      • QTL mapping for grain width using near-isogenic lines from a cross between ‘Hwaseongbyeo’ and Oryza rufipogon

        Grain size is a major determinant of grain yield in rice. In a previous study, a QTL for grain width(GW), qgw1 was detected on chromosome 1 using 96 BC3F8 lines derived from a cross between ‘Hwaseongbyeo’ as a recurrent parent and ‘O. rufipogon’ as a donor parent. At this locus, the O. rufipogon allele increased GW. Among the 96 introgression lines, three ILs with the O. rufipogon qgw1 locus showed significantly increase in grain width compare to the recurrent parent. One of the three lines, CR572 was selected and crossed to ‘Hwaseongbyeo’. A total of 494 F2:3 were evaluated for grain width and agronomic traits in the field. QTL analysis in 494 F2:3 lines indicated that QTL for grain width was located in the interval RM495–RM5443. To narrow down the position of qgw1, substitution mapping using F4 lines with different cross-over breakpoints in the region is underway. The result will be discussed.

      • Mapping quantitative trait loci for agronomic traits using introgression lines carrying wild rice (Oryza rufipogon) chromosome segments in cultivar background

        The objective of this study were to identify QTLs for agronomic traits using a set of introgression lines carrying wild rice (Oryza rufipogon) segment in cultivated rice (ssp. japonica cv. Hwaseongbyeo). Ninety-six ILs were evaluated for seven agronomic traits, amylose and protein contents. The proportion of the recurrent genome in ILs ranged from 87.8 to 100%, with an average of 96.7%. The mean number of homozygous and heterozygous donor segments were 2 (ranging 0-7) and 1.7 (ranging 0-6), respectively, and the majority of these segments had size less than 10 cM. A total of 22 quantitative trait loci were identified for 9 traits and each QTL explained 7.2% to 56.6% of the phenotypic variance. Some QTLs were clustered in a few chromosomal regions. A first cluster was located near RM527 on chromosome 6 with QTLs for culm length, panicle length, days to heading, 1000-grain weight and protein content. Three ILs with high spikelets per panicle compared to the recurrent parent were selected to detect and fine map the wild segments responsible for this variation. The results will be discussed.

        This study was carried out to elucidate the inheritance of aroma in rice. Aromatic nature was tested with leaves of two aromatic/non-aromatic crosses and six crosses among aromatic cultivars. All F₁ hybrids of non-aromatic cultivar Hwaseongbyeo and aromatic cultivars, Sanghaehyanghyeolla, Miyakaori, Della, Selatus Malam, Manong Balay, Hyangmibyeo 1 and Chokoto were not aromatic and this seemed to indicate the recessive nature of the aroma in all tested rices. F₂ segregation ratio of 3 non-aromatic : 1 aromatic individual suggested that Sanghaehyanghyeolla and Miyakaori each contained a single gene for aroma. The finding that all F₁ hybrids among six aromatic rice cultivars were aromatic suggested that the aroma in Sanghaehyanghyeolla and Miyakaori may be controlled by the same gene known to be in Della and the aroma genes in Manong Balay and Chokoto are also allelic although the number of genes controlling the aroma in these cultivars are not known. No linkage was detected between the aroma gene(fgr) in Sanghaehyanghyeolla and two genetic markers(sug and v-8) located on rice chromosome 8.

        Relationship Between Heterosis and Genetic Divergence in Tongil-type Rice(Oryza sativa L.)

        RAPDs 분석에 양친의 유전적 유사성 정도가 F1 잡종강세 예측에 효율적으로 이용될 수 있는지 알아보고자 통일형 6 품종 (밀양42, IR747, 백운찰벼, 삼강벼, 수원307, 이리 356) 간 반이면 교배 F1 15조합을 작성하였다 양친과 F1을 공시하여 조사한 수량 등 8개의 특성과 RAPDs 분석을 통해 구한 양친의 유전적거리와의 관계를 조사하였다. 120개의 Operon 프라이머를 6개 품종의 DNA 증폭에 이용한 결과 336개의 밴드가 발생되었으며 이중 168개가 6개 품종 중 1개 이상의 품종에서 변이를 보였다. 6품종간 유전적 거리는 최소 0.157(백운찰벼와 수원307호 간), 최대 0.383(삼강벼와 이리356호 간)의 분포를 보였다. 잡종강세의 정도는 정조수량(129%), 수당립수(125%), 수장(109%) 등이 높은 잡종강세를 보였고, 유전적거리는 수당립수와 통계적으로 유의한 부의 상관을 보였지만, 정조수량, 천립중등과는 유의한 상관을 보이지 않았다 In this study we evaluated genetic divergence among six Tongil-type rice varieties and assessed the relationship between genetic &stance and hybrid performance in all possible non-reciprocal crosses between them The 15 F1 hybrids along with the six parents were evaluated for eight traits of agronomic importance including yield in a replicated field trial The six parents were examined for DNA polymorphism using 120 random decamer oligonucleotide primers A total of 168 polymorphic variants were generated and based on the polymorphism data. genetic distances (GDs) ranged from 0157 to 0.333 Heterosis was observed in hybrids for most of the traits, and yield exhibited the highest heterosis among the eight traits examined The correlation values of GDs with F1 performance were not significant. Also the correlations of GDs with midparent and better-parent heterosis were not significant except for spikelets per panicle Our results have indicated that GDs based on the RAPDs markers may not be useful for predicting heterotic combinations in Tongil-type rice and are supportive of the idea that the level of correlations between hybrid performance and genetic divergence is dependent on the germplasm emploved.

      • Validation of QTLs associated with spikelets per panicle and grain weight in rice

        <P>In this study, a near-isogenic line (BC4F10) CR572 developed by introgressing a chromosomal segment from <I>Oryza rufipogon</I> (accession no. 105491) into the <I>Oryza sativa</I> subsp. <I>japonica</I> cv. Hwaseong was found to exhibit a significant increase in the number of spikelets per panicle (SPP) and grain weight compared with the recurrent parent Hwaseong. Quantitative trait locus (QTL) analysis in F2 generation derived from the cross between CR572 and Hwaseong revealed that two QTLs, <I>qSPP1</I> and <I>qTGW1</I>, were linked to a simple sequence repeat marker, RM283, on chromosome 1. The additive effect of the <I>O. rufipogon</I> allele at <I>qSPP1</I> was 13 SPP, and 21.6% of the phenotypic variance was explained by the segregation of RM283. The <I>qTGW1</I> QTL explained 19.1% of the phenotypic variance for grain weight. Substitution mapping was carried out with five F3 lines derived from F2 plants having informative recombination breakpoints within the target region. Substitution mapping indicated the linkage of <I>qSPP1</I> and <I>qTGW1</I>. The grain yield of CR572 was 18.2 and 15.8% higher than that of Hwaseong at two locations, respectively, mainly due to the increase in 1000-grain weight and SPP. These results are very useful for QTL cluster transfer by molecular marker-assisted selection in rice breeding programmes and for QTL gene cloning by map-based cloning.</P>

        Mapping of QTLs for Yield Traits Using an Advanced Backcross Population from a Cross between Oryza sativa and O. glaberrima

        An interspecific BC3F2 population (Milyang 23/O. glaberrima/3*Milyang 23) consisting of 141 lines was evaluated for8 traits associated with yield. Oryza glaberrima (IRGC #103544) was phenotypically inferior to all of the traits examined in thisicle and grain yield. A genetic linkage map generated for this population consisted of 124 SSR markers with an average intervallength of 13.1 cM. The percentage of O. glaberrima genome, which was calculated on the number of the exotic gene introgres-sions contained in the 141 BC3F2 lines, ranged from 3.9 to 25.4% with a mean of 12.8%. Of the 37 QTLs associated with yield andyield components, 24 alleles from O. glaberrima were favorable. However, eight QTLs were significantly associated with yieldand five alleles of O. glaberrimafrom 1.5 to 2.2 tons/ha. Exploitation of the genetic potential of exotic species using molecular markers might provide a new oppor-tunity to improve yield and quantitative traits in rice breeding program.
