      • Expression and Prognostic Implications of FOXO3a and Ki67 in Lung Adenocarcinomas

        To investigate the significance of FOXO3a and Ki67 in human lung adenocarcinomas. Envision immunohistochemical staining and Western blotting were used to examine the protein expression of FOXO3a in 127 cases of human lung adenocarcinoma specimens. The positive rate in lung adenocarcinoma (55.9%) was lower than that in normal tissues (80%). We found that the expression of FOXO3a was closely related with the degree of differentiation, TNM staging, lymph node metastasis and survival. In addition, significant differences in the different pathological types of lung adenocarcinoma cases (P<0.01). The FOXO3a positive rate of the acini as the main type (APA) (86.7%) and the lepidic as the main type (LPA) (82.4%) was higher than the solid as the main type (SPA) (50.0%), the papilla as the main type (PPA) (42.9%) and the micropapilla as the main type (MPA) (9.4%). Moreover, the expression of FOXO3a was negatively related with Ki67 expression. Our results suggested that the expression of FOXO3a is closely correlated with the aggressiveness of lung adenocarcinoma. It was indicated that disregulation of FOXO3a might play key roles in the occurrence and development of lung a denocarcinoma and joint detection of the two markers might play an important role in diagnosing tumors.

        Cellulose nanofibrils composite hydrogel with polydopamine@zeolitic imidazolate framework-8 encapsulated in used as efficient vehicles for controlled drug release

        Herein, we report a nanocellulose hydrogel that has a packaging structure for on-demand drug release. Zeolitic imidazolate frameworks-8 (ZIF-8) were grown on the surface of polydopamine (PDA) to producePDA@ZIF-8 nanoparticles. The obtained PDA@ZIF-8 nanoparticles were then incorporated into a networkof cellulose nanofibrils (CNFs) to fabricate a PDA@ZIF-8/CNFs composite hydrogel. The loading ratio of thedrug tetracycline hydrochloride (TH) was 58%. A slight burst release behavior was observed at the beginning,and the composite hydrogel presented a significant pH-dependent release behavior. The drug deliverytime of the PDA@ZIF-8/CNFs composite hydrogel was 3.5 times longer than that of the PDA/CNFscomposite hydrogel. Furthermore, NIR light radiation accelerated the drug delivery rate. The drug releasemechanism was mainly driven by anomalous transport. Importantly, the novel PDA@ZIF-8/CNFs compositehydrogel was found to be biocompatible according to in vitro cytotoxicity test. Because of its uniquemultiple drug release responses, the PDA@ZIF-8/CNFs composite hydrogel may be used in future clinicalapplications as a promising drug delivery carrier.

      • A management strategy for solar panel - battery - super capacitor hybrid energy system in solar car

        This paper presents an application of solar energy - battery - super-capacitor hybrid energy storage system in solar electric vehicles. The key point is the proposed energy management control algorithm. The entire system consists of a solar panel, a boost converter, a battery, a super capacitor, a bi-directional DC/DC converter, and a brushless DC motor. This paper gives the details of the control strategy to cooperate the three energy sources or storage devices: solar panel, battery and super capacitor. A hysteretic control algorithm for battery charging by solar panel is given. A novel control method for bi-directional DC/DC is proposed to keep the battery discharging current within a certain limit and make full use of super capacitor. The switching transient between super-capacitor charging mode and discharging mode is also presented. Both simulation and experimental results demonstrate the effectiveness of the proposed strategy.

      • Dietary Factors and Risk of Pancreatic Cancer: a Multi-Centre Case-Control Study in China

        Background: Pancreatic cancer is the sixth leading cause of cancer death with an increasing trend in China. Dietary intake is believed to play an important role in pancreatic cancer carcinogenesis. The aim of this paper was to evaluate associations between some dietary factors and risk of pancreatic cancer in a multi-centre case-control study conducted in China. Materials and Methods: Cases (n=323) were ascertained from four provincial cancer hospitals. Controls (n=323) were randomly selected from the family members of patients without pancreatic cancer in the same hospitals, 1:1 matched to cases by gender, age and study center. Data were collected with a questionnaire by personal interview. Odds ratios (OR) and 95% confidence intervals (95%CI) were estimated using conditional logistic regression. Results: Tea intake (OR =0.49; 95%CI: 0.30-0.80) was associated with a half reduction in risk of pancreatic cancer. Reduced vegetable consumption (P trend: 0.04) was significant related to pancreatic cancer. Although no significant association was found for meat and fruit, ORs were all above or below the reference group. A protective effect was found for fruit (OR=1.73 for consumption of 1-2 times/week vs more than 3 times/week; 95%CI: 1.05-2.86). A high intake of meat was associated to a higher risk of pancreatic cancer (OR=0.59 for consumption of 1-2 times/week vs. more than 3 times/week; 95%CI: 0.35-0.97). Conclusions: The present study supports fruit consumption to reduce pancreatic cancer risk and indicates that high consumption of meat is related to an elevated risk. Direct inverse relations with tea and vegetable intake were also confirmed.

        Fast Transient Buck Converter Using a Hysteresis PWM Controller

        In this paper, a fast transient buck converter using hysteresis PWM control is presented. The proposed control method is based on hysteresis control of the capacitor C voltage. This offers a faster transient response to meet the challenges of the power supply requirements for fast dynamic input and load changes. It also provides better stability and solves the compensation problem of the error amplifier in conversional voltage PWM control. Finally, the steady-state and dynamic operation of the proposed control method are analyzed and verified by simulation and experimental results.

      • Ablation of HRC alleviates cardiac arrhythmia and improves abnormal Ca handling in CASQ2 knockout mice prone to CPVT

        <P><B>Aims</B></P><P>Cardiac calsequestrin (CASQ2) and histidine-rich Ca-binding protein (HRC) are sarcoplasmic reticulum (SR) Ca-binding proteins that regulate SR Ca release in mammalian heart. Deletion of either CASQ2 or HRC results in relatively mild phenotypes characterized by preserved cardiac structure and function, although CASQ2 knockout (KO), or Cnull, shows increased arrhythmia burden under conditions of catecholaminergic stress. We hypothesized that given the apparent overlap of functions of CASQ2 and HRC, simultaneous ablation of both would deteriorate the cardiac phenotype compared with the single knockouts.</P><P><B>Methods and results</B></P><P>In contrast to this expectation, double knockout (DKO) mice lacking both CASQ2 and HRC exhibited normal cardiac ejection fraction and ultrastructure. Moreover, the predisposition to catecholamine-dependent arrhythmia that characterizes the Cnull phenotype was alleviated in the DKO mice. At the myocyte level, DKO mice displayed Ca transients of normal amplitude; additionally, the frequency of spontaneous Ca waves and sparks in the presence of isoproterenol were decreased markedly compared with Cnull. Furthermore, restitution of SR Ca release was slowed in DKO myocytes compared with Cnull cells.</P><P><B>Conclusion</B></P><P>Our results suggest that rather than being functionally redundant, CASQ2 and HRC modulate cardiac ryanodine receptor-mediated (RyR2) Ca release in an opposing manner. In particular, while CASQ2 stabilizes RyR2 rendering it refractory in the diastolic phase, HRC enhances RyR2 activity facilitating RyR2 recovery from refractoriness.</P>

        Insulin receptors regulate the fecundity of Nilaparvata lugens (Stål) (Hemiptera: Delphacidae)

        Two InR (insulin receptor) genes have been identified in the Nilaparvata lugens. In this study, we used RNA interference (RNAi) to investigate the role of InR genes in the fecundity of N. lugens. The expression of NLInR1 and NLInR2 genes was simultaneously silenced with mixture of dsInR1 and dsInR2 (dsInRs) injection. Our results showed that larvae RNAi against both NLInR1 and NLInR2 reduced the number of eggs laid by N. lugens and some eggs as well as ovaries were abnormal. In addition, the relative expression of Vg (vitellogenin) and VgR (vitellogenin receptor) was significantly reduced on the 4th and 6th days after insects treated with larvae RNAi reached the adult stage. We also determined the relative expression levels of insulin/insulin-like signaling (IIS) related genes in RNAi-treated larvae and found that the expression levels of Chico (homologous receptor substrate), Akt (protein kinase B), PI3K (phosphoinositide 3-kinase), and PTEN (phosphatase and tensin homolog) genes decreased whereas FOXO (forkhead box O) and GSK3 (glycogen synthase kinase-3) levels increased on the 4th and 6th days after insects reached the adult stage. These results indicate that silencing of NLInR1 and NLInR2 reduces the fecundity of N. lugens through the IIS pathway.

        Two type III polyketide synthases from Polygonum cuspidatum: gene structure, evolutionary route and metabolites

        In our recent work (Ma et al., in Planta229(3):457–469, 2009a and 229(4):1077–1086, 2009b),two three-intron type III PKS genes, PcPKS1 and PcPKS2,were isolated from Polygonum cuspidatum Sieb. et Zucc. Phylogenetic and functional analyses revealed PcPKS1 is athree-intron chalcone synthase (CHS) gene, and PcPKS2 isfound to be a three-intron benzalacetone synthase (BAS)gene. The regular CHS encoded by a single intron genehave not been isolated and characterized from P. cuspidatum. In this work a further CHS with one intron (PcPKS3)and a stilbene synthase (STS) gene with three-intron(PcPKS5) were isolated and characterized by functionaland phylogenetic analyses. In comparison with PcPKS1, abifunctional enzyme with both CHS and BAS activity, theenzymatic product of recombinant PcPKS3 was naringenin,bis-noryangonin (BNY) and 4-coumaroyltriacetic acidlactone (CTAL) occurred as side products. The PcPKS5synthesized resveratrol and a trace amount of naringeninfrom p-coumaroyl-CoA. To our knowledge, PcPKS5 is thefirst reported three-intron STS gene in flowering plants. Inthis work, we speculated that this involved a possibleevolutionary route of plant-specific type III PKS superfamilyin P. cuspidatum.
