Significance of p16 Positive Expression in Oropharyngeal Cancers

Background: The involvement of HPV in oral and oropharyngeal carcinogenesis was first proposed in 2004, based on epithelial HPV tropism and detection of HPV genotypes in oral squamous cell carcinoma samples. While 60-70% of oropharynx tumors may be HPV-positive, only 10 to 19% of tumors of the oral cavity, larynx and hypopharynx appear to have HPV infection. The aim of the study was to evaluate HPV infection associated with oropharyngeal cancer. Materials and Methods: Seventy-eight cases were selected for p16 immunoexpression reactions, and demographic data were collected for comparisons. Results: Most patients were over 60 years old, and 64.1% were smokers. Immunohistochemistry results showed that 86.3% of cases stained positive for p16 protein. Conclusion: The oropharyngeal cancer profile at Erasto Gaertner Hospital presented a high index of smokers over 60 years as well a high number of p16+ tumors, for what we can not determinate the main etiologic factor, but can be aware of the number of patients that presented HPV infection. Since prevention is still the best way to deal with cancer disease, it is important to analyze the interaction of these two etiologic factors and how to detect lesions at an early stage.
Study of the regenerator constituting material influence on a gamma type Stirling engine

A gamma Stirling engine with compressed air as working fluid was investigated. This engine operates at a maximum charge pressure of 10 bar, runs at a maximum rotation speed of 600 rpm and can provide 500 W of brake power on the shaft. The engine is equipped with several pressure sensors and thermocouples. This experimental study concentrates on the regenerator constituting material (porous medium). Four different materials were investigated: stainless steel, copper, aluminum and Monel 400. The obtained experimental results provide guidance to Stirling engine enhancement and selection of the appropriate regenerator material. As a conclusion, the regenerator has an important role to enhance the heat exchange and to improve Stirling engine performance, which closely depends on its constituting material.
Chemical Risk Assessment Screening Tool of a Global Chemical Company

Background: This paper describes a simple-to-use and reliable screening tool called Critical Task Exposure Screening (CTES), developed by a chemical company. The tool assesses if the exposure to a chemical for a task is likely to be within acceptable levels. Methods: CTES is a Microsoft Excel tool, where the inhalation risk score is calculated by relating the exposure estimate to the corresponding occupational exposure limit (OEL) or occupational exposure band (OEB). The inhalation exposure is estimated for tasks by preassigned ART1.5 activity classes and modifying factors. Results: CTES requires few inputs. The toxicological data, including OELs, OEBs, and vapor pressure are read from a database. Once the substance is selected, the user specifies its concentration and then chooses the task description and its duration. CTES has three outputs that may trigger follow-up: (1) inhalation risk score; (2) identification of the skin hazard with the skin warnings for local and systemic adverse effects; and (3) status for carcinogenic, mutagenic, or reprotoxic effects. Conclusion: The tool provides an effective way to rapidly screen low-concern tasks, and quickly identifies certain tasks involving substances that will need further review with, nevertheless, the appropriate conservatism. This tool shows that the higher-tier ART1.5 inhalation exposure assessment model can be included effectively in a screening tool. After 2 years of worldwide extensive use within the company, CTES is well perceived by the users, including the shop floor management, and it fulfills its target of screening tool.

There is an ever growing interest in the development of biochar from a large variety of agrowastes. Herein, the main objective is the conversion of pomegranate peel powder biochar and its post-functionalization by phosphoric acid treatment, followed by arylation organic reaction. The latter was conducted using in situ-generated diazonium salts of 4-aminobenzoic acid (H2N-C6H4-COOH), sulfanilic acid (H2N-C6H4-SO3H) and Azure A dye. The effect of diazonium nature and concentration on the arylation process was monitored using thermal gravimetric analysis (TGA) and Raman spectroscopy, scanning electron microscopy (SEM), and X-ray photoelectron spectroscopy (XPS). SEM pictures showed micrometer-sized biochar particles with tubular structure having about 10–20 µm-wide channels. SEM studies have shown that arylation did not affect the morphology upon arylation. The porous structure did not collapse and withstood the arylation organic reaction in acid medium did not collapse upon arylation. TGA and Raman indicated gradual changes in the arylation of biochar at initial concentrations 10–5, 10–4 and 10–3 mol L−1 of 4-aminobenzoic acid. The detailed Raman spectra peak fittings indicate that the D/G peak intensity ratio leveled off at 3.35 for 4-aminobenzoic acid initial concentration of 10–4 mol L−1, and no more change was observed, even at higher aryl group mass loading. This is in line with formation of oligoaryl grafts rather than the grafting of new aryl groups directly to the biochar surface. Interestingly, Azure A diazonium salt induced much lower extent of surface modification, likely due to steric hindrance. To the very best of our knowledge, this is the first report on diazonium modification of agrowaste-derived biochar and opens new avenues for arylated biochar and its applications.

Objectives: The aim of this study was to evaluate the profile of patients on antiresorptive therapies for cancer treatment and assess presence of oral lesions, oral hygiene status, and knowledge regarding medication-related osteonecrosis of the jaw (MRONJ). Materials and Methods: This was an observational cross-sectional study that evaluated patients treated with antiresorptive medication at a single cancer hospital. Clinical data were collected and oral examination was performed to assess patient oral health. Results: From July 2017 to December 2018, 90 patients were assessed; 64 were female and 26 were male, and the mean age was 61 years. The most common drug was an intravenous bisphosphonate, zoledronic acid. Among the 90 patients, 47 presented with some type of oral disease, isolated or associated. Among these 47 patients, 9 patients (10%) developed osteonecrosis. Oral hygiene was evaluated, and most patients, with or without MRONJ, presented with regular to poor condition. Regarding patient knowledge of the risks of MRONJ and the risks associated with dental surgery, 60% stated that they were not aware of the risks. Conclusion: Identifying the profile of patients and their needs facilitates not only the preventive process, but also the emergence of new therapeutic options. Our study shows that most patients are weakened both by metastatic disease and antineoplastic treatment as well as by issues associated with aging because most were over 60 years of age. Collectively, this information should be considered for management of preventive and therapeutic measures.

Although kidney bean (Phaseolus vulgaris L.) lectin toxicity is widely known, its effects in the gastrointestinal tract require further study. This investigation aimed to identify and characterize phytohemagglutinins (PHAs) in the small intestine and sera of rats following oral challenge with ground white beans. Twenty young, adult male rats were divided randomly into two groups of 10 animals each. The control group underwent gavage with a suspension of 300 mg of rodent pellet flour. The experimental group was administered a 300 mg Beldia bean flour suspension (BBFS). After 10 days of daily treatment, jejunal rinse liquid ( JRL) and ileum rinse liquid and secretions, as well as sera, were collected. All biological fluids were screened for lectin reactivity using competitive inhibition ELISA, Ouchterlony double immunodiffusion, and immunoelectrophoresis techniques. The results revealed the presence of immunogenic intraluminal PHAs 3–4 h after the oral intake of the BBFS in the JRLs as well as in the jejunal and ileal secretions; however, no PHA was detectable in the rat sera. Ingestion of raw Beldia beans may lead to interaction between PHAs and the mucosa of the small intestine, potentially resulting in an inflammatory response.
