        OMC-2010 추출물이 마우스의 비장세포 cytokine 생성에 미치는 영향

        Objective : This study was performed to estimate the effects of OMC-2010 extract on cytokine production in mouse spleen cells. Methods : Mouse spleen cells were pre-treated with ethanol and water extract of OMC-2010 for 1 h, then stimulated with lipopolysaccharide (LPS, 1 μg/ml) for 48 h. Then the cells were harvested for real-time reverse transcription polymerase chain reaction to detect cytokines. Results : OMC-2010 ethanol extract significantly inhibited the LPS-induced interleukin (IL)-1beta, tumor necrosis factor (TNF)-alpha and IL-5 mRNA expressions, but not shown such changes in IL-6, IL-4, IL-13. OMC-2010 water extract significantly inhibited the LPS-induced TNF-alpha, and IL-5 mRNA expressions, but not shown such changes in IL-1beta, IL-6, IL-4, IL-13. Conclusions : Theses results could suggest that both ethanol and water OMC-2010 extract could inhibit the TNF-alpha and IL-5 mRNA expression.

        OMC-2010 구성약재 배합추출물 투여가 Ovalbumin으로 유도한 마우스 알레르기성 기관지 천식에 미치는 영향

        Objectives: We recently have reported that constituents of OMC-2010 have an immuno-modulatory effects via inhibiting tumor necrosis factor (TNF)-alpha and interleukin (IL)-5. In this study, based on previous data, we investigated the effects of combinations with each OMC constituents on splenocyte cytotoxicity, cytokine productions, and ovalbumin (OVA) induced experimental allergic asthma. Methods: Mouse splenocytes were pre-treated with ethanol extract of constituents of Rehmannia glutinosa (RG), Pinellia ternata (PT), Schisandra chinensis (SC). We made 4 combinations using RG, PT, and SC (A;1:1:1, B;2:1:1, C;1:2:1, D;1:1:2). The cells were pretreated with A, B, C, or D for 1 h, then stimulated with lipopolysaccharide (LPS, 1 ㎍/ml) for 48 h. Then the cells were harvested for real-time reverse transcription polymerase chain reaction to detect cytokine productions. Then using effective combination from RG, PR and SC, we administrated the combination orally, then challenged with OVA to induce asthma. Then we analyzed the airway hyper-reactivity (AHR), lung histology and lung TNF-α and IL-5 mRNA. Results: A. B. C. and D did not showed significant cytotoxicity on splenocytes. Pre-treatment of A inhibited the expression of TNF-α and IL-5 significantly, but not B, C, and D. In experimental asthma, administration of A significantly inhibited the increase of AHR, lung damage, TNF-α and IL-5 expression. Conclusions: Theses results could suggest that inhibitory effects of the ideal combination with RG, PT and SC (1:1:1) could be applied to treatment of asthma and study of asthma mechanisms.

        LPS로 유도한 RAW 264.7 세포의 염증반응에서 자초(紫草)의 항염증 효과

        Objective: Lithospermum Erythrorhizon (LE) has been used as an anti-bacterial and anti-inflammatory agent. However, it is unclear that LE aqueous extract could show the anti-inflammatory effects in RAW 264.7cells. The purpose of this study was to investigate the anti-inflammatory effect of aqueous extract from LE on lipopolysaccharide (LPS) - induced inflammatory response. Methods: To measure out the cytotoxicity of LE, we performed the MTT assay. To evaluate the anti-inflammatory effects of LE, we examined the inflammatory mediators such as nitric oxide (NO), prostaglandin E2 (PGE2) and pro-inflammatory cytokines (tumor necrosis factor (TNF)-a, interleukin, (IL)-1β and (IL)-6) on RAW 264.7 cells. We also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-B (NF-κB) activation by western blot. Results : Aqueous Extract from LE itself did not have any cytotoxic effect in RAW 264.7 cells. Aqueous extract from LE inhibited LPS-induced productions of inflammatory mediators such as NO, PGE2, and pro-inflammatory cytokines including TNF-a, IL-1β and IL-6 in RAW 264.7cells. In addition, LE inhibited the phosphorylation of p38 kinases (p38), c-Jun NH2-terminal kinase (JNK), and NF-κB activation in RAW 264.7 cells. Conclusion : LE down-regulated LPS-induced production of inflammatory mediators through the inhibition of p38, JNK and NF-κB activation. Taken together, these results could provide the evidence for the anti-inflammatory effects of LE. Therefore, LE may be a novel target in the management of inflammation and help to support a potential strategy for prevention and therapy of inflammatory diseases.

        감초부자탕(甘草附子湯)이 흰쥐의 Adjuvant 유발 관절염에 미치는 영향

        Objectives :This study was performed to investigate the effects of Gamchobuja-tang(gancaofu□zˇl-tang) on the Complete Freund`s Adjuvant(CFA)-induced arthritis in rats. Methods :Arthritis was induced by intradermal injection of CFA into the base of tail. Arthritic rats were divided into control(n=8) and Gamchobuja-tang(gancaofu□zˇl-tang) treated(n=8) group. Control group was taken distilled water for 15 days. Gamchobuja-tang(gancaofu□zˇl-tang) treated group was taken extracts of Gamchobuja-tang(gancaofu□zˇl-tang) by orally for same duration. Normal group(n=8) was injected with light mineral oil and was taken distilled water. Gross structure of hind paw were observed at every 5 days after treatment. Body weight, paw edema volume and ankle joint thickness were measured at 0, 5, 10, 15th day after treatment. Histopathological examination on the tarsal joint and distal tibia of the right hind paw were performed at the 15th day after treatment. White blood cell counts in blood were analysed at the 15th day after treatment. The content of interleukin-1β(IL-1β) and tumor necrosis factor-α(TNF-α) in paw exudate were analysed by enzyme-linked immunosorbent assay(ELISA) at the 15th day after treatment. Results :1. Grossly, the severity of arthritis in the Gamchobuja-tang(gancaofu□zˇl-tang) treated group were alleviated compared with control group. 2. Paw edema volume(ml) of Gamchobuja-tang(gancaofu□zˇl-tang) treat group was significantly decreased compared with control group. 3. Ankle joint thickness(mm) of Gamchobuja-tang(gancaofu□zˇl-tang) treat group was significantly decreased compared with control group. 4. Histopathologically, destructive lession of synovial membrane, articular cartilage and bony tissue in Gamchobuja-tang(gancaofu□zˇl-tang) treated group were alleviated compared with control group. 5. Total WBC(103/μl) of Gamchobuja-tang(gancaofu□zˇl-tang) treated group was significantly decreased compared with control group. 6. TNF-α content(pg/ml) in paw exudation of Gamchobuja-tang(gancaofu□zˇl-tang) treat group was significantly decreased compared with control group. Conclusions :On the basis of these results, Gamchobuja-tang(gancaofu□zˇl-tang) has anti-arthritic effects on the progression of CFA-induced arthritis in rat. And it`s effect is related with reduced secretion of TNF-α.

        우슬(牛膝) 등 복합 추출물의 monosodium iodoacetate로 유발한 흰쥐 골관절염에 대한 효과

        Objectives : The present study was designed to determine the effects of mixture of Achyranthis Japonicae Radix, Scutellariae Radix, and Acanthopanacis Cortex on monosodium iodoacetate (MIA)-induced osteoarthritis in rats. The mixture was composed of Achyranthis Japonicae Radix, Scutellariae Radix, and Acanthopanacis Cortex extracts. Methods : Arthritis was induced by injection of MIA into knee joints of rats. At the end of experiment, gross examination on the articular structures of knee joints were performed. Proteoglycan (PG) contents in articular cartilages were analysed as well. Tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) contents in synovial fluids were measured by ELISA method and matrix metalloproteinase 2 (MMP2), MMP9, and tissue inhibitors of metalloproteinase 1 (TIMP1) mRNA were measured by a realtime PCR. Results : The surfaces of the articular cartilage were observed. The severity of osteoarthritis in the treated group were alleviated compared with control group. PG contents in articular cartilages of the treated group were increased compared with control group. IL-1β contents in synovial fluids of the treated group were significantly decreased compared with control group. MMP2 and MMP9 mRNA contents in articular cartilages were significantly decreased compared with control group and TIMP1 mRNA contents were increased compared with control group. Conclusions : On the basis of these results, we concluded that Achyranthis Japonicae Radix-containing mixture treatment has anti-arthritic effects on the MIA-induced osteoarthritis in rats. And the effects were related with the reduction of IL-1β in synovial membranes and the consequent reduction of MMP2 and MMP9 expressions.

        Objectives : This study was to investigate the effects of GCP treatment on the monosodium iodoacetate-induced osteoarthritis in rats. Methods : Arthritis was induced by injection of Monosodium Iodoacetate(0.5mg) into knee joints of rats. Arthritic rats were divided into control(n=8) and treated(n=8) group. Control group was taken distilled water for 20 days. Treated group was taken extracts of GCP by oraly for same duration. Normal group(n=8) was injected with normal saline and was taken distilled water for 20 days. Body weight was measured at 0, 5, 10, 15, 20 days after injection. Macroscopic examination and histopathological study on articular cartilage of knee joint were operated at 20 days after injection. Proteoglycan(PG) content of articular cartilages of knee joint was represented by safranine O staining, was measured at 20 days after injection. Tumor necrosis factor-α, Interleukin-1β, Interleukin-6 in synovial fluid were measured with ELISA kit at 20 days after injection. Immunohistochemical staining of COX-2, iNOS in knee joints were observed at 20 days after injection. Results : 1. Body weight of the treated group increased compare with control group at 20 days after injection. 2. Macroscopically, degree of osteoarthritis in the treated group were evaluated compared with the control group. 3. PG content in articular cartilage of the treated group was significantly increased compared with the control group. 4. Histopathologically, osteoarthritic scores of the treated group was significantly decreased compared with the control group. 5. TNF-α content in synovial fluid of the treated group was decreased compared with the control group. 6. IL-1β content in synovial fluid of the treated group was significantly decreased compared with the control group. 7. Positive reaction of COX-2 in chondrocytes and synovial membrane of the treated group was faint compared with the control group. Conclusions : On the basis of these results, we concluded that GCP has inhibiting effects on the IL-1β and COX-2 secretion of chondrocytes and synovial membrane in Monosodium Iodoacetate-Induced osteoarthritis model of rats.

        Objectives : This study was to investigate the effects of Gamisoyeoum-tang(Jiaweixiaoyan-tang) on the monosodium iodoacetate-induced osteoarthritis in rats. Methods : Monosodium iodoacetate induced arthritic rats were divided into control and treated group. Control group was taken distilled water for 20 days. Treated group was taken extracts of Gamisoyeoum-tang(Jiaweixiaoyan-tang) by orally for same duration. Normal group was injected with normal saline and was taken distilled water. Body weights were measured at 0, 5, 10, 15, 20 days after injection. At the end of experiment, gross and histopathological examination on the articular cartilages of the knee joints were performed. Contents of TNF-α, IL-1β, IL-6 in synovial fluids and proteoglycan contents of articular cartilages were analysed. COX-2 and iNOS immunohistochemical examination on the knee joints were performed. Results : 1. Body weights of the treated group were significantly increased compared with control group at 20 days after injection. 2. Grossly, the severity of osteoarthritis in the treated group were alleviated compared with control group. 3. PG contents in articular cartilages of the treated group were significantly increased compared with control group. 4. Histopathologically, degenerative and necrotic lesion of articular cartilages in the treated group were alleviated compared with those of the control group. 5. IL-1β contents in synovial fluids of the treated group were significantly decreased compared with control group. 6. Positive reactions of COX-2 in chondrocytes and synovial membranes of the treated group were decreased compared with the control group. Conclusions : On the basis of these results, we concluded that Gamisoyeoum-tang(Jiaweixiaoyan-tang) has anti-arthritic effects on the monosodium iodoacetate-induced osteoarthritis in rats. And it`s effects were related with reduced secretion of IL-1β and COX-2 from osteoarthritic chondrocytes and synovial membranes.

      • 가감목방기탕(加減木防己湯)이 흰쥐의 Adjuvant 유발 관절염에 미치는 영향

        Objectives : This study was to investigate the effect of Kagammokbanggi-tang on the Freund's Complete Adjuvant(FCA)-induced arthritis in rats. Methods : Arthritis was induced by intradermal injection of FCA into base of tail. Arthritic rats were divided into control(n=10) and sample(n=10) group. Control group was taken normal saline for twenty days and sample group was taken extracts of Kagammokbanggi-tang for same duration. Normal group(n=10), non-arthritic group, was injected with mineral oil and was taken normal saline for twenty days. Body weight, paw edema volume and ankle joint thickness were measured at 0, 10, 15, 20 days after treatment. $TNF-{\alpha}$, $IL-1{\beta}$, IL-6, $PGE_2$ in synovia were analysed by ELISA at 20 days after treatment. Histochemical investigation of NADPH-d in the PAG and histopathological study on the ankle joint were performed at 20 days after treatment. Results : Paw edema volume, ankle joint thickness, $TNF-{\alpha}$, $IL-1{\beta}$, IL-6, $PGE_2$, NADPH-d of sample group were significantly decreased compared with control group. Conclusions : These results indicated that Kagammokbanggi-tang has antiarthritic and analgesic effects, and inhibited expression of NOS on the progression of FCA- induced arthritis in rats.

        방풍통성산이 흰쥐의 Adjuvant 유발 관절염에 미치는 영향

        Objectives :This study was carried out to investigate the effects of Bangpungtongseong-san(Fangfengtongsheng-san) treatment on the change of the adjuvant-induced arthritis in rats. Methods :Arthritis was induced by injection of complete Freund`s adjuvant(CFA, 0.1 ml) into the tail base of rats. Arthritic rats were divided into control(n=8) and treated(n=8) group. The control group was taken distilled water and the treated group, extracts of Bangpungtongseong-san(Fangfengtongsheng-san) by orally for 15 days. At the end of the experiment(28 days after CFA injection), gross and histopathological examinations on the articular structures of ankle joints were performed. The total number of white blood cells was measured by the ADVIA(automated blood cell counter, Bayer Co., Ireland). And also, tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) contents in paw exudate of rats were measured by enzyme-linked immunosorbent assay(ELISA) method. Results :1. Body weight(g) gain rate(%) of the treated group was increased significantly compared with control group at the 15th day after injection. 2. The thickness of ankle joints of the treated group was decreased significantly compared with the control group. 3. Histopathologically, finding of fibrous ankylosis of the treated group was decreased compared with the control group. 4. The total number of white blood cells in the treated group was decreased significantly compared with the control group. 5. TNF-α content and IL-1β content in paw exudate of the treated group were decreased significantly compared with the control group. Conclusions :On the basis of these results, we suggest that Bangpungtongseong-san(Fangfengtongsheng-san) has inhibiting effects on the progression of arthritis in CFA-induced arthritis model.

        가시오가피가 흰쥐의 Collagen 유발 관절염 발생 및 치료에 미치는 영향

        Objectives : This study was carried out to investigated the anti-pathogenetic and curative effects of Radix Cortex Acanthopanacis Senticosis (CAS) on collagen-induced arthritis in rats. Methods : In experiment Ⅰ, rats(n=30) were divided into normal group, porcine type II collagen immunized control group which taken with normal saline, and collagen immunized treated group which taken with extract of CAS. Body weight, paw edema volume, and thickness of ankle joint were measured at 0, 10, 15 days after immunization. At 15 days after immunization, serum TNF-α, IL-1β and anti-collagen IgG level were analysed, and histopathological examination was performed on the ankle joint. In experiment Ⅱ, rats(n=13) that were shown the gross lesions of collagen-induced arthritis, were divided into control group and treated group which taken with extract of CAS for 15 days after onset of arthritis. Items and methods of measurement were the same as experiment Ⅰ. Results : In experiment I, the results were as follows ; Arthritic score, paw edema volume, thickness of ankle joint, TNF-α, anti-collagen IgG of treated group were significantly decreased. In experiment Ⅱ, the results were as follows ; Paw edema volume, TNF-α, IL-1β, anti-collagen IgG of treated group were significantly decreased. Conclusions : These results indicated that CAS has the inhibitory effects on the incidence and progression of collagen-induced arthritis in rats.
