Objectives: cDNA microarray is an effective method to snapshot gene expression. Functional clustering of gene expressions can identify herbal medicine mechanisms. Much microarray data is available for various herbal medicines. This study compares regulated genes with herbal medicines to evaluate the nature of the drugs. Methods: Published microarray data were collected. Total RNAs were prepared from dissociated hippocampal dissociate cultures which were given hypoxic shock in the presence of each herbal medicine. Up- or downregulated genes higher than Global M value 0.5 were selected, clustered in functional groups, and compared with various herbal treatments. Results: 1. Akt2 was upregulated by Acorus gramineus SOLAND, Arisaema amurense var. serratum $N_{AKAI}$ and Coptis chinensis $F_{RANCH}$, and they belong to Araceae herb. 2. Nf-${\kappa}b1$, Cd5, $Gn{\gamma}7$ and Sgne1 were upregulated by Arisaema amurense var. serratum $N_{AKAI}$, Coptis chinensis $F_{RANCH}$ and Rheum coreanum $N_{AKAI}$. 3. Woohwangcheongsim-won, Sohaphyang-won and Scutellaria baicalensis $G_{EORGI}$ downregulated Scp2 and upregulated Tsc2. Woohwangcheongsim-won and Sohaphyang-won upregulated Hba1 and downregulated Myf6. 4. Sohaphyang-won and Scutellaria baicalensis $G_{EORGI}$ downregulated Slc12a1. 5. Woohwangcheongsim-won and Arisaema amurense var. serratum $N_{AKAI}$ upregulated $Rar{\alpha}$, Woohwangcheongsim-won and Coptis chinensis $F_{RANCH}$ downregulated Rab5a and $Pdgfr{\alpha}$, and Woohwangcheongsim-won and Rheum coreanum $N_{AKAI}$ upregulated $Plc{\gamma}1$ and downregulated Pla2g1b and Slc10a1. Conclusions: By clustering microarray, genes are commonly identified to be either up- or downregulated. These results will provide new information to understand the efficacy of herbal medicines and to classify them at the molecular level.

<P><B>Abstract</B></P> <P>A previous study showed that small G protein signaling modulator 3 (SGSM3) was highly correlated with Cx43 in heart functions and that high levels of SGSM3 may induce Cx43 turnover through lysosomal degradation in infarcted rat hearts. Here, we investigated the protective effects of kenpaullone on cardiomyocytes following H<SUB>2</SUB>O<SUB>2</SUB>-induced oxidative stress mediated by the interaction of SGSM3 with Cx43. We found that the gap junction protein Cx43 was significantly down-regulated in an H<SUB>2</SUB>O<SUB>2</SUB> concentration-dependent manner, whereas expression of SGSM3 was up-regulated upon H<SUB>2</SUB>O<SUB>2</SUB> exposure in H9c2 cells. The effect of kenpaullone pretreatment on H<SUB>2</SUB>O<SUB>2</SUB>-induced cytotoxicity was evaluated in H9c2 cells. H<SUB>2</SUB>O<SUB>2</SUB> markedly increased the release of lactate dehydrogenase (LDH), while kenpaullone pretreatment suppressed LDH release in H9c2 cells. Moreover, kenpaullone pretreatment significantly reduced ROS fluorescence intensity and significantly down-regulated the level of apoptosis-activating genes (cleaved caspase-3, cleaved caspase-9 and cytochrome C), autophagy markers (LC3A/B), and the Cx43-interacting partner SGSM3. These results suggest that kenpaullone plays a role in protecting cardiomyocytes from oxidative stress and that the turnover of Cx43 through SGSM3-induced lysosomal degradation underlies the anti-apoptotic effect of kenpaullone.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Cx43 was down-regulated in an H<SUB>2</SUB>O<SUB>2</SUB> concentration-dependent manner, whereas SGSM3 was up-regulated in H9c2 cells. </LI> <LI> Kenpaullone pretreatment reduced ROS fluorescence intensity and apoptosis-activating gene levels. </LI> <LI> Kenpaullone plays a role in protecting cardiomyocytes from oxidative stress. </LI> </UL> </P>

<P>Abstract</P><P>Midbrain dopamine (mDA) neurons play critical roles in the regulation of voluntary movement and their dysfunction is associated with Parkinson’s disease. Pitx3 has been implicated in the proper development of mDA neurons in the substantia nigra pars compacta, which are selectively lost in Parkinson’s disease. However, the basic mechanisms underlying its role in mDA neuron development and/or survival are poorly understood. Toward this goal, we sought to identify downstream target genes of Pitx3 by comparing gene expression profiles in mDA neurons of wild-type and Pitx3-deficient aphakia mice. This global gene expression analysis revealed many potential target genes of Pitx3; in particular, the expression of vesicular monoamine transporter 2 and dopamine transporter, responsible for dopamine storage and reuptake, respectively, is greatly reduced in mDA neurons by Pitx3 ablation. In addition, gain-of-function analyses and chromatin immunoprecipitation strongly indicate that Pitx3 may directly activate transcription of vesicular monoamine transporter 2 and dopamine transporter genes, critically contributing to neurotransmission and/or survival of mDA neurons. As the two genes have been known to be regulated by Nurr1, another key dopaminergic transcription factor, we propose that Pitx3 and Nurr1 may coordinately regulate mDA specification and survival, at least in part, through a merging and overlapping downstream pathway.</P>

Transumbilical single-port laparoscopic donor nephrectomy (SPLDN) is a novel, rapidly evolving, minimally invasive treatment modality for kidney transplantation. This method causes minimal parietal injury, has cosmetic advantages, and allows rapid recovery because of low postoperative pain and short hospital stay. Like other abdominal surgeries, when conducted by experienced laparoscopic surgeons, it can meet the same graft requirements as conventional laparoscopic surgery. Here, we report the first two cases of transumbilical SPLDN at Daejeon St. Mary’s Hospital, The Catholic University of Korea. We used the umbilicus as a common path for laparoscopic procedures and as a route for specimen retrieval. The operating times were 230 and 265 minutes in cases 1 and 2, respectively. No intra- or postoperative complications were noted. In case 1, the wound length was 4 cm and duration of hospitalization was 2 days. In case 2, the wound length was only 2.5 cm, and the duration of hospitalization was only 1 day.

<P>The use of herbal medicines in the therapeutic treatment of atopic dermatitis (AD) has been suggested recently. The present study examined whether selected herbal extracts fermented in Lactobacillus plantarum (FHE) possessed anti-AD properties. In addition, the study assessed the increased bioavailability of these herbal extracts both in vitro and in vivo. The data from these experiments revealed that FHE inhibited the proliferation of splenic T and B cells in a dose-dependent manner, when activated with their mitogens. Moreover, the expression of Th1/Th2 mRNA cytokines (IL-2, IL-4, IL-5, IL-13) from mouse splenocytes was inhibited severely as was cyclosporine A. Furthermore, the release of &bgr;-hexosaminidase in RBL-2H3 mast cells was suppressed significantly. FHE also reduced the plasma level of IgE in dust mite extract-induced AD-like NC/Nga mice. More dramatic results were found in the histological changes, which were observed by hematoxylin–eosin and toluidine blue staining, as well as in the macroscopic features on dorsal lesions of AD-like NC/Nga mice. In conclusion, the results presented in this study suggest that FHE may have therapeutic advantages for the treatment of AD due to its increased immune-suppressive and increased absorptive effects, which were fortified by L. plantarum fermentation. Novel Noncontact Meibography With Anterior Segment Optical Coherence Tomography: Hosik Meibography

PURPOSE:: To present a novel noncontact meibography system with anterior segment optical coherence tomography (OCT), which is widely used in clinics for the anterior segment (cornea, anterior chamber angle, etc.), and compare the results with preexisting infrared meibography. METHODS:: This research was carried out at the Seoul St Mary’s Hospital with 2 volunteers. Preexisting infrared meibography was performed on the subjects, and photographs of the meibomian gland were taken again with the anterior segment OCT. With the anterior segment OCT, a tomogram of the meibomian gland could be taken and a picture of the whole meibomian gland could be taken from the infrared images for monitoring. RESULTS:: The resolution of the preexisting infrared meibography was a pixel size of 640 × 480. In the anterior segment OCT, the meibomian glands were clearly identified just beneath the palpebral conjunctiva. There was no problem in grading the meibomian gland with the infrared images for monitoring. The resolution was 239 × 178 or 129 × 95 pixels, depending on the save option of the photograph. CONCLUSIONS:: This novel meibography using anterior OCT yielded meibomian gland tomograms without any additional equipment, and overall infrared meibography was possible for grading. We believe this new technique will be useful in clinics for meibomian gland dysfunction and the like.
