With conventional mass spectrometry (MS), ions are separated according to mass/charge (m/z) ratios. We must speculate the z values to obtain the m values. Superconducting tunnel junction (STJ) detectors can solve this problem, and true mass spectrometry becomes possible instead of m/z spectrometry. The STJ detectors were installed in MS instruments with a variety of ion sources. As an example, we report fragmentation analysis of a non-covalent protein complex of hemoglobin.

Dark aerobic methane emission associated to leaf factors of two Acacia and five Eucalyptus species

We sought the biological factors determining variations in the methane emission rates from leaves of different plant species under aerobic conditions. Accordingly, we studied relations between the methane emission rate and leaf traits of two Acacia and five Eucalyptus species. We grew seedlings of each species in a glasshouse and measured the methane emission rate of the detached leaves under dark conditions at 30 <SUP>o</SUP>C. At the same time we measured the leaf mass per area (LMA), water content, and concentrations of carbon and nitrogen. There was no correlation between the leaf nitrogen concentration and the methane emission rate. This is consistent with previous findings that enzymatic processes do not influence methane emission. We found a significant negative correlation between LMA and the methane emission rate. Our results suggest that leaf structure is primarily responsible for differences in the rates of aerobic methane emission from leaves of different species.

Seventy-six animals including cattle, sheep, horses, 6 species of zoo animals were employed for collection of fresh feces in order to detect verotoxigenic Esherichia coli (VTEC) by safe, quick and sensitive PCR-based molecular methods. Bacterial cell disruption with bead-beating followed by bacterial DNA purification with hydroxyapatide chromatography and gel filtration allowed DNA preparation from animal feces with high recovery and purity. A mountain goat was firstly shown by PCR and sequencing to shed verotoxin 2 gene (vt2) that was used to generate vt2 probe and second primer set for nested PCR to attempt more sensitive detection. Most sensitive nested PCR revealed that 45% of tested cattle and 47% of tested zoo animals were VTEC-positive, while least sensitive normal PCR detected VTEC from none of these animals except a mountain goat. Moderately sensitive detection by PCR in combination with hybridization suggested that the VTEC density varied between the VTEC-positive cattle.
Analysis of Wide-gap Semiconductors with Superconducting XAFS Apparatus

Fluorescent yield X-ray absorption fine structure (XAFS) spectroscopy is useful for analyzing local structure of specific elements in matrices. We developed an XAFS apparatus with a 100-pixel superconducting tunnel junction (STJ) detector array with a high sensitivity and a high resolution for light-element dopants in wide-gap semiconductors. An STJ detector has a pixel size of 100 μm square, and an asymmetric layer structure of Nb(300 nm)-Al(70 nm)/AlOx/Al(70 nm)-Nb(50 nm). The 100-pixel STJ array has an effective area of 1 mm2. The XAFS apparatus with the STJ array detector was installed in BL-11A of High Energy Accelerator Research Organization, Photon Factory (KEK PF). Fluorescent X-ray spectrum for boron nitride showed that the average energy resolution of the 100-pixels is 12 eV in full width half maximum for the N-K line, and The C-K and N-K lines are separated without peak tail overlap. We analyzed the N dopant atoms implanted into 4H-SiC substrates at a dose of 300 ppm in a 200 nm-thick surface layer. From a comparison between measured X-ray Absorption Near Edge Structure (XANES) spectra and ab initio FEFF calculations, it has been revealed that the N atoms substitute for the C site of the SiC lattice.
