Background: Atopic dermatitis (AD) is a chronic and relapsing inflammatory skin disease mediated by T helper type 2 (Th2) cells in acute phase. Group 2 innate lymphoid cells (ILCs) play a role in the initiation of the Th2 response. Although mold exposure is associated with the development of AD, studies on the underlying mechanisms are lacking. This study investigated whether group 2 ILCs are involved in inflammation in AD-like skin induced by Aspergillus fumigatus (Af). Methods: We investigated changes of group 2 ILCs population in Af-induced AD-like skin lesions. To induce AD-like skin lesions, Af extracts were applied to the dorsal skin of BALB/c and Rag1−/− mice five times per week, with repeat exposures at 2-week intervals. Results: The clinical parameters were higher in the Af-treated group than in the control group. Histologic findings revealed epiderrmal and dermal thickening as well as eosinophil and mast cell infiltration into the skin of Af-treated mice. Populations of group 2 ILCs in the skin were also significantly higher in the Af-treated group. In addition, interleukin-33 mRNA expression was significantly higher in the skin lesions of the Af-treated mice. In the Rag1−/− mice lacking mature lymphocytes, AD-like skin lesions were still induced by Af and ILCs depletion using an anti-CD90.2 mAb lowered the Af-induced inflammatory response. Conclusions: Group 2 ILCs may play a role in a murine model of Af-induced AD-like skin lesions.

<P><B>Abstract</B></P> <P>The effects of 3d transition-metal alloying elements (V, Cr, Mn, Co, Ni, and Cu) on the structural stability, mechanical properties, and electronic structure of Fe<SUB>3</SUB>Al alloys have been investigated using first-principles calculations. The site preference of the alloying elements was clearly observed from the formation enthalpy calculations of the Fe<SUB>3</SUB>Al–X alloys. With regards to their mechanical properties, it was found that the alloying elements V, Cr, and Ni improve the ductility of the alloys by increasing Pugh's ratio, whereas Mn, Co, and Cu cause the materials to be more brittle. Among the tested alloying elements, V leads to the largest increase of Pugh’s ratio as well as significant reduction in the alloy’s elastic anisotropy, which implies that Fe<SUB>3</SUB>Al–V should have an enhanced ductility. An electronic structure analysis showed that the alloying elements altered the density of states of the alloys. From the projected density of states and from a crystal orbit Hamiltonian population analysis, it could clearly be seen that V leads to the stabilization of Fe<SUB>3</SUB>Al–V by reducing the magnetic moment and by relaxing the spin asymmetry of the Fe atoms. This result is in good agreement with that of a previous experiment, which reported that the alloy Fe<SUB>3</SUB>Al–V has an enhanced strength. However, Ni has antibonding states at the Fermi level that are substantially filled, which was found to be detrimental to the structural stability of the Ni alloy.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Effects of alloying elements on mechanical properties of Fe<SUB>3</SUB>Al–X were investigated. </LI> <LI> V, Cr, and Ni increase Pugh's ratio, indicating the enhancement of ductility. </LI> <LI> Notably, V was effective on improving anisotropy and stability of the alloys. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

Background/Aims: Helicobacter pylori (HP) infection is positively associated with metabolic syndrome (MS). However, the long-term effects of eradication therapy on MS and sex differences have not been thoroughly studied. We aimed to investigate the long-term effects of HP eradication on MS and sex differences. Methods: This study included 2,267 subjects who visited a tertiary referral center between May 2003 and May 2019. HP was diagnosed by histology, a Campylobacter-like organism test, and culture, and the subjects were prospectively followed up. The participants were categorized into three groups: HP uninfected, HP infected but non-eradicated, and HP eradicated. The baseline characteristics and changes in metabolic parameters after HP eradication were compared over a 5-year follow-up period. Results: Among 1,521 subjects, there was no difference in baseline metabolic parameters between the HP-uninfected (n=509) and HP-infected (n=1,012) groups, regardless of sex. Analysis of the metabolic parameters during follow-up among HP-uninfected (n=509), HP-non-eradicated (n=346), and HP-eradicated (n=666) groups showed that high-density lipoprotein (HDL) and the body mass index (BMI) increased after eradication, with a significant difference at 1-year of follow-up. In females, HDL increased after eradication (p=0.023), and the BMI increased after eradication in male subjects (p=0.010). After propensity score matching, the HDL change in female remained significant, but the statistical significance of the change in BMI in the male group became marginally significant (p=0.089). Conclusions: HP eradication affected metabolic parameters differently depending on sex. HDL significantly increased only in females over time, especially at 1-year of follow-up. In contrast, BMI showed an increasing tendency over time in males, especially at the 1-year follow-up.

<P>This study optimizes the synthesis process of pellet-type adsorbents using alum sludge. The effect of the binder and heat treatment temperature on the nanopore formation in the adsorbent is investigated. The pellet-type adsorbent prepared using the powder-type sludge from water treatment is determined to be a material that contains nanopores. The specific surface area is increased significantly after the calcination process in the range of 132-172 m2/g. With the calcination treatment, the breakthrough time in the formaldehyde adsorption increases remarkably with an optimum calcination temperature of 400 °C. The breakthrough capacity of the formaldehyde increases to a maximum 2.96 mg/g at this temperature.</P>

Immunological rejection of the organ grafted onto a primate arises from two antibody mediated processes, hyperacute rejection (HAR) and acute humoral rejection (AHR). Functional ablation of α1,3-galactosyltransferase (GalT) and concurrently overexpression of complement regulatory proteins are known to inhibit HAR and AHR. In previous study, we reported that production of porcine male fibroblasts harboring a MCP expression cassette targeted to GalT locus. In this study, we constructed a different MCP expression cassette, in which the EF1α promoter regulates MCP expression and internal ribosome entry site-mediated neomycin resistance gene expression. Subsequently, this cassette was inserted between the left and the right homologous arms to target exon 9 of the GalT gene. Female fibroblasts were isolated from ear skin of 10 days old miniature pig, and used for nucelofection of the the construct for MCP expression at GalT locus. PCR analysis showed that four clones of forty neomycin resistant clones carry MCP expression cassette at exon 9 of the GalT gene. Two clones analyzed downregulated GalT expression, as determined by quantitative reverse transcriptase polymerase chain reaction. Flow cytometry analysis showed that MCP was efficiently expressed at the cell surface.

<P><B>Abstract</B></P> <P>In the eukaryotic circadian clock machinery, negative feedback repression of CLOCK (CLK) and BMAL1 transcriptional activity by PERIOD (PER) and CRYPTOCHROME (CRY) underlies the basis for 24 h rhythmic gene expression. Thus, precise regulation of the time-dependent nuclear entry of circadian repressors is crucial to generating normal circadian rhythms. Here, we sought to identify novel kinase(s) that regulate nuclear entry of mammalian CRY1 (mCRY1) with an unbiased screening using red fluorescent protein (RFP)-tagged human kinome expression plasmids in mammalian cells. Transient expression of human vaccinia-related kinase 3 (hVRK3) reduced the nuclear presence of mCRY1. hVRK3 expression also induced alterations in the subcellular localization of other core clock proteins, including mCRY2, mPER2, and BMAL1. In contrast, the subcellular localization of mCLK was not changed. Given that singly expressed mCLK mostly resides in the cytoplasm and that nuclear localization sequence (NLS) mutation of hVRK3 attenuated the effect of hVRK3 co-expression on subcellular localization, ectopically expressed hVRK3 presumably reduces the retention of proteins in the nucleus. Finally, downregulation of <I>hvrk3</I> using siRNA reduced the amplitude and lengthened the period of the cellular bioluminescence rhythm. Taken together, these data suggest that VRK3 plays a role in setting the amplitude and period length of circadian rhythms in mammalian cells.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Screening was performed to identify kinases that regulate CRY1 subcellular localization. </LI> <LI> VRK3 alters the subcellular localization of CRY1, CRY2, PER2, and BMAL1. </LI> <LI> VRK3 knock-down alters the circadian bioluminescence rhythm in mammalian cells. </LI> </UL> </P>
Factors Affecting Depression in Young People : A hybrid machine learning approach

This study aims to select the factors affecting depression in young people and to identify the relationship by using a hybrid machine learning. From 2018 to 2022, a total of four years of community health survey data were used, and the final subjects of the study were 141,510 young people aged 19 to 34. In order to identify the factors that significantly affect depression, a feature selection with Least absolute shrinkage and selection operator (LASSO) regression has been applied. After selecting variables, logistic regression analysis was performed through a complex sampling design. Through feature selection, variables such as health behavior (smoking, drinking, sleeping) and health condition (with or without hypertension) were selected. Among the variables related to health behavior, smoking (OR 1.95; 95% CI 1.82 - 2.09) and sleep_7 hours or more (OR 0.70; 95% CI 0.66 - 0.74) had a significant effect on depression. Both the hypertension and subjective health level, which are variables related to health status, had a significant effect on depression.
