The first step of the branch pathway in tryptophan biosynthesis is catalyzed by anthranilate synthase, which is subjected to feedback inhibition by the end product of the pathway. The trpE^(FBR) gene from a mutant Escherichia coli strain coding for anthranilate synthase that was insensitive to feedback inhibition by tryptophan has been cloned. To identify the amino acid changes involved in the feedback regulation of anthranilate synthase, the nucleotide sequence of the mutant trpE^(FBR) gene was determined. Sequence analysis of the trpE^(FBR) gene revealed that four bases were changed in the structural gene while alteration was not found in the 5` control region. Among these base changes, only two base substitutions caused the alterations in amino acid sequences. From the results of restriction fragment exchange mapping, the 61st nucleotide, C to A substitution, that changed Pro^(21)→Ser was identified as the cause of the desensitization to feedback inhibition by tryptophan. Additional feedback-resistant enzymes of the E. coli anthranilate synthases were constructed by sitedirected mutagenesis to examine the effect of the Ser^(40)→Arg^(40) change found in the trpE^(FBR) gene of Brevibacterium lactofermentum. From the feedback inhibition analysis, the Pro^(21)→Ser and Ser^(40)→Arg mutants maintained about 50% and 90% of their maximal activities, respectively, even at the extreme concentration of 10 mM tryptophan. From these results, we suggest that the Pro^(21) and Ser^(40) residues are involved in the tryptophan binding in the E. coli enzyme.

A 44-year-old woman presented with asymptomatic, multiple, variable sized (3 mm to 13 mm, in diameter) brownish macule on the posterior side of right thigh and she had never been treated before. The number and the size of the lesion had not been changed much from that had first appeared 6 month before visiting our clinic. Physical examination revealed no underlying mass or nodule around the lesion and no other similar skin lesion at other site of the body. She had no previous medical history. Though the first assessment of the clinician was scar with postinflammatory hyperpigmentation, as she had no previous traumatic episodes at that site, skin biopsy was performed. There were scattered giant cells demonstrating bizarre or scalloped outlines which were positive for factor XIIIa and vimentin. There were also a proliferation of small blood vessels and a sparse inflammatory infiltrate in the interstitium. The histopathologic features were consistent with multinucleate cell angiohistiocytoma. Although the patient was recommended to get a complete excisional biopsy, she refused any further treatment.

A case of basal cell carcinoma of the nail bed
( Won Choi ),( Joon Soo Lim ),( Kyoung Geun Lee ),( Hyung Jin Hahn ),( Seung Hyun Cheong ),( Ki Bum Myung ) 대한피부과학회 2018 대한피부과학회 학술발표대회집 Vol.70 No.1
Basal cell carcinoma is widely regarded as the most common malignant skin tumor and usually seen in head and neck. Ralely, nail bed and nail folds can be atypical sites for BCC. The initial diagnosis of chronic paronychia lead to delay in diagnosis so that taking history and adequate biopsy are essential in making timely diagnosis. Previous study reported that the BCC of nail bed was more often on the fingers than on the toes and had a slight predilection to occur in men. Ulceration was the most common presentation of nail unit BCC, which was noted in more than one-half of patients. Mohs micrographic surgery with second intension healing is effective to make excellent cosmetic results. The incidence of metastasis is extremely rare and same as other BCCs, which has been reported with incidence rates of 0.0028% to 0.55%. A 55-year-old man presented with ill-defined 0.8 cm sized pigmented lesion on left thumb nail bed without trauma history. Duration of the lesion was 1 month. Specimen showed that basophilic cells formed budding tumor proliferations extending from the undersurface of the epidermis to the dermis. HMB45 and S-100 showed negative results. He has no any other past history and laboratory study showed normal findings. The patient was finally diagnosed as BCC of the nail bed and referred to plastic surgeon for wide excision and full thickness skin graft. Herein we report a relatively rare case of BCC of the nail bed.
