration method is used to obtain the response of the system where the rotor is established by FEM and the bearing outer ring and pedestal are established by lumped mass model. Firstly, modal test results and simulation results were used to verify the correctness of this model. Secondly, the role of tightening torque between bearing outer ring and pedestal is considered, and the response characteristics of bearing and rotor are analyzed when fit looseness fault is considered. Finally, comparing the simulation results with test results, the waveform and spectrum are similar, which verifies the correctness of the fit looseness model. The fit looseness fault characteristics are that the acceleration after noise reduction shows periodic impact, up and down asymmetry, multiple frequencies appear. A method by increasing tightening torque is put forward to control the vibration caused by fit looseness fault.

        Ginsenosides Rc, as a novel SIRT6 activator, protects mice against high fat diet induced NAFLD

        Background: Hepatic lipid disorder impaired mitochondrial homeostasis and intracellular redox balance, triggering development of non-alcohol fatty liver disease (NAFLD), while effective therapeutic approach remains inadequate. Ginsenosides Rc has been reported to maintain glucose balance in adipose tissue, while its role in regulating lipid metabolism remain vacant. Thus, we investigated the function and mechanism of ginsenosides Rc in defending high fat diet (HFD)-induced NAFLD. Methods: Mice primary hepatocytes (MPHs) challenged with oleic acid & palmitic acid were used to test the effects of ginsenosides Rc on intracellular lipid metabolism. RNAseq and molecular docking study were performed to explore potential targets of ginsenosides Rc in defending lipid deposition. Wild type and liver specific sirtuin 6 (SIRT6, 50721) deficient mice on HFD for 12 weeks were subjected to different dose of ginsenosides Rc to determine the function and detailed mechanism in vivo. Results: We identified ginsenosides Rc as a novel SIRT6 activator via increasing its expression and deacetylase activity. Ginsenosides Rc defends OA&PA-induced lipid deposition in MPHs and protects mice against HFD-induced metabolic disorder in dosage dependent manner. Ginsenosides Rc (20mg/kg) injection improved glucose intolerance, insulin resistance, oxidative stress and inflammation response in HFD mice. Ginsenosides Rc treatment accelerates peroxisome proliferator activated receptor alpha (PPAR-α, 19013)-mediated fatty acid oxidation in vivo and in vitro. Hepatic specific SIRT6 deletion abolished ginsenoside Rc-derived protective effects against HFD-induced NAFLD. Conclusion: Ginsenosides Rc protects mice against HFD-induced hepatosteatosis by improving PPAR-α-mediated fatty acid oxidation and antioxidant capacity in a SIRT6 dependent manner, and providing a promising strategy for NAFLD.

        Probabilistic Load Flow Analysis Based on Sparse Polynomial Chaotic Expansion

        To aim at the defciencies of the traditional second-order polynomial chaotic expansion with more input variables and larger calculation quantities applied in power systems probabilistic load fow (PLF) calculation, an improved PLF method is proposed based on sparse polynomial chaotic expansion theory in this paper. In this method, to obtain the sparse expression of the polynomial chaotic expansion (PCE) and reduce calculation quantity, the quadratic cross terms in the second-order PCE are eliminated by Sobol sensitivity analysis. And then, Nataf transformation is used to control the correlation among nonnormal input variables. Through comparison on computational efciency between probabilistic collocation point method and random sampling method in the sparse polynomial chaotic expansion, the probabilistic collocation point method is selected to suit samples allocation. By applying in IEEE-9, IEEE-30, and IEEE-118 standard test systems, respectively, the results show that the proposed method can dramatically reduce the deterministic PLF calculations quantity compared with the traditional stochastic response surface method (SRSM), and possesses the same accuracy as Monte Carlo method, and is an efective solving method for some high-dimensional input variable systems.

        Ginsenoside Rg3, a promising agent for NSCLC patients in the pandemic: a large-scale data mining and systemic biological analysis

        Introduction: Non-small cell lung cancer (NSCLC) patients are particularly vulnerable to the Coronavirus Disease-2019 (COVID-19). Currently, no anti-NSCLC/COVID-19 treatment options are available. As ginsenoside Rg3 is beneficial to NSCLC patients and has been identified as an entry inhibitor of the virus, this study aims to explore underlying pharmacological mechanisms of ginsenoside Rg3 for the treatment of NSCLC patients with COVID-19. Methods: Based on a large-scale data mining and systemic biological analysis, this study investigated target genes, biological processes, pharmacological mechanisms, and underlying immune implications of ginsenoside Rg3 for NSCLC patients with COVID-19. Results: An important gene set containing 26 target genes was built. Target genes with significant prognostic value were identified, including baculoviral IAP repeat containing 5 (BIRC5), carbonic anhydrase 9 (CA9), endothelin receptor type B (EDNRB), glucagon receptor (GCGR), interleukin 2 (IL2), peptidyl arginine deiminase 4 (PADI4), and solute carrier organic anion transporter family member 1B1 (SLCO1B1). The expression of target genes was significantly correlated with the infiltration level of macrophages, eosinophils, natural killer cells, and T lymphocytes. Ginsenoside Rg3 may benefit NSCLC patients with COVID-19 by regulating signaling pathways primarily involved in anti-inflammation, immunomodulation, cell cycle, cell fate, carcinogenesis, and hemodynamics. Conclusions: This study provided a comprehensive strategy for drug discovery in NSCLC and COVID-19 based on systemic biology approaches. Ginsenoside Rg3 may be a prospective drug for NSCLC patients with COVID-19. Future studies are needed to determine the value of ginsenoside Rg3 for NSCLC patients with COVID-19.

        Transcriptome-Wide Analyses Provide Insights into Development of the Hedychium coronarium Flower, Revealing Potential Roles of PTL

        The flower of Hedychium coronarium possesses highly specialized floral organs: a synsepalous calyx, petaloid staminodes and a labellum. The formation of these organs is controlled by two gene categories: floral organ identity genes and organ boundary genes, which may function individually or jointly during flower development. Although the floral organogenesis of H. coronarium has been studied at the morphological level, the underlying molecular mechanisms involved in particular organ morphologies that emerge in flower development still remain poorly understood. Here, we used comparative transcriptomics combined with Real-time quantitative PCR to investigate gene expression patterns of ABC-class genes in H. coronarium flowers, as well as the homolog of PETAL LOSS (HcPTL). Our studies found that stamen/petal identity or stamen fertility in H. coronarium was not necessarily correlated with the differential expression of HcAP3 and HcAG. We also found a novel spatio-temporal expression pattern for HcPTL mRNA, suggesting it may have evolved a lineage-specific role in the morphogenesis of the Hedychium flower. Our study provides a new transcriptome reference and a functional hypothesis regarding the role of a boundary gene in organ fusion that should be further addressed through phylogenetic analyzes of this gene, as well as functional studies.
