The coronavirus porcine epidemic diarrhea virus (PEDV) infects the cells lining the small intestine of a pig and, causes porcine epidemic diarrhea (PED). Owing to its highly infectious nature, PEDV has a substantial economic burden, which results in significant morbidity and mortality in piglets. In this study, the virucidal efficacy of a powder disinfectant containing a phosphate compound against PEDV was investigated. Virucidal efficacy was assessed as the infectivity of PEDV toward Vero cells after exposure of the virus to the disinfectant. PEDV was exposed to the disinfectant in the presence of either hard water (HW) or an organic matter suspension (OM). In the HW condition, PEDV was inactivated by 4-fold dilution of the disinfectant. In the presence of OM, the disinfectant showed virucidal activity with a 2-fold dilution. As the disinfectant possessed virucidal activity against PEDV, it should be an effective reagent for limiting the spread of animal viral diseases.

Statement of problem. The role of calcium sulfate in stimulating the growth of gingival soft tissue has been reported in few studies. Such a unique property of calcium sulfate could serve as a trouble-solving broker in compensating for the lack of soft tissues in various oral surgeries. Purpose. The purpose of this study was to compare the proliferating activities of human gingival fibroblasts seeded on various bone graft barrier materials of calcium sulfate, collagen, and polytetrafluorethylene (PTFE). Material and methods. Two calcium sulfates ($CAPSET^{(R)}$. and $CalForma^{(R)}$, Lifecore Biomedical Inc., St. Paul, Minnesota, USA), a resorbable natural collagen ($Bio-Gide^{(R)}$, Geistlich Pharma Ag., Wolhusen, Switzerland), and a non-resorbable PTFE ($TefGen-FD^{(R)}$, Lifecore Biomedical Inc., St. Paul, Minnesota, USA) served as the human gingival fibroblasts' substrates and comprised the four experimental groups, whereas the untreated floors of culture plastics were used in the control group, in this study. Cells were trypsinized, seeded, and incubated for 48 h. The proliferating activities of fibroblasts were determined by XTT and SRB assay and absorbance (optical density, OD) was measured. One-way ANOVA was used to analyze the differences in the mean OD values between the groups of CAPSET, CalForma, Bio-Gide, TefGen, and the control (p<0.05). Results. From the XTT assay, the mean OD value of the control group, the highest, was significantly greater than that of any of the four experimental groups followed by CalForma, CAPSET, TefGen, and Bio-Gide. Further, the mean OD value of CalForma, was significantly greater compared to that of Bio-Gide. From the SRB assay, Calforma showed the highest mean OD value, which was significantly greater than that of any other groups, followed by the control, CAPSET, Bio-Gide, and TefGen. The mean OD values of both the control and CAPSET were significantly greater compared to that of TefGen (p<0.05). Conclusion. Assessment of the viability and proliferation of cultured fibroblasts seeded and incubated for 48 h on various barrier-material substrates using XTT and SRB assay showed that calcium sulfate $CalForma^{(R)}$ promotes the proliferating activity of human gingival fibroblasts.
Effect of Ion-Pair Formation with Bile Salts on the In Vitro Cellular Transport of Berberine

The objective of this study was to examine the effect of ion-pair complexation with endogenous bile salts on the transport of a quarternary ammonium organic cationic (OC) drug, berberine, across the Caco-2 and LLC-PK1 cell monolayers. The basolateral-to-apical (BL-AP) transport of berberine in Caco-2 cells was temperature dependent and 10-fold higher than that of the apical-to-basolateral (AP-BL) transport. Similar results were observed for the transport of berberine across the LLC-PK1 cells. Moreover, the BL-AP transport in the Caco-2 cells was significantly reduced by the cis-presence of P-glycoprotein (P-gp) inhibitors such as cyclosporine A, verapamil, and digoxin. These results suggest that an efflux transporter, probably P-gp, is involved in the Caco-2 cell transport. The $K_m$ and $V_{max}$ values for the carrier-mediated transport were estimated to be 83.4 mM and 7640 pmole/h/$cm^2$, respectively. The apparent partition coefficient (APC) of berberine between n-octanol and a phosphate buffer (pH 7.4) was increased by the presence of an organic anion (OA), taurodeoxycholate (TDC, a bile salt), suggesting the formation of a lipophilic ion-pair complex between an OC (berberine) and an OA (TDC). Despite the ion-pair complexation, however, the BL-AP transport of berberine across the Caco-2 and LLC-PK1 cells was not altered by the cis-presence of bile salts or the rat bile juice. This is consistent with the reportedly unaltered secretory transport of a quarternary ammonium compound, tributylmethylammonium (TBuMA), across the Caco-2 cell monolayers in the cis-presence of bile salts or the rat bile juice, but not with our previous report in which the secretory transport of TBuMA across the LLC-PK1 cell was increased in the cis-presence of TDC. Therefore, the effect of ion-pair formation with the bile components or bile salts on the secretory transport of OCs appears to depend on the molecular properties of OCs (e.g., molecular weight, lipophilicity and affinity to relevant transporters) and the characteristics of cell strains (e.g., expression and contribution of responsible transporters to the transport).

Highly pathogenic avian influenza virus (HPAIV) damages vital organs and tissues, frequently leading to death in birds, and causes serious economic losses in the poultry industry. In addition, HPAIV can infect humans and other mammals, often with fatal outcomes. In this study, the virucidal efficacy of Clean-Zone??, which contains citric acid, malic acid and phosphoric acid, against avian influenza virus (AIV, H9N2) was investigated. Virucidal efficacy was determined by examining the viability of AIV after contact with the disinfectant in the allantoic membrane of chicken embryos. The disinfectant and AIV were reacted under hard water (HW) and organic matter suspension (OM) condition. AIV was inactivated with 200- and 50-fold dilutions of the disinfectant under HW and OM conditions, respectively. As the disinfectant, Clean-Zone??, has a virucidal efficacy against AIV, it can be used to prevent the spread of animal viral diseases.

This study was investigated the synergistic antibacterial effects from the combination of sulfamethazine (SMZ) and tiamulin (SMT) against Salmonella Typhimurium (S. Typhimurium) and Actinobacillus pleuropneumoniae (A. pleuropneumoniae) isolated from pigs. Minimum inhibitory concentrations (MICs) and fractional inhibitory concentration (FIC) index were determined for 10 bacteria from 13 swine farms in Gyeongsangnam province by the National Committee for Clinical Laboratory Standards broth microdilution procedure. Antimicrobial agents tested included SMZ, tiamulin and SMT. Against S. Typhimurium and A. pleuropneumoniae, the MIC ranges of SMZ, tiamulin and SMT were 125-500, 15.625-62.5 and 15.625-62.5 μg/mL, respectively. In addition, the FIC index ranges of SMT against S. Typhimurium and A. pleuropneumoniae were 0.19-0.38 and 0.19-0.62, respectively. In conclusion, SMT has a high antibacterial activity against S. Typhimurium and A. pleuropneumoniae, then the combination may be applied for the treatment of swine diseases caused by S. Typhimurium and A. pleuropneumoniae.

This study examined the acute toxicity of Super-Neophensan containing florfenicol and acetaminophen in male and female Sprague-Dawley rats. Rats were orally administered the drug at dose levels of 0, 250, 500, 1,000 and 2,000 mg/kg body weight (BW) for the single-dose toxicity test. BW changes in all treated groups were not significantly different to those in the control treated with saline. In hematological and blood biochemical analyses, all parameters were unaffected by the drug. In the same way, there were no significant differences between all treated groups and the control group in terms of markers for liver and kidney functions. As no adverse effects were observed with the drug in the single-dose oral toxicity test, even at the highest concentration of the drug, the lethal dose 50 (LD50) of Super-Neophensan is estimated to be greater than 2,000 mg/kg BW.

<P><B>Background/Aims</B></P><P>The purpose of this study was to investigate the malignant potential of aberrant crypt foci (ACF) by measuring the multiplicity of crypts and lectin expression in the early and late stages of 1,2-dimethylhydrazine (DMH)-induced colon carcinogenesis.</P><P><B>Methods</B></P><P>Six-week-old Wistar rats were injected subcutaneously with DMH for 27 weeks. We classified ACF according to the number of crypts per ACF as a few crypts (≤3 crypts, FC ACF) or numerous crypts (≥4 crypts, NC ACF). Immunohistochemistry was used to evaluate lectin expression.</P><P><B>Results</B></P><P>In the early stage, FC ACF (590/1,902, 31.0%) occurred more frequently than NC ACF (35/449, 7.8%); whereas in the late stage, NC ACF (176/449, 39.2%) occurred more frequently than FC ACF (324/1,902, 17.0%). The number of ACF peaked at 15 to 20 weeks. The ratio of NC/FC ACF increased gradually during carcinogenesis. The expression of both UEA1 and PNA was higher in NC ACF than FC ACF. Lectin expression increased in the late stage compared with the early stage.</P><P><B>Conclusions</B></P><P>The expression of lectin was higher in NC ACF and ACF in the late stage. Therefore, ACF with higher multiplicities in the late stage may have more malignant potential in DMH-induced colon carcinogenesis.</P>
