Molecular and genomic features of <i>Mycobacterium bovis</i> strain 1595 isolated from Korean cattle

<P>The aim of this study was to investigate the molecular characteristics and to conduct a comparative genomic analysis of <I>Mycobacterium</I> (<I>M.</I>) <I>bovis</I> strain 1595 isolated from a native Korean cow. Molecular typing showed that <I>M. bovis</I> 1595 has spoligotype SB0140 with mycobacterial interspersed repetitive units-variable number of tandem repeats typing of 4-2-5-3-2-7-5-5-4-3-4-3-4-3, representing the most common type of <I>M. bovis</I> in Korea. The complete genome sequence of strain 1595 was determined by single-molecule real-time technology, which showed a genome of 4351712 bp in size with a 65.64% G + C content and 4358 protein-coding genes. Comparative genomic analysis with the genomes of <I>Mycobacterium tuberculosis</I> complex strains revealed that all genomes are similar in size and G + C content. Phylogenetic analysis revealed all strains were within a 0.1% average nucleotide identity value, and MUMmer analysis illustrated that all genomes showed positive collinearity with strain 1595. A sequence comparison based on BLASTP analysis showed that <I>M. bovis</I> AF2122/97 was the strain with the greatest number of completely matched proteins to <I>M. bovis</I> 1595. This genome sequence analysis will serve as a valuable reference for improving understanding of the virulence and epidemiologic traits among <I>M. bovis</I> isolates in Korea.</P>

<P>Two semiconducting conjugated polymers were synthesized via Stille polymerization. The structures combined unsubstituted or (triisopropylsilyl)ethynyl (TIPS)-substituted 2,6-bis(trimethylstannyl)benzo[1,2-<I>b</I>:4.5-<I>b</I>′]dithiophene (BDT) as a donor unit and benzotriazole with a symmetrically branched alkyl side chain (DTBTz) as an acceptor unit. We investigated the effects of the different BDT moieties on the optical, electrochemical, and photovoltaic properties of the polymers and the film crystallinities and carrier mobilities. The optical-band-gap energies were measured to be 1.97 and 1.95 eV for PBDT-DTBTz and PTIPSBDT-DTBTz, respectively. Bulk heterojunction photovoltaic devices were fabricated and power conversion efficiencies of 5.5% and 2.9% were found for the PTIPSBDT-DTBTz- and PBDT-DTBTz-based devices, respectively. This difference was explained by the more optimal morphology and higher carrier mobility in the PTIPSBDT-DTBTz-based devices. Ovarian reserve as a predicting factor for successful pregnancy after adenomyomectomy

Objective: To compile the fertility outcomes of adenomyomectomy and to evaluate which demographic factors predict a successful pregnancy in patients who have undergone adenomyomectomy. Methods: We retrospectively reviewed the patient data between January 2017 and April 2020. Possible causes for infertility were excluded by hysterosalpingography, blood tests, karyotyping, hormonal assay, and semen analysis before the surgery. Candidates were selected for surgery based on the following criteria: 1) patients aged ≤45 years, 2) patients who consented to proceed with the surgery after being informed about the possibility of recurrence of the disease and operative blood loss, and 3) patients desiring pregnancy in the future. Patients were followed up monthly for 3 months. The subsequent follow-ups were conducted every 3-6 months. Patients who wanted to get pregnant could try for pregnancy 3 months after the surgery. We defined pregnancy success as the presence of fetal heartbeat at 6-7 weeks of gestation. The number of pregnancy trials was defined as the number of embryos transferred. Results: Forty-three patients were divided into two groups: the pregnancy failure group (n=28) and the pregnancy success group (n=15). The age of patients was higher (39.0 [32.046.0] vs. 37.0 [33.042.0] years, P=0.006) and the level of anti-Mllerian hormone (AMH) (0.54 [0.018.54] vs. 2.91 [0.347.92] ng/mL, P=0.002) was lower in the pregnancy failure group than in the pregnancy success group. The operative time was longer (220.0 [68.0440.0] vs. 175.0 [65.0305.0] minutes, P=0.048) and the estimated blood loss was higher (750 [1002500] vs. 500 [502000] ml, P=0.016) in the pregnancy failure group than in the pregnancy success group. No significant difference was observed in body mass index, symptoms, cancer antigen 125, preoperative uterine volume, and type of adenomyosis. In the multivariate analysis, age and AMH were significant predictive factors for successful pregnancy. Conclusion: Ovarian reserve (age and AMH) and disease severity might be predictive factors for successful pregnancy in patients who have undergone adenomyomectomy. Adenomyomectomy should be positively considered for women desiring pregnancy and having appropriate ovarian reserve. This information will be beneficial for patients and clinicians before the decision about adenomyomectomy. Larger prospective studies are needed to confirm our findings.
Induction of Microspore-derived Embryos According to Heat Shock, Dark Incubation, and Washing Medium

Heat shock pretreatment, dark culture period and washing medium could have marked effects on microspore embryogenesis. A heat shock pretreatment of microspores at 32.5°C for 48 hours gave high production rate of microspore-derived embryo (MDE) when compare to shorter and longer period. The yield of MDE increased significantly when microspore cultured for 15 days at 25℃ in dark condition followed by heat shock pretreatment. MDE were browned and lost vitality when dark treatment period extended longer than 15 days. This is caused by an insufficient oxygen and light for growing embryo which already formed during dark treatment period. The vitality of a microspore isolated from flower bud stored at 4℃ become decreased at the very first day and the vitality of microspore stored at 4℃ in the form of flower bud itself become decreased from the 5th day after storage. This shows the possibility of getting a certain period of storage for a suitable flower bud in MDE formation. The yield of MDE was most effective when isolated microspore was had with MS medium compared to B-5 and NLN medium and also showed most effective result with sucrose 130 gL-1 in additional sucrose concentration. The above result is thought to be very useful for the development of a new cultivar of radish and other many crops including Brassica using haploid breeding technology.
