IL28B Is Associated with Outcomes of Chronic HBV Infection

Purpose: The role of IL28B gene variants and expression in hepatitis B virus (HBV) infections are not well understood. Here, we evaluated whether IL28B gene expression and rs12979860 variations are associated with HBV outcomes. Materialsand Methods: IL28B genetic variations (rs12979860) were genotyped by pyrosequencingof DNA samples from 137 individuals with chronic HBV infection [50 inactive carriers (IC), 34 chronic hepatitis B (CHB), 27 cirrhosis, 26 hepatocellular carcinoma (HCC)], and 19 healthy controls. IL28A/B mRNA expression in peripheralblood mononuclear cells was determined by qRT-PCR, and serum IL28B proteinwas measured by ELISA. Results: Patients with IL28B C/C genotype had greater IL28A/B mRNA expression and higher IL28B protein levels than C/T patients. Within the various disease stages, compared to IC and healthy controls, IL28B expression was reduced in the CHB, cirrhosis, and HCC cohorts (CHB vs. IC, p=0.02; cirrhosis vs. IC, p=0.01; HCC vs. IC, p=0.001; CHB vs. controls, p<0.01; cirrhosis vs. controls, p<0.01; HCC vs. controls, p<0.01). When stratified with respect to serum HBV markers in the IC and CHB cohorts, IL28B mRNA and protein levels were higher in HBeAg-positive than negative individuals (p=0.01). Logistic regression analysis revealed that factors associated with high IL28B proteinlevels were C/C versus C/T genotype [p=0.016, odds ratio (OR)=0.25, 95% confidence interval (CI)=0.08‒0.78], high alanine aminotransferase values (p<0.001, OR=8.02, 95% CI=2.64‒24.4), and the IC stage of HBV infection (p<0.001). Conclusion: Our data suggest that IL28B genetic variations may play an important role in long-term development of disease in chronic HBV infections.

Hydrostatic bearing has the advantages of wide speed range, large bearing capacity, high precision and stability. Hydrostatic-hydrodynamic hybrid bearing can make up for the lack of stiffness of hydrostatic bearing and prevent tribological failure under the condition of high speed and heavy load. Therefore, hydrostatic bearing and hydrostatic-hydrodynamic hybrid bearing have become important bearing parts of high-end computer numerical control (CNC) machine equipment. The research on hydrostatic and hydrostatic-hydrodynamic open a new way for precision improvement of high-end CNC machine equipment. This paper reviews research progress of hydrostatic linear guideway, hydrostatic rest and ram, hydrostatic thrust bearing, hydrostatic-hydrodynamic thrust bearing and hydrostatic-hydrodynamic spindle of high-end CNC machine equipment, and discusses the influence of structure and working parameters on lubrication performance, accuracy and stability of bearing parts. Finally, the future research direction in hydrostatic bearing and hydrostatic-hydrodynamic hybrid bearing are suggested. This review provides a theoretical basis for design and development of high-end CNC machine equipment in the future.

If a centrifugal impeller is in deep stall, it requires a large amount of additional energy to break free. As such, it is crucial to study the prediction method of impeller stall and establish its relationship with force and torque. This study employs a computational fluid dynamics numerical simulation method to analyze the flow field within the impeller of a centrifugal pump, while monitoring the changes in hydraulic torque of the blades. By examining the impeller’s internal flow field characteristics and external (energy performance) characteristics curves under different working conditions, the stall process was classified into three stages: germination, transition and stabilization. The evolution of stall cells under different stall stages was also analyzed. We utilized time-frequency conversion and analysis to extract characteristic parameters from instantaneous hydraulic torque results generated under different working conditions. A more effective model for predicting impeller stall stages was built by virtue of support vector machine algorithm. The proposed model exhibits improved feasibility and effectiveness in comparison to traditional prediction methods. It is expected to have significant implications for stable operation monitoring and safety protection of centrifugal pumps.

We undertook U-Pb-Hf isotope testing of detrital zircons from Late Paleozoic sedimentary rocks in the Jilin Province, NE China, with the aim of constraining the tectonic evolution of eastern Central Asian Orogenic Belt. The age populations of the detrital zircons from the sandstone of the Jiefangcun Formation in the Songliao Block are ~2234, ~1782, ~1030–921, ~817, ~540, and ~381–254 Ma, whereas those of the zircons from the greywacke of the Permian Kedao Formation in the Songliao Block yield are ~2436, ~1905, ~1692, 1146–1170, ~554–498, and ~342–255 Ma. Combined Hf isotope characteristics, these ages reveal that: (1) rocks from the Jiefangcun and Kedao formations were deposited after 254 and 255 Ma, respectively; (2) these formations were sourced mainly from the Songliao Block and the Jiamus Terrane in the eastern CAOB and to a lesser amount, from the northern margin of the NCC; and (3) it appears that several remnants of ancient Precambrian basement occur adjacent to these Late Paleozoic strata that occur within the microcontinental blocks. Detrital zircons from the silty sandstone of the Miaoling Formation in the western margin of the Khanka Terrane yield peak age populations of 508–476, 463–396, and 348–258 Ma, having similar εHf(t) values to those obtained from zircons within the Jiamusi Terrane. These data indicate that (1) deposition of the Miaoling Formation in the western margin of the Khanka Terrane had started by 258 Ma, and (2) these sediments were sourced mainly from the Jiamusi Terrane and local magmatic rocks. The widespread occurrence of detrital zircons with ages of 480–550 Ma and positive εHf(t) values within the Late Paleozoic strata in the Songliao Block and the Jiamusi–Khanka Terrane, indicates that the amalgamation of these blocks took place during the middle stage of the Paleozoic, probably after the Silurian, prior to the Late Permian.
Four New Antioxidant Phenylpropanoid Glycosides from Microlepia pilosissima

Four new phenylpropanoid glycosides, 9-O-[β-D-glucopyranosyl-(1→2)-α-L-rhamnopyranosyl]-3,4-dimethoxy-cinnamic acid (1), 9-O-[β-D-glucopyranosyl-(1→2)-α-L-rhamnopyranosyl]-4-methoxycinnamic acid (2), 9-O-α-L-rhamnopyranosyl-3,4-dimethoxy-cinnamic acid (3), and 9-O-[6-Oacetyl-β-D-glucopyranosyl]-4-methoxy-cinnamic acid (4), together with three known compounds 9-O-α-L-rhamnopyranosyl-4-hydroxy-cinnamic acid (5), 9-O-β-D-glucopyranosyl-4-methoxycinnamic acid (6), and 9-O-β-D-glucopyranosyl-3,4-dimethoxy-cinnamic acid (7) were isolated from the 70% EtOH extract of the dry fronds of Microlepia pilosissima. Their chemical structures were elucidated by spectroscopic analysis. Moreover, 1 and 2 exhibited comparable scavenging activities with (±)-α-tocopherol against DPPH radicals, while compounds 3-7 displayed moderate antioxidant activities.

<P>As ubiquitous molecular chaperones, small heat shock proteins (sHSPs) are crucial for protein homeostasis. It is not clear why sHSPs are able to bind a wide spectrum of non-native substrate proteins and how such binding is enhanced by heat shock. Here, by utilizing a genetically incorporated photo-cross-linker (<I>p</I>-benzoyl-<SMALL>l</SMALL>-phenylalanine), we systematically characterized the substrate-binding residues in IbpB (a sHSP from <I>Escherichia coli</I>) in living cells over a wide spectrum of temperatures (from 20 to 50 °C). A total of 20 and 48 residues were identified at normal and heat shock temperatures, respectively. They are not necessarily hydrophobic and can be classified into three types: types I and II were activated at low and normal temperatures, respectively, and type III mediated oligomerization at low temperature but switched to substrate binding at heat shock temperature. In addition, substrate binding of IbpB in living cells began at temperatures as low as 25 °C and was further enhanced upon temperature elevation. Together, these <I>in vivo</I> data provide novel structural insights into the wide substrate spectrum of sHSPs and suggest that sHSP is able to hierarchically activate its multi-type substrate-binding residues and thus act as a robust chaperone in cells under fluctuating growth conditions.</P>
A Sense Embedding of Deep Convolutional Neural Networks for Sentiment Classification

Sentiment classification task has attracted considerable interest as sentiment information is crucial for many natural language processing (NLP) applications. The goal of sentiment classification is to predict the overall emotional polarity of a given text. Previous work has demonstrate the remarkable performance of Convolutional Neural Network (CNN). However, nearly all this work assumes a single word embedding for each word type, ignoring polysemy and thus inevitably casting negative impact on the downstream tasks. We extend the Skip-gram model to learn multiple sense embeddings for the word types, catering to introduce sense-based embeddings for CNN during sentiment classification. Instead of using the pipeline method to learn multiple sense embeddings of a word type, the sense discrimination and sense embedding learning for each word type are performed jointly based upon the semantics of its contextual words. We validate the effectiveness of the method on the commonly used datasets. Experiment results show that our method are able to improve the quality of sentiment classification when comparing with several competitive baselines.
