One of the major factors affecting the life span of a Reactor Pressure Vessel (RPV) is the Pressurised Thermal Shock (PTS). PTS is a thermo-mechanical load on the RPV wall due to steep temperature gradients and structural load created by internal pressure of the fluid within the RPV. Safe operating life of a nuclear power plant is ensured by carrying out fracture analysis of the RPV against thermal shock. Carrying out fracture tests on RPV/large scale components is not always feasible. Hence, studies on laboratory level specimens are necessary to validate and supplement the prototype results. This paper aims to study the fracture behaviour of standard Compact Tension [C(T)] specimens, made of RPV steel 20MnMoNi55, subjected to thermal shock through experimental and numerical investigations. Fracture tests have been carried out on the C(T) specimens subjected to thermal transient load and tensile load to quantify the effect of thermal shock. Crack resistance curves are obtained from the fracture tests as per ASTM E1820 and compared with those obtained numerically using XFEM and a good agreement was found. A quantitative study on the crack tip plastic zone, computed using cohesive segment approach, from the numerical analyses justified the experimental crack initiation toughness.
Study on acquisition of bacterial antibiotic resistance determinants in poultry litter

<P>Antibiotic resistance and the mode of transmission were investigated in bacteria isolated from poultry litter. Total aerobic heterotrophic bacteria were screened and identified for their resistance to different antibiotics such as ampicillin, streptomycin, erythromycin, tetracycline, chloramphenicol, kanamycin, tobramycin, and rifampicin. The distribution of bacteria found in the litter was Staphylococcus (29.1%), which was the predominant group, followed by Streptococcus (25%), Micrococcus (20.8%), Escherichia coli (12.5%), Salmonella (8.3%), and Aeromonas (4.1%). Fifty percent of these isolates were susceptible to ampicillin, 57% to erythromycin, 25% to tetracycline, 4% to chloramphenicol, 40% to kanamycin, 75% to streptomycin, 54% to tobramycin, and 4% to rifampicin. Three randomly selected isolates representing Staphylococcus, Streptococcus, and Micrococcus were examined for plasmids, and plasmid-curing and plasmid-induced transformation studies were conducted. Streptococcus and Micrococcus harbored a plasmid of 4.2 and 5.1 kb, respectively, whereas Staphylococcus did not harbor any plasmids. Plasmids were cured in Streptococcus and Micrococcus at a concentration of 75 and 100 microg/ mL of acridine orange, respectively, and transformation of 4.2- and 5.1-kb plasmids isolated from the Streptococcus and Micrococcus to plasmid-free E. coli DH5alpha strain was possible. In conjugation experiments, the antibiotic resistance profiles of transconjugant cells were found to be the same as the donors with the exception of Staphylococcus. The results of this study suggest that transformation and conjugation could be an important mechanism for horizontal gene transfer between bacteria in poultry litter. An understanding of the mechanism and magnitude of resistance gene transfer may provide a strategy to reduce the potential for dissemination of these genes.</P>
Detection of High-Risk Human Papillomaviruses in the Prevention of Cervical Cancer in India

Human papillomaviruses (HPVs) are small, non-enveloped, double-stranded DNA viruses that infect epithelial tissues. Specific genotypes of human papillomavirus are the single most common etiological agents of cervical intraepithelial lesions and cervical cancer. Cervical cancer usually arises at squamous metaplastic epithelium of transformation zone (TZ) of the cervix featuring infection with one or more oncogenic or high-risk HPV (HR-HPV) types. A hospital-based study in a rural set up was carried out to understand the association of HR-HPV with squamous intraepithelial lesions (SILs) and cervical cancer. In the present study, HR-HPV was detected in 65.7% of low-grade squamous intraepithelial lesions (LSILs), 84.6% of high-grade squamous intraepithelial lesions (HSILs) and 94% of cervical cancer as compared to 10.7% of controls. The association of HPV infection with SIL and cervical cancer was analyzed with Chi square test (p<0.001). The significant association found confirmed that detection of HR-HPV is a suitable candidate for early identification of cervical precancerous lesions and in the prevention of cervical cancer in India.
