Expression of Chemokines and Chemokine Receptors in Brain Tumor Tissue Derived Cells

Chemokine and chemokine receptor expression by tumor cells contributes to tumor growth and angiogenesis and thus these factors may be considered as tumor markers. Here we aimed to characterize cells directly extracted from glioma, meningioma, and secondary brain tumors as well as non-tumoral cells in vitro. Cells were isolated from brain tissues using 0.2% collagenase and characterized by flow cytometry. Expression of SDF-1, CXCR4, CXCR7, RANTES, CCR5, MCP-1 and IP-10 was defined using flow cytometry and qRT-PCR methods. Brain tissue isolated cells were observed as spindle-shaped cell populations. No significant differences were observed for expression of SDF-1, CXCR4, CXCR7, RANTES, CCR5, and IP-10 transcripts. However, the expression of CXCR4 was approximately 13-fold and 110-fold higher than its counterpart, CXCR7, in meningioma and glioma cells, respectively. CXCR7 was not detectable in secondary tumors but CXCR4 was expressed. In non tumoral cells, CXCR7 had 1.3-fold higher mRNA expression than CXCR4. Flow cytometry analyses of RANTES, MCP-1, IP-10, CCR5 and CXCR4 expression showed no significant difference between low and high grade gliomas. Differential expression of CXCR4 and CXCR7 in brain tumors derived cells compared to non-tumoral samples may have crucial impacts on therapeutic interventions targeting the SDF-1/CXCR4/CXCR7 axis.

The lowering intake of organic dyes and their auxiliaries by employing nanoparticles for some specialtextile coloration is novel and interesting topic. Nonetheless, application of reducing, stabilizing andother chemical agents in textile nano-coloration is still undesirable from both environmental andhuman/aquatic life viewpoints. This research renders a facile technique for wool coloration throughlocally forming selenium nanoparticles using sodium selenite without using other chemicals. Beingindustrially enforceable, free from use of additional chemicals, showing photo-active ability to discolorthe dyeing effluent and low-toxic colorant are the main privileges of the used method. The wool fabricswere colored light to dark orange depending on coloration time and concentration of sodium selenite. Also, the color fastness properties against washing, rubbing and acid/alkaline spotting of prepared sampleswere excellent. Moreover, the colored fabrics indicated reasonable bactericidal and fungicidal performancestoward Escherichia coli, Staphylococcus aureus and Candida albicans (95, 70 and 100 %,respectively) and still remained compatible with human skin. The in-situ synthesis of selenium nanoparticleson wool fabric not only colored the wool but also promoted the UV-protecting performance by61.82 %, photocatalytic activity through methylene blue discoloration under sunlight by 97.22 % and tensilestrength by 11.27 %.

One of the seismic retrofit methods is applying heat to transfer the failure location from the near column into the beam. In this method, by heating a section of the beam, changes the mechanical characteristics of the beam, such as enhancing the ductility and decreasing the yield as well as the ultimate strength in that section, thus causing an altered place of failure as well as changes in the failure mode. In this study, via experimental and numerical methods an investigation was conducted on the ultra-low cycle fatigue capacity in retrofitted St-37 steel connection with three models of thermal improvement with the titles HBS1, HBS1&RIB, and HBS2 which differ in the cooling rate after the heating operation. The connection without retrofit failed in the cycle of 5% drift angle of SAC protocol loading. Retrofitted connections with HBS1 and HBS1&RIB thermal models failed in the cycle of 6% drift angle of SAC protocol loading. The results of modeling these connections in software and using the ultra-low cycle fatigue failure index also revealed a failure in a cycle with a 6% drift angle. The seismic capacity of the retrofitted connection with the HBS2 thermal model, with one percent increase in seismic capacity, failed in the cycle of 7% of the drift angle. The result of numerical modeling of this connection in the software was the same and indicated the ultra-cycle fatigue failure occurring in the cycle of 7% of the drift angle.
Families of Distributions Arising from Distributions of Ordered Data

A large family of distributions arising from distributions of ordered data is proposed which contains other models studied in the literature. This extension subsume many cases of weighted random variables such as order statistics, records, k-records and many others in variety. Such a distribution can be used for modeling data which are not identical in distribution. Some properties of the theoretical model such as moment, mean deviation, entropy criteria, symmetry and unimodality are derived. The proposed model also studies the problem of parameter estimation and derives maximum likelihood estimators in a weighted gamma distribution. Finally, it will be shown that the proposed model is the best among the previously introduced distributions for modeling a real data set.

The estimation problem of expected time to failure of units is studied in a discrete set up. A simple step-stress accelerated life testing is considered with a Type-I censored sample from geometric distribution that is a commonly used distribution to model the lifetime of a device in discrete case. Maximum likelihood estimators as well as the associated distributions are derived. Exact, approximate and bootstrap approaches construct confidence intervals that are compared via a simulation study. Optimal confidence intervals are suggested in view of the expected width and coverage probability criteria. An illustrative example is also presented to explain the results of the paper. Finally, some conclusions are stated.
Nonparametric confidence intervals for quantiles based on a modified ranked set sampling

A new sampling method is introduced based on the idea of a ranked set sampling scheme in which taken samples in each set are dependent on previous ones. Some theoretical results are presented and distribution-free confidence intervals are derived for the quantiles of any continuous population. It is shown numerically that the proposed sampling scheme may lead to 95% confidence intervals (especially for extreme quantiles) that cannot be found based on the ordinary ranked set sampling scheme presented by Chen (2000) and Balakrishnan and Li (2006). Optimality aspects of this scheme are investigated for both coverage probability and minimum expected length criteria. A real data set is also used to illustrate the proposed procedure. Conclusions are eventually stated.

Background: Sinonasal malignancies are a rare group of head and neck cancers. We aimed to report the oncological outcomes based on histological types in patients who underwent radiotherapy.Materials and Methods: In this single-institution study, we retrospectively retrieved and analyzed data of patients with sinonasal carcinomas who underwent radiotherapy during 2011–2016 as part of their treatment. The 3-year rate of local, regional, and distant recurrences, and overall survival were evaluated according to the histological type.Results: A total of 28 patients were evaluated in this study, the majority of whom were male (60%). Squamous cell carcinoma (SCC), adenoid cystic carcinoma (ACC), and adenocarcinoma (ADC) were found in 15 patients (53.5%), 8 (28.5%), and 5 (18%), respectively. The highest rates of local and regional recurrences were observed in ACC and SCC, respectively. Distant recurrences were numerically more common in ADC. The 3-year OS was 48%, 50%, and 73% in SCC, ADC, and ACC, respectively.Conclusion: Different histopathologies of sinonasal cancer seem to have different patterns of failure, and this may be considered in the treatment approach.

Objective: Brain tumors cause great mortality and morbidity worldwide, and success rates with surgical treatment remain very low. Several recent studies have focused on introduction of novel effective medical therapeutic approaches. Genistein is a member of the isoflavonoid family which has proved to exert anticancer effects. Here we assessed the effects of genistein on the expression of MMP-2 and VEGF in low and high grade gliomas in vitro. Materials and Methods: High and low grade glioma tumor tissue samples were obtained from a total of 16 patients, washed with PBS, cut into small pieces, digested with collagenase type I and cultured in DMEM containing 10% FBS. When cells reached passage 3, they were exposed to genistein and MMP-2 and VEGF gene transcripts were determined by quantitative real time PCR (qRT-PCR). Results: Expression of MMP-2 demonstrated 580-fold reduction in expression in low grade glioma cells post treatment with genistein compared to untreated cells (P value= 0.05). In cells derived from high grade lesions, expression of MMP-2 was 2-fold lower than in controls (P value> 0.05). Genistein caused a 4.7-fold reduction in VEGF transcript in high grade glioma cells (P value> 0.05) but no effects were evident in low grade glioma cells. Conclusion. Based on the data of the present study, low grade glioma cells appear much more sensitive to genistein and this isoflavone might offer an appropriate therapeutic intervention in these patients. Further investigation of this possibility is clearly warranted.
