      • Quasi-static test of the precast-concrete pile foundation for railway bridge construction

        Precast concrete elements in accelerated bridge construction (ABC) extends from superstructure to substructure, precast pile foundation has proven a benefit for regions with fragile ecological environment and adverse geological condition. There is still a lack of knowledge of the seismic behavior and performance of the precast pile foundation. In this study, a 1/8 scaled model of precast pile foundation with elevated cap is fabricated for quasi-static test. The failure mechanism and responses of the precast pile-soil interaction system are analyzed. It is shown that damage occurs primarily in precast pile-soil interaction system and the bridge pier keeps elastic state because of its relatively large cross-section designed for railways. The vulnerable part of the precast pile with elevated cap is located at the embedded section, but no plastic hinge forms along the pile depth under cyclic loading. Hysteretic curves show no significant strength degradation but obvious stiffness degradation throughout the loading process. The energy dissipation capacity of the precast pile-soil interaction system is discussed by using index of the equivalent viscous damping ratio. It can be found that the energy dissipation capacity decreases with the increase of loading displacement due to the unyielding pile reinforcements and potential pile uplift. It is expected to promote the use of precast pile foundation in accelerated bridge construction (ABC) of railways designed in seismic regions.

        High Genetic Variability of Schistosoma haematobium in Mali and Nigeria

        Schistosoma haematobium is one of the most prevalent parasitic flatworms, infecting over 112 million people in Africa. However, little is known about the genetic diversity of natural S. haematobium populations from the human host because of the inaccessible location of adult worms in the host. We used 4 microsatellite loci to genotype individually pooled S. haematobium eggs directly from each patient sampled at 4 endemic locations in Africa. We found that the average allele number of individuals from Mali was significantly higher than that from Nigeria. In addition, no significant difference in allelic composition was detected among the populations within Nigeria; however, the allelic composition was significantly different between Mali and Nigeria populations. This study demonstrated a high level of genetic variability of S. haematobium in the populations from Mali and Nigeria, the 2 major African endemic countries, suggesting that geographical population differentiation may occur in the regions.

        Ardipusilloside-I stimulates gastrointestinal motility and phosphorylation of smooth muscle myosin by myosin light chain kinase

        Ardipusilloside-I is a natural triterpenoid saponin, which was isolated from Ardisia pusilla A. DC. The aim of the study was to evaluate the stimulation of ardipusilloside-I on gastrointestinal motility in vitro and in vivo. The experiment of smooth muscle contraction directly monitored the contractions of the isolated jejunal segment (IJS) in different contractile states, and the effects of ardipusilloside-I on myosin were measured in the presence of $Ca^{2+}$-calmodulin using the activities of 20 kDa myosin light chain ($MLC_{20}$) phosphorylation and myosin $Mg^{2+}$-ATPase. The effects of ardipusilloside-I on gastro emptying and intestinal transit in constipation-predominant rats were observed, and the MLCK expression in jejuna of constipated rats was determined by western blot. The results showed that, ardipusilloside-I increased the contractility of IJS in a dose-dependent manner and reversed the low contractile state (LCS) of IJS induced by low $Ca^{2+}$, adrenaline, and atropine respectively. There were synergistic effects on contractivity of IJS between ardipusilloside-I and ACh, high $Ca^{2+}$, and histamine, respectively. Ardipusilloside-I could stimulate the phosphorylation of $MLC_{20}$ and $Mg^{2+}$-ATPase activities of $Ca^{2+}$- dependent phosphorylated myosin. Ardipusilloside-I also stimulated the gastric emptying and intestinal transit in normal and constipated rats in vivo, respectively, and increased the MLCK expression in the jejuna of constipation-predominant rats. Briefly, the findings demonstrated that ardipusilloside-I could effectively excite gastrointestinal motility in vitro and in vivo.
