<P><B>Abstract</B></P> <P>Poly (ε-caprolactone) honeycomb patterned porous films were selectively functionalized on the bottom surface with polypyrrole/silver (PPy/Ag) composite via <I>in situ</I> interfacial polymerization on a reactive substrate during the breath figure procedure. For the interfacial <I>in situ</I> polymerization, poly (ε-caprolactone) solution including pyrrole was cast on a reactive ice substrate. The reactive ice substrate contains aqueous methanesulfonic acid and silver nitrate as an oxidizing agent for the pyrrole monomer. FT-IR, EDX, UV-Visible, conductivity, and wetting measurements support the formation of PPy/Ag on the bottom surface of the films. Pore size, conductivity, and wettability increased with increased concentration of pyrrole. <P><B>Abstract</B></P> <P>Moth eye-like patterned poly(vinyl alcohol) (PVA) films selectively containing titanium dioxide (TiO<SUB>2</SUB>) nanoparticles in the microdome were fabricated using polystyrene honeycomb patterned porous film as a template in which TiO<SUB>2</SUB> nanoparticles are pore-filled. The template was prepared by in situ pore filling of TiO<SUB>2</SUB> nanoparticles by the modified breath figure method, wherein the hydrolysis of titanium butoxide was performed in the condensed water droplets as microreactors. The moth eye-like patterned PVA film selectively containing TiO<SUB>2</SUB> nanoparticles in the microdome was transparent almost similar to the pristine PVA flat film. However, the film showed a reversible photochromism that changes to blue color by UV illumination and recovered by visible light. <P>Among four types of bacterial restriction enzymes that cleave a foreign DNA depending on its methylation status, type I enzymes composed of three subunits are interesting because of their unique DNA cleavage and translocation mechanisms performed by the restriction subunit (HsdR). The elucidated N-terminal fragment structure of a putative HsdR subunit from <I>Vibrio vulnificus</I> YJ016 reveals three globular domains. The nucleolytic core within an N-terminal nuclease domain (NTD) is composed of one basic and three acidic residues, which include a metal-binding site. An ATP hydrolase (ATPase) site at the interface of two RecA-like domains (RDs) is located close to the probable DNA-binding site for translocation, which is far from the NTD nucleolytic core. Comparison of relative domain arrangements with other functionally related ATP and/or DNA complex structures suggests a possible translocation and restriction mechanism of the HsdR subunit. Furthermore, careful analysis of its sequence and structure implies that a linker helix connecting two RDs and an extended region within the nuclease domain may play a central role in switching the DNA translocation into the restriction activity.</P>
Depigmenting effect of Sterculia lynchnophera on B16F10 melanoma and C57BL/6 melan-a cells

To develop a novel skin depigmenting agent from natural sources, the inhibition of melanogenesis by the Chinese herb, Sterculia lynchnophera (SL), was evaluated. Treatment of B16F10 melanoma cells and melan-a cells with SL exhibited a 32.9% and 68.2% inhibition of melanin synthesis without cytotoxicity at a concentration of 200μg/ml, respectively. This herb possessed a high free radical scavenging activity with IC_50=11.02 μM. The methanol extract of SL slightly inhibited in vitro mushroom tyrosinase activity (23.4% at a concentration of 200 μg/ml) and had a significant inhibitory effect on cellular tyrosinase activivity (48.65% and 88.56% inhibition at the concentration 200μg/ml in B16F10 cells and C57BL/6 melan-a cells, respectively). From the western blotting results, SL inhibited the expression of tyrosinase and tyrosinase related protein 1 (TRP-1). Taken together, we suggest that SL may be a safe and effective depigmentation agent.
