        Protection Against Helicobacter pylori Infection by a Trivalent Fusion Vaccine Based on a Fragment of Urease B-UreB414

        A multivalent fusion vaccine is a promising option for protection against Helicobacter pylori infection. In this study, UreB414 was identified as an antigenic fragment of urease B subunit (UreB) and it induced an antibody inhibiting urease activity. Immunization with UreB414 partially protected mice from H. pylori infection. Furthermore, a trivalent fusion vaccine was constructed by genetically linking heat shock protein A (HspA), H. pylori adhesin A (HpaA), and UreB414, resulting in recombinant HspA-HpaA-UreB414 (rHHU). Its protective effect against H. pylori infection was tested in BALB/c mice. Oral administration of rHHU significantly protected mice from H. pylori infection, which was associated with H. pylori-specific antibody production and Th1/Th2-type immune responses. The results show that a trivalent fusion vaccine efficiently combats H. pylori infection, and that an antigenic fragment of the protein can be used instead of the whole protein to construct a multivalent vaccine.

        Direct ethanol production from dextran industrial waste water by Zymomonas mobilis

        The direct production of ethanol from dextran industrial waste water was investigated by using Zymomonasmobilis via batch and semi-continuous fermentation mode. In batch fermentation, pretreated waste water (unsterilizedand sterilized), pH value (3.8 and 6.0), and Mg2+(with and without) was compared with OD600, sugar and ethanol con-centration. After 24 h fermentation, sugar in the dextran waste water was almost exhausted, and the amount of ethanolaccumulated reached 24.33-29.92 g/l, which is nearly 99% of the theoretical yield of ethanol. Kinetic parameters ofZ. mobilis in batch fermentation were also investigated. The raw dextran waste water was also used in semi-continuousfermentation. After 48 h fermentation, the production of ethanol was 28.65 g/l. These results indicated that dextranwaste water may be used as a candidate substrate and Z. mobilis could convert the raw material into ethanol directly.

        Early Diagnosis and Management of Cerebral Venous Flow Obstruction Secondary to Transsinus Fracture after Traumatic Brain Injury

        Background and Purpose Cerebral venous flow obstruction (CVFO) is a fatal complication of traumatic brain injury. To compare the outcomes of patients with CVFO secondary to traumatic-brain-injury-induced transsinus fracture who were diagnosed early versus those diagnosed late in the therapeutic course. Methods In total, 403 patients with transsinus fracture were reviewed retrospectively. The patients were divided into an early-diagnosis group (n=338) and a delayed-diagnosis group (n=65).The patients submitted to 2D time-of-flight magnetic resonance venography (2D-TOF MRV) and/or CT venography (CTV), depending upon the findings of intracranial pressure monitoring, in order to identify potentially complicated CVFO. These examinations took place within 3 days of the onset of malignant intracranial hypertension symptoms in the early-diagnosis group, and after an average of 7 days in the delayed-diagnosis group. Once diagnosed, patients received intravenous thrombolytic therapy with low-dose urokinase. Patients with massive transsinus epidural hematoma, depressed fracture, or cerebral hernia were treated surgically to relieve the compression and repair any damage to the venous sinuses. Results Cerebral venous flow obstruction was much more severe in the delayed-diagnosis group than in the early-diagnosis group (p<0.001), and hence patients in the former group were given a higher dose of urokinase (p<0.001) for thrombolytic therapy. They were also significantly more likely to need surgery (48.1% vs. 20.6%, p=0.003) and had a higher mortality rate (37.0%vs. 4.1%, p<0.001). However, patients in both groups experienced a similarly favorable prognosis, not only with regard to functional outcome but also with respect to neuroradiological improvement, as evaluated by 2D-TOF MRV/CTV at the final follow-up (p=0.218). Conclusions Delayed diagnosis can result in increased risk of surgery and death in the acute phase. Thrombolytic therapy with low-dose urokinase resulted in promising improvements in both functional and neuroradiological outcomes in all of the patients in this study, regardless of the time to diagnosis.         Study on aerodynamic coefficients and responses of the integrated catwalk of Halogaland Bridge

        Wind tunnel tests and numerical aerodynamic analyses were conducted for an integrated catwalk structure under strong winds. From the wind tunnel tests, it is found that the aerodynamic coefficients were different from those of the typical type. The drag coefficient was larger than typical and was sensitive to number of vertical meshes installed rather than the solidity ratio. Comparing with typical catwalk, the integrated one showed larger deformation under strong wind, and the large torsional deformation are mainly caused by drag force. It did not show aerodynamic divergence even the torsional deformation reaching $20^{\circ}$. The reason could be that the stiffness is smaller and thus the catwalk is able to deform to the shape compactable with higher loading. Considering safety for construction, storm rope system is introduced to the catwalk to reduce the deformation to acceptable level.

      • Association Between the Ku70-1310C/G Promoter Polymorphism and Cancer Risk: a Meta-analysis

        Ku70 plays an important role in DNA double-strand break repair. Studies revealing conflicting results on the role of the Ku70-1310C/G promoter polymorphism on cancer risk led us to perform a meta-analysis to investigate this relationship. Ten case-control studies with 2566 cases and 3058 controls were identified. Odds ratios (ORs) and 95% confidence intervals (CIs) were used to assess the strength of associations. The overall results suggested no association between the Ku70-1310C/G promoter polymorphism and total cancer risk. However, on stratified analysis, significantly increased risks were observed among the Asian population (GG vs. CC: OR=1.50, 95%CI=1.10-2.06; GG vs. CC/CG: OR=1.47, 95%CI=1.07-2.01) and population-based case-control studies (GG vs. CC: OR=1.57, 95%CI=1.12-2.22; CG vs. CC: OR=1.35, 95%CI=1.11-1.64; CG/GG vs. CC: OR=1.37, 95%CI=1.14-1.65). Additionally, variant genotypes were associated with a significantly increased breast cancer risk (GG vs. CC: OR=1.80, 95%CI=1.26-2.56; GG vs. CC/CG: OR=1.40, 95%CI=1.01-1.95).

      • Numerical studies of unsteady flow field and aerodynamic forces on an oscillating 5:1 rectangular cylinder in a sinusoidal streamwise flow

        Numerical simulations are conducted to investigate the uniform flow (UF) and sinusoidal streamwise flow (SSF) over an oscillating 5:1 rectangular cylinder with harmonic heaving motion at initial angles of attack of α = 0° and 3° using twodimensional, unsteady Reynolds-averaged Navier–Stokes (URANS) equations. First, the aerodynamic parameters of a stationary 5:1 rectangular cylinder in UF are compared with the previous experimental and numerical data to validate the capability of the computationally efficient two-dimensional URANS simulations. Then, the unsteady flow field and aerodynamic forces of the oscillating 5:1 rectangular cylinder in SSF are analysed and compared with those in UF to explore the effect of SSF on the rectangular cylinder. Results show that the alternative vortex shedding is disturbed by SSF both at α = 0° and 3°, resulting in a considerable decrease in the vortex-induced force, whereas the unsteady lift component induced by cylinder motion remains almost unchanged in the SSF comparing with that in UF. Notably, the strong buffeting forces are observed at α = 3° and the energy associated with unsteady lift is primarily because of the oscillations of SSF. In addition, the components of unsteady lift induced by the coupling effects of SSF and cylinder motion are discussed in detail.

      • Spatial correlation of aerodynamic forces on 5:1 rectangular cylinder in different VIV stages

        To better understand the vortex-induced vibration (VIV) characteristics of a 5:1 rectangular cylinder, the distribution of aerodynamic force and the non-dimensional power spectral density (PSD) of fluctuating pressure on the side surface were studied in different VIV development stages, and their differences in the stationary state and vibration stages were analyzed. The spanwise and streamwise correlations of surface pressures were studied, and the flow field structure partitions on the side surface were defined based on the streamwise correlation analysis. The results show that the variation tendencies of mean and root mean square (RMS) pressure coefficients are similar in different VIV development stages. The RMS values during amplitude growth are larger than those at peak amplitude, and the smallest RMS values are observed in the stationary state. The spanwise correlation coefficients of aerodynamic lifts increase with increase of the peak amplitude. However, for the lock-in region, the maximum spanwise correlation coefficient for aerodynamic lifts occurs in the VIV rising stage rather than in the peak amplitude stage, probably due to the interaction of vortex shedding force (VSF) and self-excited force (SEF). The streamwise correlation results show that the demarcation point positions between the recirculation region and the main vortex region remain almost constant in different VIV development stages, and the reattachment points gradually move to the tailing edge with increasing amplitude. This study provides a reference to estimate the demarcation point and reattachment point positions through streamwise correlation and phase angle analysis from wind tunnel tests.
