        유산양 유즙으로부터 분리된 세균의 분포 및 항균제 감수성 검사

        The aim of this study was to isolate microorganisms from half milk samples of dairy goats by California mastitis test (CMT) during the lactation period and to further investigate the susceptibility of isolated organisms to antimicrobial drugs. From a total of 235 half milk samples with CMT scores of 2 or above from 366 dairy goats distributed throughout Jeonnam province, microorganisms were isolated from 198 (83.5%) samples either singly (99.0%) or in combination (1.0%). The most prevalent microorganism was the coagulase-negative Staphylococcus spp., (44.4%, n=88) followed by Staphylococcus aureus (24.2%, n=48), Escherichia coli (11.1%, n=22) and Streptococcus spp. (7.6%, n=15). Isolated bacteria also included Bacillus spp. (2.5%, n=5), Pseudomonas spp. (2.5%, n=5), Micrococcus spp. (1.5%, n=3), Corynebacterium spp. (1.5%, n=3), Enterococcus facium (1.0%, n=2), Morganella morganii (0.5%, n=1) and Streptococcus agalactiae (0.5%, n=1). During the summer season, a high prevalence of all micro-organisms were observed in which Staphylococcus spp. (30.8%), Escherichia coli (8.6%), and Streptococcus spp. (5.6%) were among the most prevalent bacteria isolated. Staphylococcus spp. was also shown to be high in the winter (21.7%). In most samples, the presence of bacterial pathogens in goat milk led to the increase in the total somatic cell count (SCC). Most of the half milk samples of dairy goats with bacterial contamination showed SCC of ≥1×10(6) cells/ml (90.4%). Minor pathogens (11.4%) were more detected from milk samples with SCC of <1×10(6) cells/ml than major pathogens (4.1%), while the major pathogens tended to be higher from samples with SCC of ≥3×10(6) cells/ml. Susceptibility of these bacteria to 12 antimicrobial agents was tested by the Kirby-Bauer disc diffusion method. Results indicated that more than 90% of bacteria isolated from CMT 2+ dairy goat half milk samples were susceptible to trimethoprim/sulfamethoxazole, amoxicillin/clavulanic, enrofloxacin and cephalothin while they were resistant to tetracycline (44.7%).

        Increased Fat Due to Estrogen Deficiency Induces Bone Loss by Elevating Monocyte Chemoattractant Protein-1 (MCP-1) Production

        Ovariectomy (OVX)-induced estrogen withdrawal resulted in both bone loss and an increase in fat. We observed elevated osteoclast (OC) formation by bone marrow-derived macrophages treated with medium conditioned by fats from OVX mice, but not from sham-operated mice. Fats from OVX mice expressed and secreted higher levels of monocyte chemoattractant protein-1 (MCP-1) than those from sham-operated mice. Increased fat resulting from estrogen deficiency is thus responsible for bone loss due to enhanced OC formation, which is, at least partly, a consequence of elevated MCP-1 production.

        Tetrahydropteridines possess antioxidant roles to guard against glucose-induced oxidative stress in Dictyostelium discoideum

        Glucose effects on the vegetative growth of Dictyostelium discoideum Ax2 were studied by examining oxidative stress and tetrahydropteridine synthesis in cells cultured with different concentrations (0.5X, 7.7 g L-1; 1X, 15.4 g L-1; 2X, 30.8 g L-1) of glucose. The growth rate was optimal in 1X cells (cells grown in 1X glucose) but was impaired drastically in 2X cells, below the level of 0.5X cells. There were glucose-dependent increases in reactive oxygen species (ROS) levels and mitochondrial dysfunction in parallel with the mRNA copy numbers of the enzymes catalyzing tetrahydropteridine synthesis and regeneration. On the other hand, both the specific activities of the enzymes and tetrahydropteridine levels in 2X cells were lower than those in 1X cells, but were higher than those in 0.5X cells. Given the antioxidant function of tetrahydropteridines and both the beneficial and harmful effects of ROS, the results suggest glucose-induced oxidative stress in Dictyostelium, a process that might originate from aerobic glycolysis, as well as a protective role of tetrahydropteridines against this stress.         EPR SPECTRA OF Mn ION WITH TWO PHASES IN THE Y - Ba - Cu - Mn - O HIGH Tc SUPERCONDUCTOR

        In this paper, Mn²+ ion was doped in Y-Ba-Cu-O as an EPR probe. The following samples were prepared by conventional solid-state reaction method: YB_(a2)Cu_(2.96)Mn_(0.04)O_(7-δ) (MN-Ⅰ), annealed YB_(a2)Cu_(2.96)Mn_(0.04)O_(7-δ) (AMN) and YB_(a2)Cu_(2.94)Mn_(0.06)O_(7-δ) (MN-Ⅱ). AMN sample was obtained from MN-Ⅰ by annealing for 1 hr under the Ar gas atmosphere at 600℃. X-band (~9.05 ㎓) EPR spectra were measured from 103 K to room temperature by employing a JES-RE3X spectroscopy with a TE_(011) cylindrical cavity and 100 ㎑ modulation frequency.<br/> In MN-Ⅰ we have observed only the Cu²+ signal. The fact that no Mn²+ signal was observed, in spite of Mn²+ being a very sensitive EPR probe, indicates that most likely isolated Mn²+ ions don't exist in the MN-Ⅰ sample. Most probably Mn²+ ions in the MN-Ⅰ sample interact antiferromagnetically and hence are EPR silent. The AMN spectra of at room temperature and 103 K indicate not only the Cu²+ signal but also an extra signal, which increases with decreasing temperature. It is suggested that the extra signal originates from Mn ions that were antiferromagnetically coupled before the annealing process. In MN - Ⅱ, from 103 K to room temperature, also, the extra signal was observed together with the Cu²+ signal. The extra signal in MN-Ⅱ, however, decreases with decreasing temperature and nearly disappears at 103 K. The signal originates from Mn ions in impurity phases that include Mn²+ ions. We suppose that there exist at least two Mn²+ doped phases in Y-Ba-Cu-O. The Mn²+ signal of one phase is undectable at all temperature and that of another phase decreases with decreasing temperature and disappears around 103 K.

        Time-dependently expressed markers and the characterization for premature senescence induced by ionizing radiation in MCF7

        <P>Senescence has been suggested as a defense mechanism to block sporadic induction of cancer cells. Radiation treatment induces proliferating cancer cells to turn into non-proliferating senescent cells in vitro. To characterize transcriptional reprogramming after radiation treatment, we measured the gene expression profiles of MCF7 at different time points after treatment. In these experiments, we found that IR induced premature senescence in MCF7 cells, and IR treatment resulted in significant changes in the expression of 305 marker genes (<1% FDR), which were strongly correlated (|r|>0.9) with IR treatment in a time-dependent manner. Functional analysis of these markers indicated that the dynamics of cytoskeletal structure and lysosomal activity gradually increased. The expression of maker genes for modulator proteins, that were responsible for the dynamics of actin stress fibers and focal adhesion, displayed a particularly strong positive correlation with senescence-associated (SA) morphological changes through time. We also observed a strong induction of genes related to lysosomal metabolic activity, which was accompanied by an increase in the number of SA-beta-Gal positive cells. However, the expression of genes for cell cycle progression, post-transcription and translation activities gradually decreased after radiation treatment. Especially, we observed clear cell cycle arrest specifically at the S and G2/M phases with consistent down-regulation of genes for microtubule assembly/disassembly or spindle biogenesis.</P>

        Structure and dye-sensitized solar cell application of TiO<sub>2</sub> nanotube arrays fabricated by the anodic oxidation method

        <P>Well-ordered TiO<SUB>2</SUB> nanotube arrays were fabricated by the potentiostatic anodic oxidation method using pure Ti foil as a working electrode and ethylene glycol solution as an electrolyte with the small addition of NH<SUB>4</SUB>F and H<SUB>2</SUB>O. The influence of anodization temperature and time on the morphology and formation of TiO<SUB>2</SUB> nanotube arrays was examined. The TiO<SUB>2</SUB> nanotube arrays were applied as a photoelectrode to dye-sensitized solar cells. Regardless of anodizing temperature and time, the average diameter and wall thickness of TiO<SUB>2</SUB> nanotube arrays show a similar value, whereas the length increases with decreasing reaction temperature. The conversion efficiency is very low, which is due to a morphology breaking of the TiO<SUB>2</SUB> nanotube arrays in the manufacturing process of a photoelectrode.</P>

        x선 및 뫼스바우어 측정법을 사용하여 혼성 결정계열 Co(Cu_0.5Fe_0.5)_1-xCr_2S_4를 연구해본 결과. 결정구조는 입방스피넬구조를 갖고 있으며 격자상수는 Vegard의 법칙을 만족하고 있었다. 이성질체이동치로부터 철이온의 전하상태가 +3가 상태임을 알았다. 또한 전기사중극자 분열의 소멸로부터 철이온이 사면체자리를 차지하고 있음을 알았다. 특기할점은 80K에서 측정한 모든 뫼스바우어양들의 구성성분의 변화에 대해서 불변이라는 것이다. 이현상은 철이온 주변원자의 특이한 배치상태를 토대로 하여 설명할 수가 있다. Neel온도는 Co농도증가와 더불어 급속히 감소하였는데 이는 Co^2+-S-Cr^3+ 초교환 상호작용이 Fe^3+-S-Cr^3+ 초교환 상호작용에 비하여 상당히 약함을 의미한다. The mixed series Co_x(Cu_0.5)_(1-x)Cr_2S_4 has been studied using x-ray Mossbauer techniques. The crystal structure is found to be cubic spinel. and the lattice parameter a_0satisfies Vegard's law. The isomer shifts indicate that Fe ions are ferric and the absence of quadrupole splitting suggests the iron ions occupy. only tetrahedral sites. It is notable the all the Mossbauer Parameters at 80 K are independent of the com-position. This has been explained in terms of local atomic arrangements. The Neel temperature decreases rapidly with increasing cobalt concentration. suggesting that the superexchange interaction for the Co^2+-S-Cr^3+ link is much weaker than that for the Fe^3+-S-Cr^3+ link.

      • Combined toxicity of endosulfan and phenanthrene mixtures and induced molecular changes in adult Zebrafish (<i>Danio rerio</i>)

        <P><B>Abstract</B></P> <P>Individual and combined toxicities of endosulfan (ENDO) with phenanthrene (PHE) were evaluated using zebrafish (<I>Danio rerio</I>) adults. The 96-h LC<SUB>50</SUB> values for ENDO and PHE were 4.6 μg L<SUP>−1</SUP> and 920 μg L<SUP>−1</SUP>, respectively. To evaluate the mixture toxicity, LC<SUB>10</SUB> and LC<SUB>50</SUB> concentrations were grouped into four combinations as ENDO-LC<SUB>10</SUB> + PHE-LC<SUB>10</SUB>, ENDO-LC<SUB>10</SUB> + PHE-LC<SUB>50</SUB>, ENDO-LC<SUB>50</SUB> + PHE-LC<SUB>10</SUB>, and ENDO-LC<SUB>50</SUB> + PHE-LC<SUB>50</SUB>, and their acute toxicities were determined. The combination of LC<SUB>50</SUB>-ENDO and LC<SUB>10</SUB>-PHE exhibited a synergistic effect. In addition, acetylcholinesterase activity decreased in zebrafish bodies exposed to ENDO with or without PHE. Combined treatments induced higher glutathione <I>S</I>-transferase activity compared to individual treatments. Carboxylesterase activity increased in both heads and bodies of ENDO-treated fishes compared with PHE-treated fishes. Using RT-qPCR technique, CYP1A gene expression significantly up-regulated in all combinations, whereas CYP3A was unchanged, suggesting that enzymes involved in defense may play different roles in the detoxification. CYP7A1 gene responsible for bile acid biosynthesis is dramatically down-regulated after exposure to the synergistic combination exposure, referring that the synergistic effect may be resulted from the reduction of bile production in zebrafishes. Among gender-related genes, CYP11A1 and CYP17A1 genes in female zebrafish decreased after treatment with ENDO alone and combination of LC<SUB>50</SUB>-ENDO and LC<SUB>10</SUB>-PHE. This might be related to a reduction in cortisol production. The overall results indicated that ENDO and PHE were toxic to zebrafish adults both individually and in combination, and that their co-presence induced changes in the expression of genes responsible for metabolic processes and defense mechanisms.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Combined toxicity of endosulfan and phenanthrene up-regulated CYP1A, <I>ugt1a1</I>, and <I>sult1st1</I> genes. </LI> <LI> Combined toxicity showed a synergistic effect with a down-regulation in bile acid biosynthesis. </LI> <LI> Adrenal steroidogenesis in female zebrafishes was severely affected by endosulfan treatment. </LI> <LI> Fish gender should be considered for future toxicological studies. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
