In order to study the damage mechanism of surrounding rocks under water-rock coupling, an experimental study on the microscopic law of water absorption and mechanics of sandy mudstone was carried out in the sandy mudstone stratum of Wanfu coal mine in Shandong, China. The results indicate that the strength of sandy mudstone exhibits three stages of relative softening, rapid softening, and decelerated softening. The speed of water molecules entering the microscopic pore structure was analyzed based on the percentage of water molecules in the pore structure. The acoustic emission energy release process is divided into energy accumulation stage, stable release stage and intense release stage. With the increase of water content saturation, (Spectral width of the multifractal spectrum) tends to decrease and the distribution of events is gradually uniform. The action of water gradually expands and penetrates the cracks and pores inside the sandy mudstone, which makes f (Frequency of occurrence of the signal) change from negative to positive values.

Background: Evidence supporting an association between the 8q24 rs4242382-A polymorphism and prostate cancer (PCa) risk has been reported in North American and Europe populations, though data from Asian populations remain limited. We therefore investigated this association by clinical detection in China, and meta-analysis in Asian, Caucasian and African-American populations. Materials and Methods: Blood samples and clinical information were collected from ethnically Chinese men from Northern China with histologically-confirmed PCa (n=335) and from age-matched normal controls (n=347). The 8q24 (rs4242382) gene polymorphism was genotyped by polymerase chain reaction-high-resolution melting analysis. We initially analyzed the associations between the risk allele and PCa and clinical covariates. A meta-analysis was then performed using genotyping data from a total of 1,793 PCa cases and 1,864 controls from our study and previously published studies in American and European populations, to determine the association between PCa and risk genotype. Results: The incidence of the risk allele was higher in PCa cases than controls (0.222 vs 0.140, $P=7.3{\times}10^{-5}$), suggesting that the 8q24 rs4242382-A polymorphism was associated with PCa risk in Chinese men. The genotypes in subjects were in accordance with a dominant genetic model (ORadj=2.03, 95%CI: 1.42-2.91, $Padj=1.1{\times}10^{-4}$). Presence of the risk allele rs4242382-A at 8q24 was also associated with clinical covariates including age at diagnosis ${\geq}65$ years, prostate specific antigen >10 ng/ml, Gleason score <8, tumor stage and aggressive PCa, compared with the non-risk genotype ($P=4.6{\times}10^{-5}-3.0{\times}10^{-2}$). Meta-analysis confirmed the association between 8q24 rs4242382-A polymorphism and PCa risk (OR=1.62, 95%CI: 1.39-1.88, $P=1.0{\times}10^{-5}$) across Asian, Caucasian and African American populations. Conclusions: The replicated data suggest that the 8q24 rs4242382-A variation might be associated with increased PCa susceptibility in Asian, Caucasian and African American populations. These results imply that this polymorphism may be a useful risk biomarker for PCa in multi-ethnic populations.

A novel esterase gene, est01, was successfully unearthed from a biogas digester microbiota metagenomic library. The 1,194 bp est01 gene encodes a protein of 44,804 Da (designated Est01). The amino acid sequence of Est01 shows only moderate (33%) identity to a lipase/ esterase. Phylogenetic analysis and biochemical characterization confirmed that Est01 is a new member of family VIII esterases. The purified Est01 from recombinant Escherichia coli BL21 (DE3) showed high hydrolytic activity against short-chain fatty acid esters, suggesting that it is a typical carboxylesterase rather than a lipase. Furthermore, the Est01 was even active at 10°C (43% activity remained), with the optimal temperature at 20°C, and had a broad pH range from 5.0 to 10.0, with the optimal pH of 8.0. These properties suggest that Est01 is a cold-adaptive esterase and could have good potential for low-temperature hydrolysis application.

Multiple parallel inverters have multiple resonant frequencies that are influenced by many factors. This often results in stability and power quality problems. This paper develops a multiple input multiple output model of grid-connected inverter systems using a closed-loop transfer function. The influence factors of the resonant characteristics are analyzed with the developed model. The analysis results show that the resonant frequency is closely related to the number, type and composition ratio of the parallel inverters. To suppress resonance, a scheme based on virtual impedance is presented, where the virtual impedance is emulated in the vicinity of the resonance frequency. The proposed scheme needs one inverter with virtual impedance control, which reduces the design complexity of the other inverter controllers. Simulation and experimental tests are carried out on two single phase converter-based setups. The results validate the correctness of the model, the analytical results and the resonant suppressing scheme.

Heat shock proteins (HSPs) play important roles in the adaptation of organisms to environmental changes. In this study, we explore the role of HSPs during heat stress in Sesamia inferens (Walker) (Lepidoptera: Noctuidae), an important pest of rice. In order to understand themechanistic basis of ecological adaptability and stress tolerance, we analyzed the expression profiles of genes encoding five S. inferens heat shock proteins (Sihsps) across temperature gradients by real-time quantitative PCR. Our results indicated that genes encoding ribosomal protein S13 (RPS13) and β-actin (ACTB) were the most stable reference genes under high temperature stress. The mRNA levels of Sihsp19.6, Sihsp20.6 and Sihsp83 were significantly upregulated by heat stress; however, the expression of Sihsp21.4 and Sihsc70 did not vary significantly. The massive increase in the expression of Sihsp19.6 and Sihsp20.6 by thermal stress suggests a critical role in preventing cellular damage to S. inferens. Moreover, the threshold temperatures (39 °C for Sihsp19.6, Sihsp20.6 and Sihsp83)may partly explain the more southern distribution of S. inferens. Our results also support the contention that the absence of introns may favor the expression of stress-responsive genes (e.g. Sihsp19.6, Sihsp20.6 and Sihsp83 in the present study).

The fallwebworm, Hyphantria cunea (Drury) (Lepidoptera: Arctiidae), is an invasive and important pest in China. Understanding the physiological–biochemical characteristics of the overwintering pupae is essential for its cold tolerance and potential distribution in the northeast of China. Results indicated that the supercooling point (SCP) of H. cunea overwintering pupae varied significantly among Jilin city (43°N, 126°E), Jilin Province, Shenyang (41.8°N, 123.4°E), Liaoning Province and Yingkou (40.39°N, 122.13°E), Liaoning Province. The average SCP in Jilin was the lowest (−23.49 ± 0.46 °C) in those of three areas. The water content in overwintering pupae had a significant positive correlation with the SCP (R = 0.848,P = 0.03). Contents of alanine (ALA) (26.00%), lysine (LYS) (30.51%) and arginine (ARG) (22.88%) in the overwintering pupae's hemolymph were higher than others. During the overwintering, the fat content in pupae's bodies decreased gradually. Analysis of variance (ANOVA) indicated that the content of trehalose in the body of H. cunea was remarkably different (P= 0.007) month by month, with the maximum in March (2.88 mg·mL−1) and the minimum (0.40 mg·mL−1) in November. Pearson's correlation test indicated that SCP had a significant negative correlation with the SOD enzyme activity (R = −0.850; P = 0.008). Thermal hysteresis activity (THA) of hemolymph was 0.68°C and 0.7–0.8°C for H. cunea pupae and larva, respectively, using differential scanning calorimetry, which suggests that the H. cunea can improve their cold tolerance through producing antifreeze proteins. These results could help us to predict the distribution of the H. cunea in the northeast of China.

Xylanase inhibitors are known to play important roles in response to fungal pathogens attack. WRKY transcription factors are also involved in plant defence against pathogens. However, whether WRKY transcription factors interacting with rice xylanase inhibitor RIXI remains unknown. 13 W-boxes, binding sites for WRKY transcription factors were found in RIXI promoter sequence of rice by analyzing RIXI promoter. ChIP-PCR analysis and dual luciferase reporter system assay were used to identify interaction between xylanase inhibitor RIXI and WRKY transcription factors. Results showed OsWRKY6 and OsWRKY46 directly bound to RIXI promoter and regulated RIXI expression diferently. Over-expressing OsWRKY6 and OsWRKY46 transgenic plants were achieved, and OsWRKY6 and OsWRKY46 overexpressed transgenic lines showed higher expression levels of RIXI and higher resistance to Magnaporthe oryzae G11 (M. oryzae G11) compared to wild type (WT). Increased RIXI expression in OsWRKY6 and OsWRKY46 overexpressed transgenic lines was accompanied by up-regulation of pathogenesis-related genes (PR genes). Moreover, higher levels of H2O2 and higher activities of antioxidant enzymes were found in transgenic lines compared to WT plants. Results indicated that over-expression of OsWRKY6 and OsWRKY46 in transgenic lines maybe enhanced rice disease resistance by regulating RIXI expression.
