<P><B>Abstract</B></P> <P>The ciliate <I>Miamiensis avidus</I> causes scuticociliatosis in Japanese flounder <I>Paralichthys olivaceus</I>. We previously reported three serotypes of this ciliate distinguishable by serotype-specific antigenic polypeptides (serotype I, 30kDa; serotype II, 38kDa; serotype III, 34kDa). In this study, we determined the localization site of the serotype-specific polypeptides in the ciliate and determined the genes encoding the polypeptides, using the isolates IyoI (serotype I), Nakajima (serotype II), and Mie0301 (serotype III). SDS-PAGE and immunoblot analysis of cilia, membrane proteins, and cytoskeletal elements of the ciliates revealed that the polypeptides were abundant in the former two. Scanning electron microscopy of ciliates immobilized by homologous antiserum showed morphological changes in the cilia. These evidences suggested that the polypeptides were ciliary membrane immobilization antigens. The ciliary genes identified showed low identity scores—<51.5% between serotypes. To differentiate the serotypes, we designed serotype-specific PCR primer sets based on the DNA sequences. The PCR-based serotyping results were completely consistent with conventional serotyping methods (immobilization assay and immunoblot analysis). Twenty of 21 isolates were classified as either serotype I or II, and one isolate was undistinguishable. The combination of species-specific PCR previously reported and three serotype-specific PCR could be useful for identifying, serotyping, and surveillance for occurrences of new serotypes of <I>M. avidus</I>.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Three serotypes of <I>M. avidus</I> were reported in Japan and Korea. </LI> <LI> Serotype-specific polypeptides were identified as ciliary membrane proteins. </LI> <LI> The nucleotide sequences of ORFs were determined. </LI> <LI> Serotype-specific PCR revealed that the pandemic serotypes were serotype I and II. </LI> </UL> </P>

<P>A novel sustained release formulation of erythropoietin (EPO) was developed using hyaluronic acid (HA) hydrogels. For the preparation of HA hydrogels, adipic acid dihydrazide grafted HA (HA-ADH) was synthesized and analyzed with <SUP>1</SUP>H NMR. The degree of HA-ADH modification was about 69%. EPO was in situ encapsulated into HA-ADH hydrogels through a selective cross-linking reaction of bis(sulfosuccinimidyl) suberate (BS<SUP>3</SUP>) to hydrazide group (pK<SUB>a</SUB> = 3.0) of HA-ADH rather than to amine group (pK<SUB>a</SUB> > 9) of EPO. The denaturation of EPO during HA-ADH hydrogel synthesis was drastically reduced with decreasing pH from 7.4 to 4.8. The specific reactivity of BS<SUP>3</SUP> to hydrazide at pH = 4.8 might be due to its low pK<SUB>a</SUB> compared with that of amine. In vitro release of EPO in phosphate buffered saline at 37°C showed that EPO was released rapidly for 2 days and then slowly up to 4 days from HA-ADH hydrogels. When the hydrogels were dried at 37°C for a day, however, longer release of EPO up to 3 weeks could be demonstrated. According to in vivo release test of EPO from HA-ADH hydrogels in SD rats, elevated EPO concentration higher than 0.1 ng/mL could be maintained from 7 days up to 18 days depending on the preparation methods of HA-ADH hydrogels. A Study on the Effect of Fluoride-containing Pit and Fissure Sealant

100개의 우식이 없는 발거된 소구치를 산부식전 법랑질면의 기계적 처리유무와 처리방법, 산부식 처리유무에 따라 각각 6개군으로 분류하여 전색재를 도포, 광중합한 후 소와열구부위와 전색재의 경계면을 따라 색소 침투정도를 관찰하여, 연마를 시행하지 않은 상태에서 산부식 여부는 색소침투정도에 유의한 영향을 미치니 못하였고, 산부식하지 않은 상태에서 연마속도, 즉 저속과 표준속도로 연마한 경우와 저속과 표준속도로 연마한 후 산부식을 시행한 경우에서는 유의한 차이가 없었으며, 다음과 같은 결론을 얻었다. 1. 산부식없이 연마를 시행하지 않은 군과 저속으로 연마한 군, 저속으로 연마한후 산부식을 시행한 군과 시행하지 않은 군간의 색소침투정도가 유의한 차이를 보여 연마여부와 산부식여부가 전색재의 변연보쇄성에 중대한 영향을 미침을 알았다.(P<0.05). 2. 연마와 산부식을 시행하지 않은 군과 표준속도로 연마후 산부식하지 않은 군, 산부식전 연마를 시행하지 않은 군과 표준속도로 연마한 군사이의 색소침투정도 비교시 유의한 차를 발견할 수 있어 산부식 유무에 관계없이 연마여부에 따라 변연봉쇄성의 차이를 보임을 알 수 있었다.(P<0.01). 3. 저속으로 연마후 산부식한 군과 연마하지 않고 산부식한 군간에 P<0.001 수준에서 염료침투정도의 차이를 보였다.
Magnetic field effects on synthesis process of high-Tc superconductors

Materials synthesis processes in high magnetic fields are investigated for high-Tc superconductors at the High Field Laboratoryfor Superconducting Materials, Institute for Materials Research, Tohoku University. On a melt growth process of YBa2Cu3O7 bulkalso the decrease of the crystal growth rate by the magnetic field, which is suggested from a dierential temperature analysismeasurement in magnetic fields. On a chemical vapor deposition process of YBa2Cu3O7 lms deposited on the polycrystalline Agsubstrates in high magnetic elds, the microstructure changes drastically from the rectangular large grains with 10l m in size to thesmall complex shaped grains. The spiral growth mode is observed for the samples prepared at 0 T but it changes into the island2Sr2CaCu2O8 tapes in magneticelds, the homogeneous texture is obtained and it results in the increase of the critical current density.. 2003 Elsevier B.V.
