Epigenetic fields for cancerization are involved in development of human cancers, especially those associated with inflammation and multiple occurrences. However, it is still unclear when such field defects are formed and what component of inflammation is involved in induction of aberrant DNA methylation. Here, in a mouse colitis model induced by dextran sulfate sodium (DSS), we identified three CpG islands specifically methylated in colonic epithelial cells exposed to colitis. Their methylation levels started to increase as early as 8 weeks after DSS treatment and continued to increase until colon cancers developed at 15 weeks. In contrast to the temporal profile of DNA methylation levels, infiltration of inflammatory cells spiked immediately after the DSS treatment and then gradually decreased. Exposure of cultured colonic epithelial cells to DSS did not induce DNA methylation and it was indicated that inflammation triggered by the DSS treatment was responsible for methylation induction. To clarify components of inflammation involved, severe combined immunodeficiency (SCID) mice that lack functional T- and B-cells were similarly treated. Even in SCID mice, DNA methylation, along with colon tumors, were induced at the same levels as in their background strain of mice (C.B17). Comparative analysis of inflammation-related genes showed that Ifng, Il1b and Nos2 had expression concordant with methylation induction whereas Il2, Il6, Il10, Tnf did not. These results showed that an epigenetic field defect is formed at early stages of colitis-associated carcinogenesis and that functional T and B cells are non-essential for the formation.
Cardenolides and β-Sitosterol Glucoside from Pergularia tomentosa L.

The aerial parts of Pergularia tomentosa L. afforded three cardenolides, desglucouzarin, coroglaucigenin and uzarigenin, in addition to β-sitosterol glucoside. The isolated compounds were identified by physical and spectral means, including IR, UV, [α]_D 1D-, 2D-NMR and FAB-MS experiments. The cardenolides, ghalakinoside, calactin and pergularoside previously reported from roots, were also identified in the aerial parts.

The present study was done to examine whether inoculated and established bacteria in the digestive tract of germ-free (GF) chickens affect growth performance, energy availability, nitrogen utilization and neutral detergent fibre (NDF) digestibility of the host bird fed a high-fibre diet. Gnotobiotic (GB) chicks were made from GF birds by co-inoculating with Ruminococcus albus, and Staphylococcus warneri, only the latter of which was established in the chicken gut. No difference was detected among conventional (CV), GF and GB birds in body weight gain, food intake or food efficiency from 7 to 21 d of age. The amount of nitrogen retained was larger in CV than in GF and GB chicks. DE and ME values of the diet and NDF digestibility were higher in CV birds than in GF and GB counterparts. It was concluded, therefore, that the established bacterium S. warneri did not give any beneficial effects on the host bird as judged by growth performance, energy availability, nitrogen utilization, and NDF digestibility.
EFFECT OF GRANULOSA AND CUMULUS CELLS ON IN VITRO DEVELOPMENT OF THE BOVINE FOLLICULAR OOCYTES

The effect of co-culture with cumulus cells and granulosa cells during maturation and development on in vitro developmental potency of follicular oocytes was examined. TCM-199 supplemented with 15% FCS and hormones was used as maturation medium. Sperm from frozen semen was capacitated in modified mTALP medium containing 0.3% BSA, $10{\mu}g/ml$ heparin and 5 mM/ml caffeine. The fertilized embryos were co-cultured on monolayer of cumulus cells or granulosa cells in TCM-199. The embryo co-cultured with cumulus cells showed higher percentage of embryo developed to morula and blastocyst (73.3%) than the embryo co-cultured without cumulus cells (30.8%). The percentage of oocytes developed to morula and blastocyst among cleaved oocytes was significantly (p < 0.05) higher in the oocytes co-cultured with cumulus cells during development (62.4%) than in the oocytes co-cultured with granulosa cells during maturation and with cumulus cells during development (52.3%), and in the oocytes co-cultured with granulose cells during development (52.8%). The results of this study indicate that co-culture of in vitro fertilized embryos with cumulus cells in the development medium increased the rate of embryos developed to morula and blastocyst among cleaved oocytes.
