        Effect of Panax ginseng and Fructus Mume on Intestinal Barrier and Gut Microbiota in Rats with Diarrhea

        Panax ginseng and Fructus mume (Renshen Wumei in Chinese, RW) are natural medicines with high nutritional and pharmacological value. They have been widely used together in China to treat gastrointestinal diseases, especially persistent diarrhea, but the potential mechanisms remain elusive. In this study, a diarrhea model was established in rats using a 30% aqueous extract of senna. The therapeutic effects of RW were evaluated by recording the prevalence of loose stools, the diarrhea index, and histopathological changes in colon tissue. The levels of mucins, tight junction (TJ) proteins, inflammatory cytokines, and phosphoinositide 3-kinase/Akt/nuclear factor-κB (PI3K/Akt/NF-κB) signaling pathway proteins were measured. Metagenomic sequencing was used to analyze the gut microbiota. Treatment with RW alleviated injury to the intestinal barrier in rats with diarrhea and also upregulated levels of Muc2 and TJ proteins, such as occludin, zonula occludens-1, and claudin-1. Administration of RW regulated the structure of the gut microbiota in diarrheal rats. Furthermore, RW suppressed levels of interleukin (IL), tumor necrosis factor (TNF)-α, IL-1, PI3K, Akt, and p-NF-κB p65 and also increased IL-4 levels. Our study indicates that P. ginseng and Fructus mume help improve the symptoms of diarrhea, possibly by alleviating the intestinal barrier injury, regulating intestinal flora composition, and inhibiting the PI3K/Akt/NF-κB signaling pathway.

        KLF9 promotes autophagy and apoptosis in T-cell acute lymphoblastic leukemia cells by inhibiting AKT/mTOR signaling pathway

        Background T -cell acute lymphoblastic leukemia (T-ALL) is considered a malignant tumor with a high mortality rate. To combat this disease, exploring the mechanism of T-ALL progression is urgently needed. Krüppel-like factors (KLFs) are known as the transcription factors and mediate series of biological processes. KLF9 is a member of the KLF family which could serve as a tumor suppressor gene in most solid tumors. GEO Database analysis showed that KLF9 expression in normal T cells was higher than T-ALL cell lines and patients. However, the possible role of KLF9 in T-ALL progression is still unclear. Objective To uncover the possible eff ects of Krüppel-like transcription factor 9 (KLF9) on the progression of T-Acute lymphoblastic leukemia (T-ALL). Results The expression of KLF9 was low in human T-ALL cells. KLF9 suppressed the viability of T-ALL cells. In addition, KLF9 stimulated the apoptosis as well as autophagy of T-ALL cells. Mechanically, KLF9 suppressed AKT/mTOR pathway in T-ALL cells. Conclusion KLF9 suppressed viability and promoted autophagy as well as apoptosis in T-ALL cells by inhibiting AKT/ mTOR pathway.

        Extraction of rare earths using mixtures of sec-octylphenoxy acetic acid and organophosphorus acids

        The extraction of rare earths from nitrate medium using three organophosphorus acids, 2-ethylhexyl phosphonic acid mono-2-ethylhexyl ester (HEHEHP), di-(2-ethylhexyl) phosphoric acid (D2EHPA), bis(2,4,4-trimethylpentyl)phosphinic acid (Cyanex272), and their mixtures with sec-octylphenoxy acetic acid (CA12) has been studied in detail. The mixtures have different extraction effects on various rare earths. Synergistic extraction effects are only found when light rare earths and yttrium (III) are extracted with mixtures of D2EHPA and CA12. The possibilities of separating the rare earths with these mixtures are investigated according to the extractabilities. It is feasible and advantageous to separate yttrium (III) from the lanthanoids (III) with HEHEHP + CA12 and D2EHPA+CA12 mixtures at proper extractant ratios. The separation of yttrium (III) from heavy rare earths is also possible with mixtures of Cyanex272 and CA12.

      • Human Telomerase Gene and High-Risk Human Papillomavirus Infection are Related to Cervical Intraepithelial Neoplasia

        Our aims were to evaluate the clinical performance of human telomerase RNA gene component (hTERC gene) amplification assay with high-risk human papillomavirus (HR-HPV) DNA test of Hybrid Capture 2 DNA test (HC2), for the detection of high grade cervical precancerous lesions and cancer (CIN 2+). In addition, the association shown between hTERC gene amplification and HPV DNA test positive in women with and without cervical neoplasia was assessed. There were 92 women who underwent cytology, HR-HPV DNA test, hTERC gene amplification test, colposcopy and biopsy. We compared the clinical performance of hTERC gene test along with HR-HPV DNA test of women with colposcopy and routine screening. The samples were histology-confirmed high-grade cervical intraepithelial neoplasia (CIN 2) or worse (CIN2+) as the positive criterion. The test of hTERC gene showed the hTERC gene amplification positivity increased with the severity of histological abnormality and cytological abnormality. The test of hTERC gene showed higher specificity than HR-HPV DNA test for high-grade lesions (84.4% versus 50%) and also higher positive predictive value (90.4% versus 76.5%). Our results predicted that hTERC gene amplification demonstrated more specific performance for predicting the risk of progression and offer a strong potential as a tool for triage in cervical cancer screening, with the limited sensitive as HR-HPV DNA test.

        Adsorption of zinc (Ⅱ), cadmium (Ⅱ), and copper (Ⅱ) with PTFE Selective Resin containing primary amine N1923 and Cyanex925

        In the present study, a synergistic extraction mixture, primary amine N1923 (RNH2) and Cyanex925 (B), was loaded on PTFE powder to prepare PTFE Selective Resin and was employed for the adsorption of zinc, cadmium, and copper from chloride medium. The effects of shaking time, resin amounts, concentration of metal ions, and experimental temperature on the distribution coefficients were determined. The stoichiometry of the adsorbed complexes was determined to be MCl2·(1/2)RNH2·B with the Selective Resin. The equilibrium constants and thermodynamic quantities (ΔH, ΔG, and ΔS) were calculated. Furthermore, the Freundlich isotherm proved to be more suitable than the Langmuir isotherm to indicate the effect of concentration of M2+ on the adsorption with PTFE Selective Resin.

      • Matrix Metalloproteinase-9 as a Prognostic Factor in Gastric Cancer: A Meta-Analysis

        Background: Matrix metalloproteinase-9 (MMP-9) is associated with disruption of basement membranes of blood vessels and promotion of metastasis through the lymphatics. However, its prognostic value for survival in patients with gastric cancer remains controversial. Method: We therefore conducted a meta-analysis of the published literature in order to clarify the impact of MMP-9. Clinical studies were selected for further analysis if they provided an independent assessment of MMP-9 in gastric cancer and reported analysis of survival data according to MMP-9 expression. Results: A total of 11 studies, covering 1700 patients, were included for meta-analysis. A summary hazard ratio (HR) of all studies and sub-group hazard ratios were calculated. The combined HR suggested that a positive MMP-9 expression had an impact on overall survival: 1.25 (95% confidence interval 1.11-1.40) in all eligible studies; 1.13 (1.06-1.20) in 8 studies detecting MMP-9 by immunohistochemistry; 1.36 (1.12-1.65) in 7 studies from Asia. Only one study for DFS showed a significant impact on disease free survival (HR 1.73, 95%CI 1.27-2.34). Conclusions: Our findings suggested that MMP-9 protein expression might be a factor for a poor prognosis in patients with gastric cancer. However, the association was rather weak, so that more prospective studies should further explore the prognostic impact of MMP-9 mRNA and correlations between MMP-9 and clinicopathological characteristics.

      • Indoor Location Algorithm Based on Kalman Filter and Multi-Source Data Integration

        For onboard single-station passive direction-finding and location, if there is any abnormal error in the observation data, the extended Kalman filter (EKF) algorithm adopted thereby will cause inaccurate location result. In order to improve algorithm robustness, the robust equivalent gain matrix is constructed according to the standardized prediction residual error and the robust EKF algorithm is applied to the onboard single-station passive direction-finding and location. In allusion to the low efficiency of the robust EKF algorithm, the single-station passive location algorithm based on the improved extended Kalman filter is proposed in this article on the basis of combining F distribution statistic, and meanwhile single abnormal error and continuous abnormal error are added in the observation value to test the algorithm resistance to different abnormal errors. The simulation shows that the algorithm proposed in this article can well weaken the influence of abnormal errors on position estimation and the algorithm based on F distribution discriminant can improve location efficiency.

        Energy-Efficient Base Station Sleep Scheduling in Relay-Assisted Cellular Networks

        We Relay-assisted cellular network architecture has been developed to cover cell-edge users and to improve capacity. However, the deployment of relay stations (RSs) in cellular networks may increase the total energy consumption. Though energy efficiency has become a major concern in cellular networks, little work has studied the energy efficiency of relay-assisted cellular networks by sleep scheduling. In this paper, a distributed base stations (BSs) sleep scheduling scheme in relay-assisted cellular networks is proposed. The goal is to maximize the energy efficiency under the spectral efficiency constraint. Firstly, whether the BSs should be sleeping or active is determined by the traffic profile. Then, the transmission powers of the active BSs are optimized within the game-theoretic framework, by using an interior-point method, so as to maximize the energy efficiency. Simulation results demonstrate that the effectiveness of the proposed scheme is superior to that turning on all the BSs without sleep scheduling.

        Modified Renshen Wumei Decoction Alleviates Intestinal Barrier Destruction in Rats with Diarrhea

        Modified Renshen Wumei decoction (MRWD), a famous traditional Chinese medicine, is widely used for treating persistent diarrhea. However, as the mechanism by which MRWD regulates diarrhea remains unknown, we examined the protective effects of MRWD on intestinal barrier integrity in a diarrhea model. In total, 48 male rats were randomly distributed to four treatment groups: the blank group (CK group), model group (MC group), Medilac-Vita group (MV group) and Chinese herb group (MRWD group). After a 21-day experiment, serum and colon samples were assessed. The diarrhea index, pathological examination findings and change in D-lactate and diamine oxidase (DAO) contents illustrated that the induction of diarrhea caused intestinal injury, which was ameliorated by MV and MRWD infusion. Metabolomics analysis identified several metabolites in the serum. Some critical metabolites, such as phosphoric acid, taurine, cortisone, leukotriene B4 and calcitriol, were found to be significantly elevated by MRWD infusion. Importantly, these differences correlated with mineral absorption and metabolism and peroxisome proliferator-activated receptor (PPAR) pathways. Moreover, it significantly increased the expression levels of TLR4, MyD88 and p-NF-κB p65 proteins and the contents of IL-1 and TNF-α, while the expression levels of occludin, claudin-1 and ZO-1 proteins decreased. These deleterious effects were significantly alleviated by MV and MRWD infusion. Our findings indicate that MRWD infusion helps alleviate diarrhea, possibly by maintaining electrolyte homeostasis, improving the intestinal barrier integrity, and inhibiting the TLR4/NF-κB axis.
