        The Usefulness of Preoperative Colonoscopic Tattooing with Autologous Blood for Localization in Laparoscopic Colorectal Surgery

        Purpose: In colorectal cancer surgery, it is important to have accurate resection margins. But, there is difficulty to find lesions in laparoscopic surgery. To reduce surgical errors, many preoperative localizing methods have been introduced. In this study, we aimed to assess the preoperative feasibility and safety of autologous blood tattooing. Methods: From August 2017 to February 2020, a total of 11 patients underwent preoperative colonoscopic autologous blood tattooing. At the start of surgery, the surgeon assessed for the presence of visibility and other complications such as abscess or spillage. The characteristics of patients, outcomes, and complications were collected retrospectively. Results: The study comprised 8 men and 3 women, with an average age of 63 years. Except for one imprecise patient during the surgery, all other 10 patients showed precise visibility, and no localization errors were observed. No complication was observed in all patients. Conclusion: Preoperative autologous blood tattooing is a very useful and safe technique because it has high visibility and no complications. This method does not require additional agents or facilities. Large scale study will be required for standard guidelines.

        Transplantation of betacellulin-transduced islets improves glucose intolerance in diabetic mice

        <P>Type 1 diabetes is an autoimmune disease caused by permanent destruction of insulin-producing pancreatic β cells and requires lifelong exogenous insulin therapy. Recently, islet transplantation has been developed, and although there have been significant advances, this approach is not widely used clinically due to the poor survival rate of the engrafted islets. We hypothesized that improving survival of engrafted islets through <I>ex vivo</I> genetic engineering could be a novel strategy for successful islet transplantation. We transduced islets with adenoviruses expressing betacellulin, an epidermal growth factor receptor ligand, which promotes β-cell growth and differentiation, and transplanted these islets under the renal capsule of streptozotocin-induced diabetic mice. Transplantation with betacellulin-transduced islets resulted in prolonged normoglycemia and improved glucose tolerance compared with those of control virus-transduced islets. In addition, increased microvascular density was evident in the implanted islets, concomitant with increased endothelial von Willebrand factor immunoreactivity. Finally, cultured islets transduced with betacellulin displayed increased proliferation, reduced apoptosis and enhanced glucose-stimulated insulin secretion in the presence of cytokines. These experiments suggest that transplantation with betacellulin-transduced islets extends islet survival and preserves functional islet mass, leading to a therapeutic benefit in type 1 diabetes.</P>

        A Prenylated Flavan from <i>Broussonetia kazinoki</i> Prevents Cytokine-Induced β-Cell Death through Suppression of Nuclear Factor-κB Activity

        <P>The generation of nitric oxide (NO) <I>via</I> inducible NO synthase (iNOS) and reactive oxygen species plays a key role in cytokine-mediated pancreatic β-cell damage. Oxidative stress due to reactive oxygen species activates the nuclear factor-κB (NF-κB) transcription factor, which regulates iNOS expression. In this regard, suppression of the NF-κB pathway is a novel strategy for protecting β-cells from damage. This study was performed to explore the effects of kazinol U, a prenylated flavan from <I>Broussonetia kazinoki</I>, on the NF-κB activation pathway in interleukin-1β (IL-1β)- and interferon-γ (IFN-γ)-treated β-cells. The cytotoxic effects of cytokines were completely abolished when RINm5F cells or islets were pretreated with kazinol U. Kazinol U inhibited the nuclear translocation and DNA binding of NF-κB subunits, which correlated with the inhibitory effects on IκB kinase (IKK) phosphorylation and IκBα degradation. In addition, kazinol U suppressed NO and hydrogen peroxide production and apoptotic cell death by cytokines in RINm5F cells. The protective effects of kazinol U were further demonstrated by normal insulin secretion of cytokine-treated islets in response to glucose. Taken together, these results suggest that using kazinol U to block the NF-κB pathway in pancreatic β-cells reduces cell damage. Therefore, kazinol U may have therapeutic value in delaying pancreatic β-cell destruction in type 1 diabetes.</P>

        Three known triterpenoids were isolated from MeOH extract of C. apiifolia (Ranunculaceae). Their structures were identified as oleanolic acid (1), ursolic acid (2), hederagenic acid (3) by comparison of their physicochemical and spectral data with the literature values. Among them, 2 was isolated for the first time from this plant. The isolated compounds were evaluated for their cytotoxicity against L1210, HL-60, SK-OV-3 tumor cell lines. All compounds 1-3 were shown good activities with IC50 values ranging from 7.7 to 25.6 μg/ml. This result suggests that triterpenoids 1-3 are main cytotoxic principles of this plant.

        Transforming Growth Factor β Inhibits MUC5AC Expression by Smad3/HDAC2 Complex Formation and NF-κB Deacetylation at K310 in NCI-H292 Cells

        Airway mucus secretion is an essential innate immune response for host protection. However, overproduction and hypersecretion of mucus, mainly composed of the gel-forming MUC5AC protein, are significant risk factors for patients with asthma and chronic obstructive pulmonary disease (COPD). The transforming growth factor β (TGFβ) signaling pathway negatively regulates MUC5AC expression; however, the underlying molecular mechanism is not fully understood. Here, we showed that TGFβ significantly reduces the expression of MUC5AC mRNA and its protein in NCI-H292 cells, a human mucoepidermoid carcinoma cell line. This reduced MUC5AC expression was restored by a TGFβ receptor inhibitor (SB431542), but not by the inhibition of NF-κB (BAY11-7082 or Triptolide) or PI3K (LY294002) activities. TGFβ-activated Smad3 dose-dependently bound to MUC5AC promoter. Notably, TGFβ-activated Smad3 recruited HDAC2 and facilitated nuclear translocation of HDAC2, thereby inducing the deacetylation of NF-κB at K310, which is essential for a reduction in NF-κB transcriptional activity. Both TGFβ-induced nuclear translocation of Smad3/HDAC2 and deacetylation of NF-κB at K310 were suppressed by a Smad3 inhibitor (SIS3). These results suggest that the TGFβ-activated Smad3/HDAC2 complex is an essential negative regulator for MUC5AC expression and an epigenetic regulator for NF-κB acetylation. Therefore, these results collectively suggest that modulation of the TGFβ1/Smad3/HDAC2/NF-κB pathway axis can be a promising way to improve lung function as a treatment strategy for asthma and COPD.

        엉덩관절 안쪽돌림 조절을 위한 비탄력 테이핑 방법이 엉덩관절 근활성도에 미치는 영향 -예비연구-

        Purpose: This study investigated the effects of the non-elastic taping method for controlling internal hip joint rotation on internal and external hip rotator muscle activity in healthy people. Methods: In this study, 18 healthy volunteers were instructed to perform the small knee bending (SKB) test. All participants completed the test following two methods (using non-elastic taping and not using taping). Muscle activation during the two methods was measured using a surface electromyography (EMG) device. Surface EMG data were collected from the gluteus medius, gluteus maximus, and tensor fasciae femoris muscles while performing the SKB test with and without non-elastic taping. Results: Muscle activity in the gluteus maximus was significantly higher during the SKB test with non-elastic taping than during the conventional SKB test with taping (p < 0.05). Tensor fasciae latae muscle activity was lower during the SKB test with non-elastic taping than during the conventional SKB test (p < 0.05). Conclusion: The findings suggest that the non-elastic taping method for controlling internal hip joint rotation effectively activates the hip's external rotator muscles and minimizes unwanted internal rotator muscle use during the SKB test. Therefore, the non-elastic taping method for controlling internal hip joint rotation could be an effective intervention for those who cannot control the internal rotation of their hips.

        제주지역 차나무(Camellia sinensis L.) 품종 및 차기에 따른 신초 부위별 품질특성 변화

        제주지역 환경조건에서의 품종 및 차기에 따른 신초의부위별 품질 구성 성분의 특성 분석을 통해 품질특성에 미치는 요인을 평가하고자 본 연구를 수행하였다. 조생종 사에미도리, 중생종 야부기다 및 만생종 후슌의 3개 품종에 대해 1번차, 2번차 및 3번차 시기에 심, 상위 1엽부터 5엽과 줄기 등 7부위로 분리하여 각 부위의 품질 특성을 조사하였다. 품질 특성은 총유리아미노산(%), 데아닌(%), 총 카테킨(%), 개별 카테킨(%), 카페인(%), 탄닌(%), 비타민 C(㎎/㎏) 및 섬유소(%) 함량에 대해 근적외선분광광도계를 이용하여 분석하였다. 총 유리아미노산과 데아닌 함량의 부위별 특성은 유사하게 나타났는데, 상위엽에서 하위엽으로 갈수록 감소하는 경향이었고, 줄기 부분의 함량은 하위 4엽과 5엽보다 높게 나타났다. 총 카테킨과 탄닌 함량은 비슷한 경향을 나타내어 하위엽으로 갈수록 낮아졌으나, 그 감소 정도는 크지 않았으며 줄기 부분의 총 카테킨 함량은 총 유리아미노산과는 달리 현저히 떨어지는 경향이었다. 개별 카테킨에 있어서도 EC, ECG와 EGCG 모두 총 카테킨과 유사한 경향을 나타내었다. 카페인 함량은 총 유리아미노산 및 데아닌과 같이 하위엽으로 갈수록 현저하게 낮아지는 경향이었고, 줄기부분에서는 상위 3엽과 비슷한 수준의 함량을 보였다. 비타민C 함량은 잎 위치에 따른 변화가 거의 없었으나, 줄기에서 약간 낮았고, 차기별로는 2-3번차 보다는 1번차에서 높게 나타났다. 조섬유 함량은 품종과 차기간에 관계없이 상위엽에서 하위엽으로 갈수록 증가하는 경향이었고, 줄기부분은 하위 5엽과 비슷한 수준이었다. 본 연구결과 차품종과 차기에 따라 부위별 차 품질에 미치는 영향이 매우 크게 작용할 수 있음을 나타내었다. This study examined the factors affecting the tea quality by analyzing the quality characteristics of different shoot parts in different tea cultivars and tea seasons in Jeju, Korea. New growing shoots were plucked from ‘Saemidori’ (early-maturing), ‘Yabukita’ (mid-maturing), and ‘Hushun’ (late-maturing) at each tea season and separated into seven parts bud, 1<sup>st</sup> leaf, 2<sup>nd</sup> leaf, 3<sup>rd</sup> leaf, 4<sup>th</sup> leaf, 5<sup>th</sup> leaf, and stem to analyze the quality components. The tea quality components including the total free amino acids, theanine, catechins, caffeine, tannin, vitamin C, and fiber were determined by NIR analysis. The changes in the total free amino acids and theanine content in the different shoot parts were similar in the three cultivars and three different tea seasons. These contents decreased gradually in the lower leaves and were slightly higher at the stem than at the 4<sup>th</sup> and 5<sup>th</sup> leaf. The changes in the total cathechins and tannin contents showed a similar tendency decline gradually in leaves and decrease rapidy in the stem. The EC, ECG and EGCG content in the individual cathechins were similar to the total cathechins. The caffeine content in the different shoot parts showed a similar tendency to those of the total the amino acids and theanine at the three cultivars and three tea seasons. Caffein content decreased gradually at the lower leaves and was higher at the stem than at 3<sup>th</sup> and 4<sup>th</sup> leaf. The vitamin C content was relatively constant at different leaf positions, but was slightly low in the stem with a higher tendency at the 1<sup>st</sup> crop than at the 2<sup>nd</sup> and 3<sup>rd</sup> crop in all three cultivars. The crude fiber content was similar at the different cultivars and tea seasons, whereas it increased gradually at the lower shoot parts, and was similar at the stem to that of the 5<sup>th</sup> leaf. These results suggest that the shoot parts at different tea cultivars and tea seasons can be major factors affecting the tea quality.
