        Inductive expression patterns of genes related to Toll signaling pathway in silkworm (Bombyx mori) upon Beauveria bassiana infection

        Innate immunity is critical to insects and plays an important role in pathogen elimination and wound healing. Toll signaling pathway is the major signaling pathway associated with insect innate immunity mediating synthesis of anti-fungal/bacterial peptides. To better understand the Toll signaling pathwaymediated immune response in Bombyx mori against Beauveria bassiana infection, expression patterns of genes encoding sixteen putative components of Toll signaling pathway in the silkworm larvae challenged with B. bassiana, including four pattern recognition receptors (PRRs, i.e. BmβGRP 1, 2, 3, 4), eight Toll-like receptors (TLRs, i.e. Bm18w, BmToll 1, 3, 6, 9, 7, 10, 11) and four effectors (BmMoricin 1, BmGloverin 2, BmDefensin 1 and BmLysozyme 1), were analyzed using quantitative real-time PCR. At the same time, the changes in their expression by RNAi knock-down of the four PRRswere also detected.Moreover, the effects of Toll signaling pathway inhibitors on antifungal activity in larvae hemolymphwere also analyzed. The results showed that the expression levels of genes encoding sixteen putative components of Toll signaling pathway were obviously altered by the challengewith B. bassiana, but their temporal regulation mode was significantly different. Based on the expression patterns of the genes related to Toll signaling pathway, two sub-paths of immune signal recognition and transduction might be proposed in the response of silkworm larvae against B. bassiana infection. Besides, Toll signaling pathway inhibitor could significantly inhibit the antifungal activity in hemolymph and resulted in increased sensitivity of silkworm larvae to the B. bassiana infection, while the treatment with heat-inactivated B. bassiana could induce antifungal activity in the hemolymph and led to stronger resistance of the silkworm. These results implied that Toll signaling pathway played important roles in the antifungal immune response of the silkworm larvae, in which different components of Toll signaling pathway might play a specific regulatory function. These findings yield insights into the innate immune mechanisms underlying Toll signaling pathway in silkworm.

        Radiological gastrostomy: A comparative analysis of different image-guided methods

        Background: Radiographic guided percutaneous gastrostomy has become a safe and effective enteral nutrition method for patients who can not eat by mouth. Fluoroscopy, computed tomography (CT) and cone-beam CT have been routinely used clinically. The aim of this study was to compare the advantages and disadvantages of percutaneous gastrostomy using different radiographic guided methods. Methods: We retrospectively analyzed the clinical data of 538 patients undergoing percutaneous gastrostomy in our department. According to the image guidance method used in gastrostomy, the patients were divided into groups A by fluoroscopy guidance, group B by fluoroscopy combined with C-arm CT guidance, and group C with the whole process CT guidance. The gastrostomy success rate, complication rate, procedure time, and patient radiation dose were analyzed in the three groups. Results: Among 538 patients, 534 were successful and the success rates are 94.3%, 99.3%, and 100% in group A, B, and C, respectively (P > 0.05). There were 3 cases occurred postoperative bleeding as serious adverse events and transferred to surgical gastrostomy. The minor complications include local infection, hyperplasia of granulation tissue, tube obstruction or prolapse, and local pain of the ostomy. The minor complication rates were 10.5%, 10.4%, and 7.7% in group A, B, and C, respectively (P > 0.05). The average procedure time was 25.57 ± 5.99 minutes, 29.01 ± 6.63 minutes, and 45.47 ± 8.98 minutes, respectively (χ2 = 87.98, P < 0.001). The average radiation dosage was 27.30 ± 19.27 mGy, 145.07 ± 106.08 mGy, and 2,590.26 ± 1,088.22 mGy, respectively (χ2 = 204.44, P < 0.001). Conclusion: There were no significant differences in the success rates and complication rates of gastrostomy under the three guiding methods. For difficult cases, CT-guided gastrostomy may be a very useful supplemental method.

      • Cloud Data Migration Method Based on ABC Algorithm

        Cloud storage systems has played an important role in the support of large-scale and high-performance cloud application. For the purposes of the cloud storage system, data migration is the key technology to the elastic load balancing.In this paper, we take data migration issues into the load-balancing scenarios and propose the data migration method based on the ABC algorithm. At the same time, we validated the method.In the method,we compute the load of each storage node through the comprehensive evaluation system.The system contains the following four aspects, including the available CPU,available memory, data access heat and the system response time.The cloud storage system performs the data migration operation according to the data obtained from the comprehensive evaluation system.The results show that this method can satisfy the application needs.At the same time, this method can reflect the integrated load of each node, and it also can achieve the optimal performance of the cloud storage system.

        Inhibition of miR-128 Abates Aβ-Mediated Cytotoxicity by Targeting PPAR-γ via NF-κB Inactivation in Primary Mouse Cortical Neurons and Neuro2a Cells

        Purpose: Alzheimer’s disease (AD) is the sixth most common cause of death in the United States. MicroRNAs have been identifiedas vital players in neurodegenerative diseases, including AD. microRNA-128 (miR-128) has been shown to be dysregulated inAD. This study aimed to explore the roles and molecular mechanisms of miR-128 in AD progression. Materials and Methods: Expression patterns of miR-128 and peroxisome proliferator-activated receptor gamma (PPAR-γ) messengerRNA in clinical samples and cells were measured using RT-qPCR assay. PPAR-γ protein levels were determined by Westernblot assay. Cell viability was determined by MTT assay. Cell apoptotic rate was detected by flow cytometry via double-stainingof Annexin V-FITC/PI. Caspase 3 and NF-κB activity was determined by a Caspase 3 Activity Assay Kit or NF-κB p65 TranscriptionFactor Assay Kit, respectively. Bioinformatics prediction and luciferase reporter assay were used to investigate interactions betweenmiR-128 and PPAR-γ 3'UTR. Results: MiR-128 expression was upregulated and PPAR-γ expression was downregulated in plasma from AD patients andamyloid-β (Aβ)-treated primary mouse cortical neurons (MCN) and Neuro2a (N2a) cells. Inhibition of miR-128 decreased Aβ-mediated cytotoxicity through inactivation of NF-κB in MCN and N2a cells. Moreover, PPAR-γ was a target of miR-128. PPAR-γupregulation attenuated Aβ-mediated cytotoxicity by inactivating NF-κB in MCN and N2a cells. Furthermore, PPAR-γ downregulationwas able to abolish the effect of anti-miR-128 on cytotoxicity and NF-κB activity in MCN and N2a cells. Conclusion: MiR-128 inhibitor decreased Aβ-mediated cytotoxicity by upregulating PPAR-γ via inactivation of NF-κB in MCNand N2a cells, providing a new potential target in AD treatment.

      • In Vivo Evaluation of Curcumin-loaded Nanoparticles in a A549 Xenograft Mice Model

        Curcumin (Cum) has been reported to have potential chemo-preventive and chemotherapeutic activity through influencing various processes, inducing cell cycle arrest, differentiation and apoptosis in a series of cancers. However, the poor solubility of Cum limits its further applications in the treatment of cancer. We have previously reported Cum-loaded nanoparticles (Cum-NPs) prepared with amphilic methoxy poly(ethylene glycol)-polycaprolactone (mPEG-PCL) block copolymers. The current study demonstrated superior antitumor efficacy of Cum-NPs over free Cum in the treatment of lung cancer. In vivo evaluation further demonstrated superior anticancer effects of Cum-NPs by delaying tumor growth compared to free Cum in an established A549 transplanted mice model. Moreover, Cum-NPs showed little toxicity to normal tissues including bone marrow, liver and kidney at a therapeutic dose. These results suggest that Cum-NPs are effective to inhibit the growth of human lung cancer with little toxicity to normal tissues, and could provide a clinically useful therapeutic regimen. They thus merit more research to evaluate the feasibility of clinical application.

        Isolation and Identification of Paracoccus sp. FD3 and Evaluation of Its Formaldehyde Degradation Kinetics

        A formaldehyde-degrading bacterium strain,FD3, was isolated from contaminated soil and identified as Paracoccus sp. based on partial 16S rRNA gene sequence analysis. In batch culture, the bacterium metabolized 5,000and 8,000 mg/L formaldehyde completely within 16 and 18 h, respectively, at 30oC (pH 7.0) with agitation at 150 rpm. The degradation kinetics was found to follow a first-order model at all initial formaldehyde concentrations with regression values greater than 0.99. Formaldehyde degradation rates increased from 532.37 to 2283.04 mg/L/h as the initial concentration of formaldehyde was increased from 1,000 to 8,000 mg/L. The growth of strain FD3 on formaldehyde as a sole carbon and energy source was well described by the Luong model with a maximal specific growth rate of 0.1754/h, a half-saturation constant of 309.02 mg/L, and a maximum substrate concentration of 3875.53 mg/L. Due to its high tolerance and degradation capacity to formaldehyde, Paracoccus sp., FD3 is considered an excellent candidate for use in degrading formaldehyde in wastewaters.

        Trends in Specific Immunotherapy for Allergic Rhinitis: A Survey of Chinese ENT Specialists

        Purpose: Specific immunotherapy (SIT) is a suitable but uncommon treatment option for allergic rhinitis (AR) in China. The current understandingand attitude of Chinese ENT (ear, nose, and throat) specialists in regards to SIT is unclear. This study investigates current trends in the awarenessand application status of SIT among Chinese ENT specialists. Methods: We performed a nationwide, cross-sectional survey with a specially designedquestionnaire given to 800 ENT specialists in China. A member of the trained research group conducted face-to-face interviews with eachrespondent. Results: Most of the respondents considered AR (96.0%) and allergic asthma (96.0%) the most suitable indications for SIT. Of all respondents,77.0% recommended the application of SIT as early as possible; in addition, SIT was considered ‘relatively controllable and safe’ bymost respondents (80.6%). The highest allergen-positive rate in AR was associated with house dust mite (47.7%) and obvious differences existedamong geographical regions. Conventional subcutaneous immunotherapy was the most highly recommended treatment option (96.2%). ‘The highcost of SIT’ (86.6%) and ‘lack of patient knowledge of SIT’ (85.2%) were probably the main reasons for the lower clinical use of SIT in China. Conclusions:Most cases showed that the opinions of Chinese ENT specialists appeared to be in agreement with recent SIT progress and internationalguidelines; however, many areas still need to enhance the standardization and use of SIT in China. Clinical guidelines for SIT require improvement;in addition, Chinese ENT specialists need continuing medical education on SIT.
