        Biocatalysis and Fermentation Technology : Thermostable Sites and Catalytic Characterization of Xylanase XYNB of Aspergillus niger SCTCC 400264

        In order to improve the expression of heat-resistant xylanase XYNB from Aspergillus niger SCTCC 400264, XynB has been cloned into Pichia pastoris secretary vector pPIC9K. The XynB production of recombinant P. pastoris was four times that of E. coli, and the Vmax and specific activity of XynB reached 2,547.7 umol/mg and 4,757 U/mg, respectively. XynB still had 74% residual enzyme activity after 30 min of heat treatment at 80°C. From the van der Waals force analysis of XYNB (ACN89393 and AAS67299), there is one more oxygen radical in AAS67299 in their catalytic site, indicating that the local cavity is much more free, and it is more optimal for substrate binding, affinity reaction, and proton transfer, etc, and e ventually i ncreasing enzyme activity. The H-bonds analysis of XYNB indicated that there are two more H-bonds in the 33rd Ser of XYNB (AAS67299) than in the 33rd Ala(ACN89393 ), and two H-bonds between Ser70 and Asp67.

        Magnetostrictive and Magnetic Properties of Tb<SUB>0.29</SUB>Dy<SUB>0.48</SUB>Ho<SUB>0.23</SUB>Fe<SUB>1.9</SUB> Fiber/Epoxy Composites

        In this paper, we fabricated novel magnetostrictive composites by embedding <110>-oriented Tb-Dy-Ho-Fe fibers in an epoxy matrix. The magnetostrictive and magnetic properties (magnetostriction, magnetization, piezomagnetic coefficient and relative permeability) of the proposed composites were measured, analyzed, and compared to those of Tb0.29Dy0.48Ho0.23Fe1.9 alloy and Tb0.3Dy0.7Fe₂ continuous-fiber/epoxy composites. Tb0.29Dy0.48Ho0.23Fe1.9 continuous-fiber/epoxy composites have a saturation magnetostriction (λs) of 840 ppm and saturation magnetization (Ms) of 0.75 T. Their piezomagnetic coefficient exhibits a maximum value (8.2 μm/kA) at 19 kA/m. These proposed composites exhibit a large magnetostriction in high magnetic fields (> 400 kA/m) and a large relative permeability in low magnetic fields (< 100 kA/m). This result indicates that the given composites perform better than the Tb0.29Dy0.48Ho0.23Fe1.9 alloy and Tb0.3Dy0.7Fe₂ fiber/epoxy composites. Thus, the composites with characteristics of high sensitivity and large magnetostriction can be used in the field of ultrasonic sensing.

        Mixtures of Tea and Citrus maxima (pomelo) Alleviate Lipid Deposition in HepG2 Cells Through the AMPK/ACC Signaling Pathway

        Tea and citrus maxima are natural, medicinal homologous plants, typically used for making beverages, which have anticancer, antiobesity, and antioxidation properties. Green tea, yellow tea, and black tea were combined with citrus maxima to obtain green tea and Citrus maxima (GTCM), yellow tea and Citrus maxima (YTCM), and black tea and Citrus maxima (BTCM). The biochemical components of these mixtures were analyzed, and their possible effects and mechanisms on relieving liver lipid deposition were explored. The tea polyphenols, free amino acids, phenolamine ratio, and caffeine were comparable in YTCM and GTCM, being significantly higher than those in BTCM. In addition, the content of esterified catechins, nonesterified catechins, and total catechins in YTCM was significantly higher than those in GTCM and BTCM. All three mixtures of Citrus maxima tea significantly reduced lipid deposition in HepG2 cells, with GTCM and YTCM being slightly more effective than BTCM. Regarding the possible mechanism, Western blot analysis revealed that the three Citrus maxima tea mixtures could activate the AMPK/ACC signaling pathway, upregulate the expression of p-AMPK, p-ACC, and CPT-1 proteins, and downregulate the expression of SREBP1c and fatty acid synthase proteins to inhibit fat synthesis, thereby relieving lipid deposition in liver cells. In conclusion, as a novel and healthy beverage, Citrus maxima tea has the potential to alleviate liver lipid deposition, and further could be responsible for obesity treatment.

      • Hole-filling Based on Disparity Map and Inpainting for Depth-Image-Based Rendering

        Due to the changes in viewpoint, holes may appear in the novel view synthesized by depth-image-based rendering (DIBR). A hole-filling method combining disparity-map-based hole-filling and inpainting is proposed. The method first eliminates matching errors in destination image according to the disparity map. Then, holes are classified into different types according to their sizes. Finally, a disparity-map-based approach and an improved exemplar-based inpainting algorithm are used to fill different types of holes according to the type of hole. Experimental results show that the artifacts at the edge of foreground objects can be reduced in synthesized view since the matching errors are eliminated before hole-filling. In addition, the proposed method can achieve more natural and satisfactory results in filled areas in comparison with the disparity-map-based approach and Gautier’s inpainting algorithm, though it may result in higher time complexity.

        Sensitive and Extraction-Free Detection of Methicillin-Resistant Staphylococcus aureus through Ag + Aptamer-Based Color Reaction

        Refractory infections, such as hospital-acquired pneumonia, can be better diagnosed with the assistance of precise methicillin-resistant Staphylococcus aureus (MRSA) testing. However, traditional methods necessitate high-tech tools, rigorous temperature cycling, and the extraction of genetic material from MRSA cells. Herein, we propose a sensitive, specific, and extraction-free strategy for MRSA detection by integrating allosteric probe-based target recognition and exonuclease-III (ExoIII)-enhanced color reaction. The penicillin-binding protein 2a (PBP2a) aptamer in the allosteric probe binds with MRSA to convert protein signals to nucleic acid signals. This is followed by the DNA polymerase-assisted target recycle and the production of numerous single-strand DNA (ssDNA) chains which bind with silver ion (Ag+ ) aptamer to form a blunt terminus that can be identified by Exo-III. As a result, the Ag+ aptamer pre-coupled to magnetic nanoparticles is digested. After magnetic separation, the Ag+ in liquid supernatant catalyzes 3,3’,5,5’-tetramethylbenzidine (TMB) for a color reaction. In addition, a concentration of 54 cfu/mL is predicted to be the lowest detectable value. Based on this, our assay has a wide linear detection range, covering 5 orders of magnitude and demonstrating a high specificity, which allows it to accurately distinguish the target MRSA from other microorganisms.

        Synthesis and Characterization of Poly(lactic acid-co-glycolic acid-co-ε-caprolactone) and Its Pressure Sensitive Adhesive Property

        Random copolymers of poly(lactic acid-co-glycolic acid-co-ε-caprolactone) (PLGA-CL) based on lactic acid (LA), glycolic acid (GA), citric acid (CA), and ε-caprolactone (CL) were synthesized and characterized. ¹H NMR spectra showed that the polymerization conversion of various monomers was high under the experimental conditions, and the degree of polymerization of the polycaprolactone (PCL) segment increased with the growth of CL content. The content of CL has a remarkable effect on the decrease of glass transition temperature (Tg) of the copolymers. The adhesive properties of random copolymers as pressure-sensitive adhesive (PSA) were evaluated. With the increase of CL content, 180° peel strength, the shear strength and loop tack decreased. When CL content was 16.7 wt%, 180° peel strength was 4.76 N/25 ㎜, and loop tack was 8.45 N/25 ㎜.

        Huber Second-order Variable Structure Predictive Filter for Satellites Attitude Estimation

        This work presents a novel filtering approach to the high-accuracy attitude estimation problem of satellites. A new second-order variable structure predictive filter is first designed with the measurement errors and theirdifference reduced. The key feature of this filter is that the noise handled is not constrained to be the Gaussianwhite noise. Hence, it is a new solution to filtering problem in the presence of modeling errors or heavy-tailednoise. Then, the robust version of the preceding filter is developed by using the Huber technique. This robustfilter can ensure great robustness and perfect estimation accuracy/precision for the satellite attitude. The Lyapunovstability analysis proves that the measurement error and its difference can be stabilized into a small set with a fasterrate of convergence. The effectiveness of the presented attitude estimation filters is validated via simulation bycomparing with the traditional cubature Kalman filter.

      • Precoding Method for Broadband PLC MIMO System of Eliminating Interference

        Power line carrier communication, with the widespread application, broadband multi-input multi-output system will have a co-channel interference, the interference is in addition to asynchronous pulse noise and pulse cycle factors influencing the multi-input multi-output (MIMO) system's largest broadband power line carrier. Aiming at this problem, this paper, by using multiple input multiple output channel state information, the sender with the method of block diagonalization, eliminate the interference between the channel, so as to improve the capacity of broadband power line carrier system, at the same time reduce the bit error rate of the system. Through the simulation results and analysis can show that the proposed block diagonalization pre-coding method effectively eliminates the common channel interference, improve the drying ratio, thus improve the maximum mutual information of the system and reduce the bit error performance of the system.

        Analysis of thermal and wet stress of corn kernel based on microwave drying

        In order to reveal the relationship between temperature, moisture content and stress during microwave drying, the paper establishes a mathematical model of heat and mass transfer as well as stress during microwave drying using corn kernels as the medium, and couples the two models to solve the electromagnetic equation, energy and momentum conservation and stress equation, and conducts experimental verification. The results show that the maximum errors between the simulated and experimental values of temperature and moisture content are 8.04 % and 9.21 %, respectively, and the theoretical model can be used to simulate the changes of temperature, moisture content and stress of corn kernels. Under the set drying conditions, the thermal stress increased with increasing temperature, and the stress maximum was obtained on the upper and lower surfaces of the seeds. The wet stress increased with increasing moisture gradient, and the stress and crack index were maximum at a moisture content (dry basis) of 16.13 %.
