IAN5 polymorphisms are associated with systemic lupus erythematosus

<P>Systemic lupus erythematosus (SLE) is a representative autoimmune disease, which is frequently associated with lymphopenia. Biobreeding (BB) rat is a typical animal model which develops autoimmune diseases with lymphopenia which results from a frame-shift mutation in the immune-associated nucleotide (<I>IAN</I>) <I>5</I> gene. <I>IAN5</I> is involved in the regulation of T-cell activation and survival. To examine the association of <I>IAN5</I> gene with SLE, we scrutinised the single nucleotide polymorphisms (SNPs) in the <I>IAN5</I> gene. We conducted a case–control study where 132 SLE patients, 505 rheumatoid arthritis (RA) patients, and 546 controls were genotyped for four SNPs in the <I>IAN5</I> gene. Two SNPs (+2071C?>?T and +2677G?>?A) were associated with susceptibility to SLE (<I>P</I>?=?0.040 and 0.045, respectively), and −4432G?>?A SNP was associated with the development of leukopenia (<I>P</I>?=?0.028) and the requirement of steroid pulse therapy (<I>P</I>?=?0.040) in SLE patients. Haplotype analyses showed that Ht1(CTCG) was associated with susceptibility to SLE (<I>P</I>?=?0.036), and Ht4(ACCG), Ht5(ACTA) and Ht6(GCCG) were associated with the development of nephritis (<I>P</I>?=?0.017, 0.019, 0.022, respectively). In conclusion, the <I>IAN5</I> polymorphisms were associated with susceptibility to SLE and the development of clinical disease manifestations in a strictly Korean population.</P>

A comprehensive knowledge on the relationship between soil salinity and arbuscular mycorrhizal fungi (AMF) is vital for a deeper understanding of ecosystem functioning under salt stress conditions. The objective of this study was to determine the effects of soil salinity on AMF root colonization, spore count, glomalin related soil protein (GRSP) and community structure in Saemangeum reclaimed land, South Korea. Soil samples were collected and grouped into five distinct salt classes based on the electrical conductivity of soil saturation extracts (ECse). Mycorrhizal root colonization, spore count and GASP were measured under different salinity levels. AMF community structure was studied through three complementary methods; spore morphology, terminal restriction fragment length polymorphism (T-RFLP) and denaturing gradient gel electrophoresis (DGGE). Results revealed that root colonization (P < 0.01), spore count (P < 0.01) and GRSP (P < 0.01) were affected negatively by soil salinity. Spore morphology and T-RFLP data showed predominance of AMF genus Glomus in Saemangeum reclaimed land. T-RFLP and DGGE analysis revealed significant changes in diversity indices between non (ECse < 2 dS/m) and extremely (ECse > 16 dS/m) saline soil and confirmed dominance of Glomus caledonium only in soils with ECse < 8 dS/m. However, ribotypes of Glomus mosseae and Glomus proliferum were ubiquitous in all salt classes. Combining spore morphology, T-RFLP and DGGE analysis, we could show a pronounced effect in AMF community across salt classes. The result of this study improve our understanding on AMF activity and dominant species present in different salt classes and will substantially expand our knowledge on AMF diversity in reclaimed lands. Manufacturing progress on the first sector and lower ports for ITER vacuum vessel

Manufacturing design of Korean sectors and ports for the ITER Vacuum Vessel (VV) has been developed to comply with the tight tolerance and severe inspection requirements. The first VV sector and lower ports are being fabricated slowly under strict regulations after verification using several real scale mock-ups and qualifications for welding, forming and NDE. During three years after start of fabrication, manufacturing progress on four poloidal segments of the first sector is that (1) all inner shells were welded, (2) forgings for complicate components have been machined, (3) port stubs and poloidal T-ribs were assembled, and (4) machined components are welded on the inner shells by narrow-gap TIG welding and electron beam welding. The progress of lower ports is that (1) inner shells of stub extensions were bent and treated with heat, (2) T-ribs were fabricated and examined by qualified phased array UT, (3) supporting pads and gussets have been machined, and (4) inner shells are assembled with T-ribs and machined forgings. The progress rate of manufacturing is around 40% up to the end of 2015 for the first sector and lower port stub extensions.

A computer code of finite element method is developed by using the ?? programming language. The concept of class, inheritance, and polymorphism in ?? improves the modularity and the flexibility(such as potability, implantability, and expandability) of engineering software. The graphic user interface (GUI) is designed by using the MFC(Microsoft Foundation Class) based on ??, and combined successfully with the main code of FEM solver. This study shows the superior of OOP(Object-Oriented programming) such as ?? to other progamming languages when applied to complicated engineering problems.

Background/Aims: No clear data have been established and validated regarding whether rectal retroflexion has an important and therapeutic impact. The aim of the present study was to evaluate the diagnostic yield and therapeutic impact of rectal retroflexion compared with straight view examination. Methods: A prospective single-blind study was conducted. Consecutive patients evaluated between October 2011 and April 2012 were included. Results: A total of 934 patients (542 women [58%]) were included. The mean age was 57.4±14.8 years. Retroflexion was successful in 917 patients (98.2%). Distinct lesions in the anorectal area were detected in 32 patients (3.4%), of which 10 (1%) were identified only on retroflex view and 22 (2.4%) on both straight and retroflex views. Of the 32 identified lesions, 16 (50%) were polyps, nine (28.1%) were angiodysplasias, six (18.8%) were ulcers, and one (3.1%) was a flat lesion. All 10 patients (1%) in whom lesions were detected only by rectal retroflexion showed a therapeutic impact. Conclusions: Rectal retroflexion has minimal diagnostic yield and therapeutic impact. However, its low rate of major complications and the possibility of detecting lesions undetectable by straight viewing justify its use.

Background: Physiatrists are facing with survivors from disasters in both the acute and chronic phases of muscle and nerve injuries. Similar to many other clinical conditions, neuromusculoskeletal ultrasound can play a key role in the management of such cases (with various muscle/nerve injuries) as well. Accordingly, in this article, a recent single-center experience after the Turkey-Syria earthquake will be rendered. Methods: Ultrasound examinations were performed for various nerve/muscle lesions in 52 earthquake victims referred from different cities. Demographic features, type of injuries, and applied treatment procedures as well as detailed ultrasonographic findings are illustrated. Results: Of the 52 patients, 19 had incomplete peripheral nerve lesions of the brachial plexus (n=4), lumbosacral plexus (n=1), and upper and lower limbs (n=14). Conclusion: The ultrasonographic approach during disaster relief is paramount as regards subacute and chronic phases of rehabilitation. Considering technological advances (e.g., portable machines), the use of on-site ultrasound examination in the (very) early phases of disaster response also needs to be on the agenda of medical personnel.

The CYP2B6 enzyme is involved in the metabolism of environmental toxins and endogenous compounds. The expression and activity of the cyp2b6 gene is important for the brain’s ability to detoxify harmful compounds. In rats, the Cyp2b1 gene is homologous to the human Cyp2b6 gene and has an isoform, Cyp2b2, with nearly the same sequence. Although the distinction between them is fundamental, most studies report global changes in CYP2B enzymes due to the complexity of the techniques to detect both genes separately. We instrumented a semi-quantitative technique combining multiplex end point RT-PCR with restriction enzyme assays, allowing us to establish a relatively simple, reproducible and affordable method to distinguish Cyp2b1 gene expression from Cyp2b2. Using this technique, we showed differential Cyp2b1 expression after phenobarbital and ethanol induction in the brain and liver and detected increased Cyp2b1 expression after ethanol treatment in the brain striatum that had not been detected before.
