Effects of Green Tea Catechins (GTC) on the Treatment of Hangover and Prevention of Liver Disease

Over-consumption of alcohol leads to many side-effects such as malnutrition, liver disease, and neuronal disorders and many investigators have tried to identify methods for preventing the side-effects of drinking. This study was carried out to investigate the effect of the beverage contained green tea catechins (GTC) on the alcohol administered rats. We observed that blood alcohol concentration level decreased significantly in plasma. GTC (200 mg/kg) also reduced the aspartate aminotransferase (AST) and alanine aminotransferase (ALT) level of the intoxicated rats. These results suggest that GTC may be useful for the prevention and therapy of hepatotoxic pathogenesis.
Inhibitory Effects of Moutan Cortex Radicis Extracts and Paeonol on Rabbit Platelet Aggregation

The present study was undertaken to investigate the effects of Moutan Cortex Radicis extracts and paeonol, a major component, on rabbit platelet aggregation and thromboxane (TX) B₂ formation. Moutan Cortex Radicis methanol and butanol layers (100 μg/mL) showed the weak inhibitions, and water layer (100 μg/ mL) had no effect on the collagen-induced platelet aggregation. Whereas, hexane and EtOAc layers potently inhibited the collagen (3 μg/mL)-induced platelet aggregation with the IC_(50) values of 10.9±1.0 and 31.5±0.8 μg/mL, respectively. Paeonol isolated from the hexane-acetone layer specifically inhibited the collagen-induced platelet aggregation with the IC_(50) value of 113.1 ± 0.9 μM, whereas it had little effects on the other agonists such as AA-, thrombin-, A23187- and thapsigargin-induced platelet aggregations. Paeonol also potently inhibited the collagen-induced TXB₂ formation in rabbit platelet in a concentration-dependent manner. These results suggest that paeonol may inhibit rabbit platelet aggregation by interfering with an essential step in collagen-induced platelet activation and TXA₂ formation. Paeonol may be a promising candidate for an antiplatelet agent.
USP14 inhibition regulates tumorigenesis by inducing apoptosis in gastric cancer

Deubiquitinases (DUBs) are an essential component of theubiquitin-proteasome system (UPS). They trim ubiquitin fromsubstrate proteins, thereby preventing them from degradation,and modulate different cellular processes. Ubiquitin-specificprotease 14 (USP14) is a DUB that has mainly been studied forits role in tumorigenesis in several cancers. In the present study,we found that the protein levels of USP14 were remarkablyhigher in gastric cancer tissues than in the adjacent normaltissues. We also demonstrated that the inhibition of USP14activity using IU1 (an USP14 inhibitor) or the inhibition ofUSP14 expression using USP14-specific siRNA markedly reducedthe viability of gastric cancer cells and suppressed their migratoryand invasive abilities. The reduction in gastric cancercell proliferation due to the inhibition of USP14 activity was aresult of the increase in the degree of apoptosis, as evidencedby the increased expression levels of cleaved caspase-3 andcleaved PARP. Furthermore, an experiment using the USP14inhibitor IU1 revealed that the inhibition of USP14 activitysuppressed 5-fluorouracil (5-FU) resistance in GC cells. Collectively,these findings indicate that USP14 plays critical roles ingastric cancer progression and suggest its potential to serve asa novel therapeutic target for gastric cancer treatment.

Although phoscorites and carbonatites form only a minor proportion of the earth's crustal rocks, these unusual rocks and their intimate relation are of both academic and economic importance. Rare metal (Nb, Zr, Ta) and REEs mineralizations are in close relation with the differentiation of these phoscorite-carbonatite complexes (PCCs). Recent integrated petrological and geochemical data on PCCs in the Kola Alkaline Province, Arctic, indicate that phoscorites and associated carbonatites are differentiated from common 'carbonated silicate patental magma'. Various hypotheses for the genesis of phoscorite-carbonatite complexes have been proposed during the last half-century. A simple magmatic fractionation scheme can not explain the chemical and mineralogical characteristics of phoscorite and conjugate carbonatite. Instead, the hypotheses involving liquid immiscibility and coeval accumulation processes are favored to explain the mineralogical and geochemical characteristics of phoscorite and carbonatite association.
Antiplatelet Activity of KR-32558, a Novel Selective Sodium/hydrogen Exchanger-1 Inhibitor

We investigated the antiplatelet effect of a newly synthesized guanidine derivative KR-32558, a sodium-hydrogen exchanger-1 (NHE-1) inhibitor, together with the elucidation of the possible mechanisms of action. KR-32558 concentration -dependently inhibited the aggregation of washed rabbit platelets induced by collagen (10 μg/ml) with an IC_(50) value of 85.9 μM, but with much weaker potency against aggregation induced by thapsigargin (0.5 μM) or A23187 (5 μM). And had no effect on platelet aggregation induced by arachidonic acid (100 μM), thrombin (0.05 U/ml) and U46619 (1 μM) up to 100 μM. KR-32558 completely inhibited the [Ca^(2+)] mobilization induced by collagen at concentration of 100μiM. Taken together, these observations suggest that KR-32558 selectively inhibited collagen-mediated platelet aggregation by blocking the cytoplasmic calcium mobilization in addition to NHE-1 inhibition.
