A Population-Based Genomic Study of Inherited Metabolic Diseases Detected Through Newborn Screening

<P><B>Background</B></P><P>A newborn screening (NBS) program has been utilized to detect asymptomatic newborns with inherited metabolic diseases (IMDs). There have been some bottlenecks such as false-positives and imprecision in the current NBS tests. To overcome these issues, we developed a multigene panel for IMD testing and investigated the utility of our integrated screening model in a routine NBS environment. We also evaluated the genetic epidemiologic characteristics of IMDs in a Korean population.</P><P><B>Methods</B></P><P>In total, 269 dried blood spots with positive results from current NBS tests were collected from 120,700 consecutive newborns. We screened 97 genes related to NBS in Korea and detected IMDs, using an integrated screening model based on biochemical tests and next-generation sequencing (NGS) called NewbornSeq. Haplotype analysis was conducted to detect founder effects.</P><P><B>Results</B></P><P>The overall positive rate of IMDs was 20%. We identified 10 additional newborns with preventable IMDs that would not have been detected prior to the implementation of our NGS-based platform NewbornSeq. The incidence of IMDs was approximately 1 in 2,235 births. Haplotype analysis demonstrated founder effects in p.Y138X in <I>DUOXA2</I>, p.R885Q in <I>DUOX2</I>, p.Y439C in <I>PCCB</I>, p.R285Pfs<SUP>*</SUP>2 in <I>SLC25A13</I>, and p.R224Q in <I>GALT</I>.</P><P><B>Conclusions</B></P><P>Through a population-based study in the NBS environment, we highlight the screening and epidemiological implications of NGS. The integrated screening model will effectively contribute to public health by enabling faster and more accurate IMD detection through NBS. This study suggested founder mutations as an explanation for recurrent IMD-causing mutations in the Korean population.</P>

<P>In amorphous silicon solar cells, degradation is directly related to V(oc), FF and cell performance. The dependence of the stability of thin film amorphous silicon solar cells is studied in terms of the volume fraction of B2H6 in the p-layer. When the volume fraction of B2H6 is increased by an order of magnitude, the doping-induced defects tend to increase quite rapidly. Low-doped p-type a-SiO(x) layers had better initial properties but rapidly degraded. Heavily doped p-type a-SiO(x) layers had lower initial properties but displayed better stability. The improvement in stability is explained in conjunction with the capacitance and resistance values of impedance spectroscopy. When the B2H6 gas flow rate is increased, the cell is degraded showing a capacitance decay decrease from 51.75% to less than 18.18%. In addition, the increase in the resistance decreased from 90.90% to 11.73%.</P>

<P>We report on an Er-doped fiber oscillator that produces 146 fs pulses with 156 mW average power at a repetition rate of 49.9 MHz. The pulse energy reaches 3.13 nJ, surpassing the conventional power limit in the dispersion-managed soliton regime. Such high pulse power is obtained by devising a hybrid mode-locking scheme that combines saturable absorption with nonlinear polarization evolution. The oscillator also offers excellent temporal purity in the generated pulses with high power, providing a robust fiber-based frequency comb well suited for industrial uses.</P>
Multivariate Nonparametric Tests for Grouped and Right Censored Data

In this paper, we propose a nonparametric test procedure for the multivariate, grouped and right censored data for two sample problem. For the construction of the test statistic, we use the linear rank statistics for each component and apply the permutation principle for obtaining the null distribution. For the large sample case, the asymptotic distribution is derived under the null hypothesis with the additional assumption that two censoring distributions are also equal. Finally, we illustrate our procedure with an example and discuss some concluding remarks. In appendices, we derive the expression of the covariance matrix and prove the asymptotic distribution.

In this study, we propose a simultaneous test procedure based on the individual - values for each sub-null hypothesis with several well-known combining functions. We then compare the efficiency of our procedure with existing tests by obtaining empirical powers through a simulation study. Finally, we discuss some interesting features related to simultaneous test and point out a misconduct for the simulation study published in the previous work.
Coherent supercontinuum generation using Er-doped fiber laser of hybrid mode-locking.

<P>Supercontinuum generation is performed using femtosecond laser pulses produced by combining two distinct mode-locking mechanisms of nonlinear polarization evolution with saturable absorption on an Er-doped fiber oscillator. The generated supercontinuum yields a high level of spectral visibility of 93%, without the presence of spontaneous emission of post-amplification, being well suited for precision spectroscopy, broadband interferometry, and coherent telecommunications.</P>
