<P><I>Purpose:</I> The anti-cancer effect of &bgr;棺-lapachone (&bgr;棺-lap) is positively related to the cellular activity of NAD(P)H:quinone oxidoreductase (NQO1). Heat shock has been reported to elevate cellular NQO1. The effect of heating on the NQO1 expression in human osteosarcoma cells (HOS) and the response of the cells to the combined treatment with &bgr;棺-lap and hyperthermia was investigated.</P><P><I>Materials and methods:</I> The effects of &bgr;棺-lap alone, hyperthermia alone and in combination to cause clonogenic death and apoptosis in HOS cells were elucidated. The effect of heating on the NQO1expression was evaluated with western blot analysis. The effect of &bgr;棺-lap on the cell cycle distribution was elucidated with flow cytometry and to cause DNA damage was determined by assessing the <I>&ggr;款</I>H2AX foci formation.</P><P><I>Results:</I> Treatment of HOS cells with &bgr;棺-lap at 42°째C was markedly more effective than that at 37°째C in causing clonogenic cell death. Heating caused a long-lasting up-regulation of NQO1 in the cells, and sensitised the cells to &bgr;棺-lap. The <I>&ggr;款</I>H2AX foci formation was increased immediately after &bgr;棺-lap treatment and preheating increased the &bgr;棺-lap-induced <I>&ggr;款</I>H2AX foci formation.</P><P><I>Conclusions:</I> The sensitivity of HOS cells to &bgr;棺-lap was increased not only during heating but also after heating as demonstrated by the increase in the clonogenic cell death and <I>&ggr;款</I>H2AX foci formation. The increase in &bgr;棺-lap sensitivity after heating appeared to be due to the heat-induced elevation of NQO1 activity.</P>

Purpose: As the concept of late-onset hypogonadism (LOH) has gained increased attention, the treatment of eugonadal patients with LOH symptom has become a clinical problem. Previous studies have shown the possible benefits of 5-aminolevulinic acid (5-ALA) on the somatic, psychological and sexual functions. We therefore conducted this randomized, double‑blind, placebo‑controlled study to confirm the efficacy and safety of 5-ALA for LOH symptoms. Materials and Methods: Thirty-two eugonadal subjects with LOH symptoms were randomly divided into a 5-ALA group (n=15) and a placebo group (n=17). Treatment was continued for 8 weeks. The change of the Aging Males’ Symptoms (AMS) scale score and several biochemical and endocrinological variables during treatment were compared between the groups. Results: After treatment, the change in the total AMS in the 5-ALA group was significantly greater than that in the placebo group (-7.4±4.7 vs. -4.9±4.9, p=0.029). However, the differences between the groups in the change of the somatic, psychological, and sexual sub-scores of the AMS did not reach the statistical significance, although these changes in the 5-ALA group were greater than those in the placebo group. Furthermore, the change in the biochemical and endocrinological variables in the two groups did not differ to a statistically significant extent. During the 8-week treatment period, no patients discontinued 5-ALA due to treatment-emergent adverse events (TEAEs). Conclusions: The intake of 5-ALA for 8 weeks was beneficial for eugonadal patients with symptoms of LOH and no severe TEAEs was experienced. 5-ALA should be considered as an option for those patients.

Objective: This study aimed to determine if the discrepancy between depression severity rated by clinicians and that reported by patients depends on key behavioral/psychological features in patients with mood disorders. Methods: Participants included 100 patients with mood disorders. First, we examined correlations and regressions between scores on the Hamilton Depression Rating Scale (HAMD) and Beck Depression Inventory (BDI). Second, we divided the participants into those who provided 1) greater ratings for the BDI compared with the HAMD (BDI relative- overrating, BO) group, 2) comparable ratings for the BDI and HAMD (BDI relatively concordant, BC) group, or 3) less ratings for the BDI (BDI relative-underrating, BU) group. Adverse childhood experiences, autistic-like traits, and coping styles were evaluated with a six-item short version of the Childhood Trauma Questionnaire (CTQ-6), the Social Responsiveness Scale for Adults (SRS-A), and the Ways of Coping Checklist (WCCL), respectively. Results: A significant correlation was found between HAMD and BDI scores. Total and emotional abuse subscale scores from the CTQ-6, and the self-blame subscale scores from the WCCL were significantly higher for the BO group compared with the BU group. The BO group also elicited significantly higher SRS-A total scores than did the other groups. Conclusion: These findings suggest that patients with adverse emotional experiences, autistic-like traits, and self-blame coping styles perceive greater distress than that evaluated objectively by clinicians. The results indicate the need for inclusion of subjective assessments to effectively evaluate depressive symptoms in patients deemed to have these psycho- behavioral concerns.

Purpose: Robot-assisted radical cystectomy (RARC) was originally intended to replace open radical cystectomy (ORC) as a minimally invasive surgery for patients with invasive bladder cancer. The purpose of this study was to evaluate the advantages of robotic surgery, comparing perioperative and oncologic outcomes between RARC and ORC. Materials and Methods: Between June 2012 and August 2016, 49 bladder cancer patients were given a radical cystectomy, 21 robotically and 28 by open procedure. We compared the clinical variables between the RARC and ORC groups. Results: In the RARC group, the median estimated blood loss (EBL) during cystectomy, total EBL, operative time during cystectomy, and total operative time were 0 mL, 457.5 mL, 199 minutes, and 561 minutes, respectively. EBL during cystectomy (p<0.001), total EBL (p<0.001), and operative time during cystectomy (p=0.003) in the RARC group were significantly lower compared with the ORC group. Time to resumption of a regular diet (p<0.001) and length of stay (p=0.017) were also significantly shorter compared with the ORC group. However, total operative time in the RARC group (median, 561 minutes) was significantly longer compared with the ORC group (median, 492.5 minutes; p=0.015). Conclusions: This Japanese study presented evidence that RARC yields benefits in terms of BL and time to regular diet, while consuming greater total operative time. RARC may be a minimally invasive surgical alternative to ORC with less EBL and shorter length of stay.
Alteration of Starch-Sucrose Transition in Germinating Wheat Seed under Sodium Chloride Salinity

Alterations in starch-sucrose transition during germination were studied in wheat seeds under saline conditions. NaCl significantly reduced the speed of germination and resultant seedling growth, but delayed the degradation of seed storage components. The endogenous level of ABA increased while osmotic potential decreased. NaCl also inhibited the expression of α-amylase. Increasing the concentration of NaCl induced the expression of sucrose phosphate synthase, and sugars, including sucrose, were accumulated in the seedlings. This accumulation of sugar closely correlated with an increase in ABA. However, sugar accumulation was reversible when the salt stress was removed. Overall our results strongly suggest that the germinating wheat seeds alter the starch-to-sucrose conversion to adapt for salt stress. This is probably mediated by the increase in ABA.
