      • GOLPH3, a Good Prognostic Indicator in Early-stage NSCLC Related to Tumor Angiogenesis

        Lu, Ming,Tian, Yu,Yue, Wei-Ming,Li, Lin,Li, Shu-Hai,Qi, Lei,Hu, Wen-Si,Gao, Cun,Si, Li-Bo,Tian, Hui Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.14

Background: Golgi phosphoprotein-3 (GOLPH3) is implicated in cancer development and progression. The aim of this study was to evaluate the prognostic significance of GOLPH3 protein and its association with tumor angiogenesis in patients with early-stage NSCLC. Materials and Methods: Immunohistochemistry was performed to determine GOLPH3 protein expression and allow assessment of intratumoral microvessel density (MVD) by counting CD-34 positive immunostained endothelial cells. Correlations of expression with MVD, clinicopathologic features and clinical prognosis were analyzed. Results: A notably higher level of GOLPH3 expression was found in early-stage NSCC tissues at the protein level. However, we do not find any correlation between GOLPH3 expression and clinicopathologic features (p>0.05), although higher MVD was positively associated with GOLPH3 overexpression (p<0.001). Expression of GOLPH3 was found to be an independent prognostic factor in early-stage NSCLC patients, those expressing high levels of GOLPH3 exhibiting a substantially lower 5-year overall survival than GOLPH3-negative patients (adjusted HR =1.899, 95% CI: 1.021-3.532, p=0.043). Conclusions: High expression of the GOLPH3 protein is common in early-stage NSCC, and is closely associated with tumor progression, increased tumor angiogenesis, and poor survival. We conclude a possibility of its use as a diagnostic and prognostic marker in early-stage NSCC patients.

Plant Virome Analysis by the Deep Sequencing of Small RNAs of Fritillaria thunbergii var. chekiangensis and the Rapid Identification of Viruses

        Lu-xi Chen,Hang-kai Pan,Yu-tian Tao,Dang Yang,Hui-min Deng,Kai-jie Xu,Wen-bin Chen,Jun-min Li 한국식물병리학회 2022 Plant Pathology Journal Vol.38 No.5

Thunberg fritillary (Fritillaria thunbergii), a perennial used in traditional Chinese herbal medicine, is a members of the family Liliaceae. The degeneration of germplasm is a severe problem in the production of Fritillaria thunbergii var. chekiangensis. However, no information about viral infections of F. thunbergii var. chekiangensis has been reported. In this study, we sequenced the small RNAs of F. thunbergii var. chekiangensis from leaves and bulbs, and viruses were identified using a phylogenetic analysis and BLAST search for sequence. In addition, multiplex reverse transcriptase-polymerase chain reaction (RT-PCR) was used to rapidly detect viruses in this variety. Our study first reported that five viruses infected F. thunbergii var. chekiangensis. Among them, fritillary virus Y (FVY), lily mottle virus (LMoV), Thunberg fritillary mosaic virus (TFMV), and hop yellow virus (HYV) had been reported in F. thunbergii, while apple stem grooving virus was first reported in the genus Fritillaria. A multiplex RT-PCR method was developed to rapidly test the four viruses FVY, LMoV, TFMV, and HYV in F. thunbergii var. chekiangensis. Our results provide a better understanding of the infection of F. thunbergii var. chekiangensis by viruses and a basic reference for the better design of suitable control measures.

Selection of reference genes for RT-qPCR analysis in Trichogramma chilonis (Hymenoptera: Trichogrammatidae)

        Xie Lian-Cheng,Tian Jun-Ce,Lu Yan-Hui,Xu Hongxing,Zang Lian-Sheng,Lu Zhongxian,Jin Lin-Hong 한국응용곤충학회 2021 Journal of Asia-Pacific Entomology Vol.24 No.3

Trichogramma chilonis is an important natural enemy for control of various Lepidoperan crop pests. The biology of T. chilonis is well-studied, but the molecular mechanisms of this biology require further study. Screening suitable reference genes is a vital step for use of RT-qPCR to understand underlying molecular physiology. In the present study, nine candidate reference genes including elongation factor 2 (EF2), ribosomal proteins (RPS23, RPL13, and RPL44), malate dehydrogenase (MDH), eukaryotic translation initiation factor 3 subunit F (EIF3F), zinc finger protein 268 (ZFP268), muscle specific protein 20 (MP20), and ATP synthase subunit alpha (ATP5F1A) were evaluated at different conditions including development stage, diet, temperature, and insecticide treat ments. Four common algorithms (the Delta Ct method, geNorm, BestKeeper, and NormFinder) and RefFinder were used to analyze gene expression stability. Our results indicated that two reference genes used for normalization were sufficient, and the optimal combinations were: RPS23 and EF2 for developmental stages, ZFP268 and EF2 for feeding with different diets, ZFP268 and RPL13 for temperature treatments, and EF2 and RPL44 for insec ticide treatments. The results provide preliminary determination of suitable reference gene for standard RT-qPCR analyses in T. chilonis, which might establish the foundation for further molecular biology research.

NH₂-Fe-MILs for effective adsorption and Fenton-like degradation of imidacloprid: Removal performance and mechanism investigation

        Mao-Long Chen,Tian-Hui Lu,Ling-Li Long,Zhou Xu,Li Ding,Yun-Hui Cheng 대한환경공학회 2022 Environmental Engineering Research Vol.27 No.2

This investigation enables amino-functionalized metal–organic frameworks (MOFs) materials for the removal of imidacloprid (IMC). Two Fe-based MOF materials of NH₂-MIL-88B(Fe) and NH₂-MIL-101(Fe) both exhibited high adsorption capacity and Fenton-like degradation ability for IMC which were utilized to remove IMC from aqueous solution. Although the adsorption capacity of NH₂-MIL-101(Fe) was higher than that of NH₂-MIL-88(Fe), the degradation abilities of both MOF materials were similar. The removal efficiencies were evaluated through several basic studies, including concentrations of catalyst (0.12-0.3 g/L) and IMC (20-100 mg/L), pH of solution (3-11), and amounts of 30% H₂O₂ (0-2.0 μL/mL). By optimizing the above factors, the total removal ratio of IMC by NH₂-MIL-88B(Fe) was as high as 93%, whereas the removal ratio of NH₂-MIL-101(Fe) was 97%. Moreover, these MOF materials were proven to be stable and recyclable. The free radical quenching experiment and density functional theory calculation were applied to research the removal mechanism, and the hydroxyl radicals (·OH) was found to be the key active intermediate. The high catalytic efficiency can be attributed to the synergy of the Fe3+/Fe2+ redox cycle.

Restoring force model for circular RC columns strengthened by pre-stressed CFRP strips

        Changdong Zhou,Xilin Lu,Hui Li,Teng Tian 국제구조공학회 2014 Steel and Composite Structures, An International J Vol.17 No.4

This paper presents a tri-linear restoring force model based on the test results of 12 circular RC columns strengthened by CFRP strips under low cyclic loading. The pre-stress of CFRP strips and axial load ratio of specimens are considered as the affect parameters of the proposed model. All essential characteristics of the hysteretic behavior of the proposed model, including the hysteretic rules, main performance points, strength degradation, stiffness degradation and confinement effects are explicitly analyzed. The calculated results from the proposed model are in good agreement with the experimental results, which shows that the recommended model can be reliably used for seismic behavior predictions of circular RC columns strengthened by pre-stressed CFRP strips.

Identification and characterization of CONSTANS-like genes from Curcuma alismatifolia

        Yuan-Yuan Li,Xiao-Huang Chen,Hui-Wen Yu,Qi-Lin Tian,Luan-Mei Lu 한국원예학회 2021 Horticulture, Environment, and Biotechnology Vol.62 No.2

The CONSTANS ( CO ) and CONSTANS - like ( COL ) genes have an important role in the regulation of fl owering in photoperiod-sensitive plants. However, information on the molecular characterization of COL genes in Curcuma alismatifoliais very limited. The objective of this study was to identify and evaluate 8 COL homologs ( CaCOLs ) in the C. alismatifoliatranscriptome. Their structures, phylogenetic relationships and expressing patterns under short-day (SD) and long-day (LD)contexts were investigated. Our results showed that CaCOLs were classifi ed into three groups: CaCOL3 and CaCOL6 in groupI were the most CO - like genes; CaCOL7 and CaCOL8 in group II contains only one B-box; CaCOL1 to 2 and CaCOL4 to 5formed the group III. Phylogenetic analysis of the CaCOLs in Arabidopsis, Rice, Barley, Malaccensis and C. alismatifoliademonstrated that both the B-box and CCT domains were conserved in dicot and monocot plants, but diff erent groups ofCOL genes evolved independently. Diff erent patterns of mRNA accumulation in C. alismatifolia leaves in response to SDand LD treatments were observed. CaCOL1 , CaCOL2 , CaCOL4 and CaCOL7 showed signifi cantly higher expressions inLD treatment than that of SD, indicating that they were potential fl owering inducers, and are candidate genes for research infl owering regulation and circadian controlling in C. alismatifolia plants. Altogether, our study facilitates successful regulationof fl owering in Curcuma species and provides insights for future molecular breeding programs.
