Although, one of the etiologies of localized lipodystrophy of the subcutaneous connective tissue (cellulite) is the histological alternation of adipose tissue, the characteristics of expression of the components of extracellular matrix (ECM) components during adipogenesis are not uncovered. In this study, the effects of caffeine and Ishige okamurae originated diphlorethohydroxycarmalol (DPHC) on the expression of extracellualr fibers was analyzed with quantitative RT-PCR during differentiation induction of mouse subcutaneous adipose derived stem cells (msADSC) into adipocyte. The expression levels of Col1a, Col3a1, and Col61a were decreased by the adipogenci induction in a time-dependent manners. However, Col2a mRNA and Col4a1 mRNA expressions were oposit to them. Caffeine and DPHC stimulated the changes of the expression of these collagens. Eln mRNA expression was increased by induction. DPHC stimulated the expression of it. Mfap5 mRNA expression was deceased in both adipogenic cell and matured adipocytes. Caffeine suppressed the expression of Mfap5 but the effect of DPHC was different by the concentration. The expression of bioglycan, decorin, and lumican were also modified by caffeine and DPHC in a concentration-dependent manner. Based on this study, we revealed firstly the effects of caffeine and DPHC on the expression of collagens, elastin, and glycoproteins during adipogenesis of msADSCs. Those results suggest that DPHC may have antiadipogenic effect and has more positive effets on normal adipose tissue generation and work as suppressor the abnormality of ECM structure. Such results indicate that DPHC can be applied in keeping the stability of the ECM of adipogenic tissues.

Windbreak forests, which have a windproof effect against strong winds, are known to be effective in reducing the spread of odors and dust emitted from livestock farms. The effect of reducing the spread of odors and dust can be estimated through numerical models such as computational fluid dynamics, which require aerodynamic coefficients of the windbreaks for accurate prediction of their performance. In this study, we aimed to evaluate the aerodynamic coefficients, Co, C1, C2, and α, of two windbreaks, Thuja occidentalis and a mesh net, through wind tunnel experiments. The aerodynamic coefficients were derived by the relation between the incoming wind speed and the pressure loss due to the windbreaks which was measured by differential pressure sensors. In order to estimate the change in the aerodynamic coefficient concerning various leaf density, the experiments were conducted repeatedly by removing the leaves gradually in various stages. The results showed that the power law regression model more suitable for coefficient evaluation compared to the Darcy-Forchheimer model.
3′‐Sialyllactose protects against osteoarthritic development by facilitating cartilage homeostasis

Abstract

3′‐Sialyllactose has specific physiological functions in a variety of tissues; however, its effects on osteoarthritic development remain unknown. Here, we demonstrated the function of 3′‐sialyllactose on osteoarthritic cartilage destruction. In vitro and ex vivo, biochemical and histological analysis demonstrated that 3′‐sialyllactose was sufficient to restore the synthesis of Col2a1 and accumulation of sulphated proteoglycan, a critical factor for cartilage regeneration in osteoarthritic development, and blocked the expression of Mmp3, Mmp13 and Cox2 induced by IL‐1β, IL‐6, IL‐17 and TNF‐α, which mediates cartilage degradation. Further, reporter gene assays revealed that the activity of Sox9 as a transcription factor for Col2a1 expression was accelerated by 3′‐sialyllactose, whereas the direct binding of NF‐κB to the Mmp3, Mmp13 and Cox2 promoters was reduced by 3′‐sialyllactose in IL‐1β‐treated chondrocytes. Additionally, IL‐1β induction of Erk phosphorylation and IκB degradation, representing a critical signal pathway for osteoarthritic development, was totally blocked by 3′‐sialyllactose in a dose‐dependent manner. In vivo, 3′‐sialyllactose protected against osteoarthritic cartilage destruction in an osteoarthritis mouse model induced by destabilization of the medial meniscus, as demonstrated by histopathological analysis. Our results strongly suggest that 3′‐sialyllactose may ameliorate osteoarthritic cartilage destruction by cartilage regeneration via promoting Col2a1 production and may inhibit cartilage degradation and inflammation by suppressing Mmp3, Mmp13 and Cox2 expression. The effects of 3′‐sialyllactose could be attributed in part to its regulation of Sox9 or NF‐κB and inhibition of Erk phosphorylation and IκB degradation. Taken together, these effects indicate that 3′‐sialyllactose merits consideration as a natural therapeutic agent for protecting against osteoarthritis.

Jinan Poonghyeol in Chonbuk province is famous for emitting cool air in summer and warm air in winter. Cause for these phenomena is studied by measuring temperature, humidity and air velocity of several locations around Jinan Poonghyeol. Temperature variations under the ground are also measured. Analyzing data compiled from July 21, 2005 through May 26, 2006, it is presumed that storage effect of rocks and soil in this area and buoyance effect are the main causes for this mysterious natural air-conditioning phenomena.

Recently, acupuncture has demonstrated extraordinary clinical results in the treatment of several categories of health conditions worldwide. The mechanisms of action of acupuncture (including immune mechanisms) have been investigated by biomedical studies over the last few decades. The immune mechanisms of representative clinical conditions and their clinical effects were thoroughly assessed, with a comprehensive investigation into the mechanisms of action of acupuncture (including immune responses) in this study. Conditions such as stroke, migraine, depression, chronic fatigue syndrome, lower back pain, hypertension, irritable bowel syndrome, sepsis, and allergic diseases were meticulously examined. This in-depth analysis aims to ensure a foundational understanding of the immune mechanisms involved in acupuncture, thereby serving as an initial step toward integrating the impact of acupuncture on the immune system.
