        Deletion of Superoxide Dismutase Gene of Bombyx mori Nuclear Polyhedrosis Virus Affects Viral DNA Replication

        Superoxide dismutase (SOD) is an important enzyme which catalyzes superoxide radicals to hydrogen peroxide. A Cu, Zn sod-like gene was found in Bombyx mori nuclear polyhedrosis virus encoding 151 amino acids. To demonstrate its function, a recombinant virus named dsBmNPV with deleted sod gene was constructed. It was discovered that the sod gene was not essential for viral replication. Studies on growth of budded virus in BmN cells and superoxide dismutase and catalase activities in vivo after dsBmNPV infection showed that the titer of dsBmNPV decreased obviously comparing to wild type BmNPV, the sod gene was effective on genomic DNA replication of baculovirus, the peak of SOD activity of silkworm infected with wt-BmNPV appeared between 36 and 48 hrs post infection, and with dsBmNPV, it did not appear. And the changes of CAT activity after infection were similar to SOD activity.

      • Semi-Global Output Feedback Tracking Control for Fully Actuated Ships

        In this paper a new output feedback trajectory tracking controller for fully actuated ships is designed whenonly the position and the heading measurements are available. The considered 3 DOF ship model includes the Coriolisand Centripetal induced forces and moments and a nonlinear damping term. The gains of the constructed observer arenonlinear and time-variant, which can be used to globally dominate the difficult term resulting from the Coriolis andCentripetal term in the observer error dynamics. The acquired output feedback tracking control system is proveduniformly semi-globally exponential stable by applying nonlinear cascaded system theory. Furthermore, simulationstudies are carried out by utilizing Matlab’s Simulink Toolbox. Simulation results on a model ship Cybership II arepresented to validate the proposed output feedback tracking controller with a separate stability result for observer.

        Degradation of 1,4-dioxane by Newly Isolated Acinetobacter sp. M21 with Molasses as the Auxiliary Substrate

        The elimination of 1,4-dioxane (dioxane), a persistent organic pollutant, is a great challenge owing to its high hydrophilicity and chemical stability. Cometabolic bioremediation technology is an effective approach to remove many organic pollutants. Because of its eco-friendly and inexpensive properties, molasses is widely used as an auxiliary biomaterial to clean up compound-contaminated sites. In this study, a newly isolated bacterium Acinetobacter sp. M21 could effectively remove dioxane using molasses without any apparent lag phase. Under the optimized molasses dosage of 0.3%, M21 could remove 500 mg/L dioxane by 60.0 ± 2.8% within 20 days with a maximum dioxane degradation rate of 1.3 ± 0.2 mg-dioxane/L/h in the first day, and exhibited extraordinary dioxane tolerance up to 1,000 mg/L, while so high dose of dioxane negatively affected the cell growth. The degradation pathway of dioxane was also determined, and was supported by the detection of 2-hydroxyethoxyacetic acid as the key metabolite of dioxane. High level degradation activity of M21 to 20 mg/L dioxane was maintained over a variable of pH (5-11), temperatures (15-45°C), and salinities (up to 8%, as NaCl wt). This is the first report linking the cometabolism of dioxane and molasses by Acinetobacter sp. M21, a bacterium that shows great potential for field dioxane bioremediation.

      • SIFT-Based 3D Feature Matching and Reconstruction with Binocular Stereo Vision

        In this paper, we address the problem of 3D (three-dimensional) feature matching and reconstruction method with SIFT(scale invariant feature transform)-based algorithm under binocular stereo vision. The technique is fulfilled by camera calibration, image processing, feature detection and 3D calculation. Zhang plane-based calibration and Harris corner point detection method are utilized for Camera calibration. SIFT algorithm is focused as feature extraction and matching algorithm to obtain the image coordinates of matching points. The obtained 3D coordinates are validated by the measurement results with a laser sensor. Finally the depth map of 3D environment is produced with the measured results. VC++.NET and OpenCV are used to compile the algorithm for the experiment in this research.

        A Bayesian Network approach for risk analysis of composite subsea pipelines subjected to falling objects

        Thermoplastic Composite Pipes (TCPs) have potentials to be used as subsea pipelines. TCPs are superior in corrosion resistance, yet vulnerable to impact damages. In this study, a Bayesian Network (BN) model was developed taking the factors, such as water depth, weather condition, seabed stiffness, material aging, etc., as variables of the BN model to evaluate the probability of failure of TCP subsea pipelines when they were subjected to falling objects from passing vessels. Case study examples of an assumed TCP pipeline in shallow water area of Bohai sea of China were used to illustrate the topology structure of the BN model and the modeling procedures of the Conditional Probability Table (CPT) of the BN model variables. The case study examples illustrated that the BN model can predict the probability of failure of a TCP pipeline and diagnose the likely causes of the failure. Additionally, the BN model can be updated by taking into account new data such as aging of materials for the entire service life of the pipeline. The predicted failure probability of the assumed TCP pipeline was not benchmarked due to lack of observation data; however, the proposed BN model is a useful tool for the integrity management of TCP pipelines when widely installed in the future.

        Robust Fuzzy Controller Design for Dynamic Positioning System of Ships

        This paper presents a robust fuzzy controller design approach for dynamic positioning (DP) system of ships using optimal H∞ control techniques. The H∞ control technique is used to exterminate the effects of environmental disturbances. Firstly, a Takagi-Sugeno (TS) fuzzy model is applied to approximate the nonlinear DP system. Next, linear matrix inequality (LMI) and general eigenvalue problem (GEVP) methods are employed to find a positive definite matrix and controller gains. The stability of the controller is proven by using Lyapunov stability theorems. A positive definite matrix is determined by solving LMI equations using robust control toolbox available in MATLAB. The obtained positive definite matrix proves that the designed fuzzy controller is stable. Finally, a uniformly ultimately bound (UUB) and control performance for the dynamic position system is guaranteed. Simulation is carried out, and results are presented to validate the effectiveness and performance of the proposed control system.

        Experimental study of cactus-like body shape on flow-induced vibration mitigation of clustered cylinders

        Vortex-Induced Vibration (VIV) is a major contributor to the fatigue damage of marine risers which are often arranged in an array configuration. In addition to helical strakes and fairings, studies have been strived in searching for possible VIV suppression techniques. Inspired by giant Saguaro Cacti, flexible cylinders of different cactus-shaped cross sections were tested in a water tunnel facility, and test results showed that cactus-like body shapes reduced VIV responses of a cylinder at no cost of significant increase of drag. A series of experiments were conducted on a pair of two tandem-arranged flexible cylinders and an array of four cylinders in a square configuration to investigate the effects of wake on the dynamic responses of cylinders and the VIV mitigation effectiveness of the cactus-like body shape. Results showed that the cylinders in a square configuration, either at the upstream or downstream positions, might have larger dynamic responses than those of a single cylinder. The cactus-like body shape could mitigate VIV responses of cylinders at upstream positions in an array configuration; however, similar to helical strakes, the mitigation efficiency was reduced on downstream cylinders. Note that the cactus-like cross-sectional shape investigated was not optimized for VIV suppression. The present study indicates that the modification of the cross-sectional shape of a cylinder to a well-designed cactus-like shape may be used as an alternative technique to mitigate the VIV of marine risers.

      • Design and Implementation of Vision-Based Transport for a Mobile Robot via Kalman Filter

        In this paper, we propose a vision-based scheme for object recognition and transport with a mobile robot. At first, the camera calibration is experimentally performed with Zhenyou Zhang"s method. A distance measurement method with monocular camera is presented ant tested. And then the proposed scheme uses HSI model as the input and the seed filling algorithm as the image segmentation approach, a Kalman Filtering algorithm is used to predict the movement of the moving target. Finally, the motion control of the pan-tilt camera and mobile robot is designed to fulfill the tracking and transport task. The experiment results on a recognition and transport task have demonstrated the robust object recognition and fast tracking capabilities of the proposed scheme.

        Porous NiS@Ni2P nanoframe as a multi-functional catalyst for enhanced oxygen evolution and urea oxidation reactions

        During water electrolysis, multifunctional electrocatalysts with outstanding functionality and endurancemust be thoughtfully constructed and designed. Below, we report a hollow NiS@Ni2P nanoframeheterostructure that works well as a substitute catalyst for the urea oxidation and general water splittingreactions. The hollow NiS@Ni2P nanoframe was synthesized by ion exchange, sulfurization and phosphating,which is favorable for achieving a heterostructure. The NiS@Ni2P catalyst exhibits excellent catalyticactivity and strong long-term stability in 1.0 M KOH solutions for the hydrogen evolution reaction, whichneeds 121 mV to achieve 10 mA cm2. And for the oxygen evolution reaction, the catalyst needs 311 mVto acquire 50 mA cm2. These advantages come from the optimal electronic structural configuration,hierarchical hollow nanoframe structure, and large surface area. It could attain 10 mA cm2 at 1.41 Vwhen utilized as a urea oxidation reaction anode, which is lower than the oxygen evolution reaction. For total water splitting and urea oxidation process, NiS@Ni2P as a bifunctional catalyst holds tremendouspromise due to its strong electrocatalytic activity, ease of manufacture, and low cost of raw ingredients.
