In eukaryotic cells, a key RNA processing step to generate mature mRNA is the coupled reaction for cleavage and polyadenylation (CPA) at the 3′ end of individual transcripts. Many transcripts are alternatively polyadenylated (APA) to produce mRNAs with different 3′ ends that may either alter protein coding sequence (CDS-APA) or create different lengths of 3′UTR (tandem-APA). As the CPA reaction is intimately associated with transcriptional termination, it has been widely assumed that APA is regulated cotranscriptionally. Isoforms terminated at different regions may have distinct RNA stability under different conditions, thus altering the ratio of APA isoforms. Such differential impacts on different isoforms have been considered as post-transcriptional APA, but strictly speaking, this can only be considered “apparent” APA, as the choice is not made during the CPA reaction. Interestingly, a recent study reveals sequential APA as a new mechanism for post-transcriptional APA. This minireview will focus on this new mechanism to provide insights into various documented regulatory paradigms.
A high-stability neutron generator for industrial online elemental analysis

The yield stability of the neutron generator directly affects the accuracy of elemental analysis. This paper presents an industrial fully automatic neutron generator with a 48 mm neutron tube based on PLC to improve the stability and reliability of the neutron generator in industrial applications. By integrating a Kalman Filter with the PID algorithm in a PLC, the neutron yield of the generator is remarkably stabilized, achieving 1 × 10<sup>8</sup>n/s. The neutron generator has been employed for industrial online elemental analysis. The results demonstrate that only a slight fluctuation of ±0.82 % exists in the neutron yield, and the reproducibility of the generator holds at a significant level of 0.05. This improved neutron generator can be applied to the online bulk analysis of carbon in coal-fired power stations and absolute measurement of neutron source emission rate.

Two transition metal ions (Fe3+ and Cu2+) and a rare earth metal ion (Ce3+) were selected to coordinate with amidoximated polyacrylonitrile (PAN) nanofibrous membrane for preparing three metal modified PAN nanofibrous membrane complexes (M-AO-n-PANs, M = Fe, Cu, or Ce) as the heterogeneous Fenton catalysts for the dye degradation in water under visible irradiation. The coordination kinetics of three metal ions with modified PAN nanofibrous membranes was studied and the catalytic properties of the resulting complexes were also compared. The results indicated that increasing metal ion concentrations in solution or higher coordination temperature led to a significant increase in metal content, particularly in Fe and Cu contents of the complexes. Their coordination process could be described using Langmuir isotherm and pseudo-second-order kinetic equations. Moreover, Fe-AO-n-PAN had the best photocatalytic efficiency for the dye degradation in acidic medium, but a lower photocatalytic activity than Cu-AO-n-PAN in alkali medium.
Mineralization of calcium phosphate in reverse microemulsion

In the present study, reverse microemulsion was prepared to regulate the mineralization of calcium phosphate. Calcium chlorideand sodium hydrogen phosphate aqueous solution were, respectively, dropped into the mixture of Span 80, Tween 80, andn-heptaneto get two kinds of emulsions.n-Buranol was used to adjust the emulsions to transparent state. Calcium phosphate was prepared byadding phosphate microemulsion to Calcium microemulsion. Heating method was used to induce the phase separation. After beingwashed with acetone and ethanol, the mineralized deposition were studied with scanning electron microscopy (SEM), X-ray dirac-tion (XRD) and transmission electron microscopy (TEM). The results indicated that the minerals in the deposition were mainlymonetite (DCPA) and the nanosized needle-like DCPA exhibited the preferential orientation along 200 direction..

The α-gliadins from Crithopsis delileana (Schult) Roshev (2n=2x=14, KK) were investigated by Acid polyacrylamide gel electrophoresis (A-PAGE) analysis. It was indicated that the electrophoresis mobility of gliadins from C.delileana had obvious difference with those from common wheat in α, γand ω region. Using primers designed from published sequences of α-gliadin genes, three α-gliadin genes were isolated from C. delileana, which were designated as gli-ka1,gli-ka2 and gli-ka3, respectively. Two in-frame stop codons were found in the coding sequences of gli-ka3, indicating that gli-ka3 could be a pseudogene. The gli-ka2 was a gliadin with an odd number of cysteines, resulting from a non-synonymous mutation. This change might lead to the interactive behavior of gli-ka2. Three α-gliadin genes of C. delileana had the similar but not identical primary structures to the corresponding gene sequences from other wheat related species. By the alignment of α-gliadin genes from Triticeae,phylogenetic analysis indicated that three α-gliadin genes of C. delileana clustered together with all α-gliadin genes from Ee genome of Lophopyrum elongatum by an interior paralleled branch.

The first tuber mustard calmodulin-like (CML) gene BjAAR1 (Brassica juncea var. tumida Tsen et Lee Abiotic stress and Abscisic acid (ABA) Responsive gene 1) was cloned and characterized. The protein encoded by BjAAR1 contains four predicted Ca2+ binding sites (EF-hand motif) and its recombinant protein can bind Ca2+ in vitro. qRT-PCR showed that the expression level of BjAAR1 was rather high in non-swollen stem of tuber mustard and largely reduced in swollen stem. Expression of BjAAR1 enhanced ABA- and stress-induced gene expression in Arabidopsis (Arabidopsis thaliana). Transgenic plants also exhibited hypersensitivity to NaCl, mannitol, and ABA during the seed germination and post-germination stages. ABA biosynthesis inhibitor, norflurazon (NF), rescued hypersensitivity phenotype of transgenic plants to NaCl and mannitol, indicating that BjAAR1 functions in multiple abiotic stresses response through ABA-dependent process.
MicroRNA Expression Profile Analysis Reveals Diagnostic Biomarker for Human Prostate Cancer

Prostate cancer is a highly prevalent disease in older men of the western world. MicroRNAs (miRNAs) are small RNA molecules that regulate gene expression via posttranscriptional inhibition of protein synthesis. To identify the diagnostic potential of miRNAs in prostate cancer, we downloaded the miRNA expression profile of prostate cancer from the GEO database and analysed the differentially expressed miRNAs (DE-miRNAs) in prostate cancerous tissue compared to non-cancerous tissue. Then, the targets of these DE-miRNAs were extracted from the database and mapped to the STRING and KEGG databases for network construction and pathway enrichment analysis. We identified a total of 16 miRNAs that showed a significant differential expression in cancer samples. A total of 9 target genes corresponding to 3 DE-miRNAs were obtained. After network and pathway enrichment analysis, we finally demonstrated that miR-20 appears to play an important role in the regulation of prostate cancer onset. MiR-20 as single biomarker or in combination could be useful in the diagnosis of prostate cancer. We anticipate our study could provide the groundwork for further experiments.

Fangchinoline (Fan) inhibits cell proliferation and induces apoptosis in several cancer cell lines. The effects of Fan on cell growth and proliferation in breast cancer cells remain to be elucidated. Here, we show that Fan inhibited cell proliferation in the MDA-MB-231 breast cancer cell line through suppression of the AKT/Gsk-3beta/cyclin D1 signaling pathway. Furthermore, Fan induced apoptosis by increasing the expression of Bax (relative to Bcl-2), active caspase 3 and cytochrome-c. Fan significantly inhibited cell proliferation of MDA-MB-231 cells in a concentration and time dependent manner as determined by MTT assay. Flow cytometry analysis demonstrated that Fan treatment of MDA-MB-231 cells resulted in cell cycle arrest at the G1 phase, which correlated with apparent downregulation of both mRNA and protein levels of both PCNA and cyclin D1. Further analysis demonstrated that Fan decreased the phosphorylation of AKT and GSK-3beta. In addition, Fan up-regulated active caspase3, cytochrome-c protein levels and the ratio of Bax/Bcl-2, accompanied by apoptosis. Taken together, these results suggest that Fan is a potential natural product for the treatment of breast cancer.
