This study was carried out to select effective preventive pesticides against pine wilt disease caused by pinewood nematode (PWN), Bursaphelenchus xylophilus on trunk injection. 1,000 fold aquatic solution of abamectin 1.8% EC and emamectin benzoate 2.15% EC were lower mortality (7.3% and 8.3% respectively) against PWN on 1 day after treatment. However effects of abamectin 1.8% EC, emamectin benzoate 2.15% EC, fosthiazate 30% SL and fenitrothion 30% SL were inhibited the reproduction of PWN over 99.6% in Botrytis cineria media. Effect of trunk injection of abamectin 1.8% EC and emamectin benzoate 2.15% EC at the rate of 10 ㎖ per 10 ㎝ in diameter of breast height (DBH) on mortality of Japanese black pine, Pinus thungergii by inoculated PWN was 0% and 3.3%, respectively at the applied year however when injection of fosthiazate 30% SL were treated with the rate of 5 ㎖ per 10 cm tree DBH, mortality of tree was 63.3%. Abamectin 1.8% EC and emamectin benzoate 2.15% EC was showed high preventive efficacy representing >90% against PWN at the following year. PWN preventing efficacy of trunk injection was lower in naturally occurred area (mortality of pine tree in control was 11.7% at the first year) of PWN than artificially infected site (mortality of pine tree in control was >76.7% at the first year), PWN preventing efficacy of trunk injection of abamectin 1.8% EC and emamectin benzoate 2.15% EC at the rate of 10 ㎖ per 10 ㎝ in DBH was 91.5% and 82.9%, respectively, at the applied year and 89.5% and 82.6% respectively at the following year in PWN naturally occurred site. Control efficacy by trunk injection of abamectin 1.8% EC and emamectin benzoate 2.15% EC was more higher in 10 fold dilution with 10 fold high amount of aquatic solution than no dilution with 10 fold less amount of aquatic solution. The preventive effect of trunk injection of abamectin 1.8% EC and emamectin benzoate 2.15% EC at the rate of 5 ㎖ per 10 ㎝ in DBH was showed 100% at the applied year in PWN inoculated tree.

Transcatheter arterial chemoembolization (TACE) is suitable treatment method of hepatocellular carcinoma (HCC) for patients with decreased underlying liver function or unresectable disease. But this procedure can induce some embolic events. We report a case of a gallbladder infarction caused by the embolic event in TACE procedure. A 61-year-old man had been going to undergo 6th TACE for marginal recurrence of HCC. After the procedure, gallbladder infarction occurred and was detected by abdominal computed tomography (CT) scan for response evaluation of the treatment at out-patient-clinic. Then, he was treated with oral antibiotics and conservative therapy at out-patient clinic. In 6th week after the event, abdominal CT scan showed improved state of gallbladder.
Improvement of Peptide Expression Vector by Mutagenesis

The goal of the present study is to improve the preS2-expression vector in such a way that purification of the recombinant preS2 peptide of hepatitis B virus surface antigen is facilitated. Previously we have succeeded in expressing the β-galactosidase-preS2 fusion protein in E. coli with a yield of 50% of total cellular proteins. Since there is no internal Met residue in the preS2 sequence, we could use CNBr cleavage in isolating the preS2 peptide out of the fusion protein. In amino acid sequence of the fusion protein encoded by pCTHB20 plasmid, there exist three internal Met residues: two in the β-galactosidase portion and one at the beginning of preS2 sequence. In the present study we altered those two Met codons in the lacZ portion by oligonucleotide-directed mutagenesis. DNA fragment containing the target sequence was cloned into single strand DNA phage M13 mp18 vector and two mutagenic oligonucleotides (17-mer) were used as primers in synthesizing the complementary strand. Mutant clones were selected by hybridization with ^(32)P-labeled mutant oligonucleotides as probes. DNA sequencing confirmed that those two target Met codons (ATG) were changed into Val codons (GTG). The mutagenized sequence of lacZ in the recombinant M13 vector was isolated and replaced for the wild type counterpart of pCTHB20 plasmid. The resulting plasmid was named as pCMHB20. Expression of β-galactosidase-preS2 fusion proteins was carried out and the presence of preS2 sequence in the fusion protein was confirmed by Western blotting method with preS2-specific monoclonal antibody and peroxidase-conjugated anti-mouse IgG antibody.

<P>Amorphous In-Ga-Zn-O (a-IGZO) thin-film transistors (TFTs) fabricated by low-temperature processes on a flexible substrate can easily be degraded by mechanical deformation. Furthermore, lower performance in terms of the initial characteristics and reliability levels compared to those fabricated on glass substrates with relatively high heat treatments is inevitable. To solve these problems, a local electro-thermal annealing (ETA) method was applied to flexible a-IGZO TFTs processed at low temperature to enhance the inferior initial characteristics and reliability under a bending state. The enhancement of the characteristics and reliability by ETA can be attributed to the reduction of defects related to the oxygen through a localized Joule heat treatment with an extremely short duration (similar to 1 ms). In addition, the effectiveness of ETA to recovery from bending stress even under harsh cyclic bending operation (strain condition of 0.833%) is verified.</P>

The effects of column length and particle size on the efficiency of separation and characterization of phospholipids (PLs) are investigated using nanoflow liquid chromatography-electrospray ionization-tandem mass spectrometry (nLC-ESI-MS-MS). Since PLs are associated with cell proliferation, apoptosis, and signal transduction, it is of increasing interests in lipidomics to establish reliable analytical methods for the qualitative and quantitative profiling of PLs related to biomarker development in adult diseases. Due to the complexity of PLs, the preliminary separation of PLs is necessary prior to MS analysis. In this study, length of capillary column and the particle size of reversed phase ($C_{18}$) packing materials are varied to find a reliable condition for the high speed and high resolution separation using 8 PL standard mixtures. From experiments, it was found that a capillary column of nLC-ESI-MS-MS analysis for PL mixtures can be minimized to a 5 cm long pulled tip column packed with 3 ${\mu}m$ $C_{18}$ particles without losing resolution.

<P>Given their important role in neuronal function, there has been an increasing focus on altered lipid levels in brain disorders. The effect of a high-fat (HF) diet on the lipid profiles of the cortex, hippocampus, hypothalamus, and olfactory bulb of the mouse brain was investigated using nanoflow ultrahigh pressure liquid chromatography-electrospray ionization-tandem mass spectrometry in the current study. For 8 weeks, two groups of 5-week-old mice were fed either an HF or normal diet (6 mice from each group analyzed as the F and N groups, respectively). The remaining mice in both groups then received a 4-week normal diet. Each group was then subdivided into two groups for another 4-week HF or normal diet. Quantitative analysis of 270 of the 359 lipids identified from brain tissue revealed that an HF diet significantly affected the brain lipidome in all brain regions that were analyzed. The HF diet significantly increased diacylglycerols, which play a role in insulin resistance in all regions that were analyzed. Although the HF diet increased most lipid species, the majority of phosphatidylserine species were decreased, while lysophosphatidylserine species, with the same acyl chain, were substantially increased. This result can be attributed to increased oxidative stress due to the HF diet. Further, weight-cycling (yo-yo effect) was found more critical for the perturbation of brain lipid profiles than weight gain without a preliminary experience of an HF diet. The present study reveals systematic alterations in brain lipid levels upon HF diet analyzed either by lipid class and molecular levels.</P>

<P><B>Abstract</B></P> <P>A comprehensive lipid analysis was performed at the plasma lipoprotein level in patients with acute coronary syndrome (ACS) and stable coronary artery disease (CAD). Because the lipids in lipoproteins are related to the pathology of the cardiovascular system, lipoprotein-specific lipid analysis can be useful for understanding the mechanism of lipid-associated cardiovascular diseases. Lipoproteins were size-sorted into high density lipoproteins (HDL) and low density lipoproteins (LDL) using asymmetrical flow field-flow fractionation, then lipids of each lipoprotein were analysed using nanoflow ultrahigh performance liquid chromatography-electrospray ionization-tandem mass spectrometry. A total of 365 lipids were structurally identified and quantified by selected reaction monitoring method. Two high abundance lysophosphatidylcholines (16:0 and 18:0) were significantly increased only in the HDL of the ACS group (vs. the stable CAD group). Phosphatidylethanolamines (38:5 and 40:5) significantly increased in ACS by >2-fold in both lipoproteins. (18:0, 22:6)-diacylglycerol increased in ACS by 3.5-fold only in LDL; however, most high abundance triacylglycerols decreased 2-fold in both lipoproteins. The present study revealed the usefulness of lipoprotein-specific analysis of lipids in distinguishing ACS from stable CAD, and the selected lipids analysed in this study may be useful in the development of lipid markers for the early detection of ACS.</P>

Fault prognostics of an air compressor is a challenging issue for condition-based maintenance of a railway vehicle. In this paper, we propose a mathematical model of a two-stage reciprocating air compressor to obtain normal and abnormal data for the pressure and temperature for various parts of the air compressor. The mathematical model includes kinematic equations of crank mechanism, thermodynamic equations of cylinders, valve flow equations, and heat transfer equations of cylinder blocks. Especially, heat transfer effects at cylinder blocks are simulated for the cases with and without heat transfer. Simulation results show that the mathematical model is valid and heat transfer effect at cylinder blocks is minimal to cylinder pressure but affects cylinder temperature significantly.
