        Attenuation of Experimental Autoimmune Hepatitis in Mice with Bone Mesenchymal Stem Cell-Derived Exosomes Carrying MicroRNA-223-3p

        MicroRNA-223-3p (miR-223-3p) is one of the potential microRNAs that have been shown to alleviate inflammatory responses in pre-clinical investigations and is highly encased in exosomes derived from bone mesenchymal stem cells (MSC-exosomes). MSC-exosomes are able to function as carriers to deliver microRNAs into cells. Autoimmune hepatitis is one of the challenging liver diseases with no effective treatment other than steroid hormones. Here, we examined whether MSC-exosomes can transfer miR-223-3p to treat autoimmune hepatitis in an experimental model. We found that MSC-exosomes were successfully incorporated with miR-223-3p and delivered miR-223-3p into macrophages. Moreover, there was no toxic effect of exosomes on the macrophages. Furthermore, treatments of either exosomes or exosomes with miR-223-3p successfully attenuated inflammatory responses in the liver of autoimmune hepatitis and inflammatory cytokine release in both the liver and macrophages. The mechanism may be related to the regulation of miR-223-3p level and STAT3 expression in the liver and macrophages. These results suggest that MSC-exosomes can be used to deliver miR-223-3p for the treatment of autoimmune hepatitis.

      • A Mechanism to Improve the Performance of Zone-Based Broadcasting Protocol in the Recovery Phase

        Wireless Sensor Networks (WSNs) have been widely used in environment monitoring to collect useful information. Packet broadcasting is an essential communication behavior for establishing a communication path from sink node to an interested region of sensor nodes. ZBP is a cellular based broadcast protocol, and it successfully outperformed simple flooding manner broadcasting and common cellular based approach in two main aspects: first it reduced the number of sensor nodes that forward transmission packets and thus save energy consumption; second, it can efficiently avoid collision by properly arranging broadcasting schedule. However it still faces broadcasting failure caused by blank region of sensors. This paper improves ZBP by introducing an energy efficient recovery mechanism which recovers broadcasting when blank region interrupts the request forwarding. Compared with the simple over-network flooding recovery used by ZBP, our proposed protocol can reduce much energy consumption.

        Engineered exosomes enriched in netrin-1 modRNA promote axonal growth in spinal cord injury by attenuating inflammation and pyroptosis

        Spinal cord injury (SCI) brings a heavy burden to individuals and society, and there is no effective treatment at present. Exosomes (EX) are cell secreted vesicles containing molecules such as nucleic acids and proteins, which hold promise for the treatment of SCI. Netrin-1 is an axon guidance factor that regulates neuronal growth. We investigated the effects of engineered EX enriched in netrin-1 chemically synthetic modified message RNA (modRNA) in treating SCI in an attempt to find a novel therapeutic approach for SCI.Netrin-1 modRNA was transfected into bone marrow mesenchymal stem cells to obtain EX enriched with netrin-1 (EX-netrin1). We built an inflammatory model in vitro with lipopolysaccharide (LPS) in vitro to study the therapeutic effect of EX-netrin1 on SCI. For experiments in vitro, ELISA, CCK-8 assay, immunofluorescence staining, lactate dehydrogenase release experiments test, real-time quantitative polymerase chain reaction, and western blot were conducted. At the same time, we constructed a rat model of SCI. MRI, hematoxylin-eosin and Nissl staining were used to assess the extent of SCI in rats.In vitro experiments showed that EX had no effect on the viability of oligodendrocytes and PC12 cells. EX-netrin1 could attenuate LPS-induced inflammation and pyroptosis and accelerate axonal/dentritic growth in PC12 cells/oligodendrocytes. In addition, netrin-1 could activate the PI3K/AKT/mTOR signalling pathway upon binding to its receptor unc5b. When Unc5b and PI3K were inhibited, the effect of EX-netrin1 was weakened, which could be reversed by PI3K or mTOR activator. Our in vivo experiments indicated that EX-netrin1 could promote recovery in rats with SCI.We found that EX-netrin1 regulated inflammation, pyroptosis and axon growth in SCI via the Unc5b/PI3K/AKT/mTOR pathway, which provides a new strategy for the treatment of SCI.

        The relationship between myofiber characteristics and meat quality of Chinese Qinchuan and Luxi cattle

        Objective: The objectives of this study were to explore the expression patterns of myosin heavy chain (MyHC) genes of different skeletal muscles from Chinese cattle, and to investigate the relationship between myofiber characteristics and meat quality of M. longissimus lumborum (LL), M. psoas major (PM), and M. semimembranosus (SM) from Chinese Luxi and Qinchuan cattle. Methods: Three major muscles including LL, PM, and SM from Chinese Luxi cattle and Chinese Qinchuan cattle were used in this study. The myofiber characteristics were measured by histochemical analysis. The MyHC isoforms expression was evaluated by real-time quantitative polymerase chain reaction. Quality traits including pH value, meat color, cooking loss, Warner-Bratzler shear force (WBSF) and sarcomere length were determined at day 5 postmortem. Results: PM muscle had higher pH value, a* value, sarcomere length and lower WBSF value compared to LL and SM muscles (p<0.05). Numbers of type I myofiber and the relative expression of MyHC I mRNA in PM muscle were higher than those of LL and SM muscles (p<0.05). Myofiber diameter of PM muscle was lower than that of LL and SM muscles, regardless of myofiber types (p<0.05). Conclusion: According to the stepwise linear regression analyses, tenderness was influenced by myofiber characteristics in all three examined muscles. Tenderness of beef muscles from Qinchuan and Luxi cattle could be improved by increasing numbers of type I myofiber.         Ambient Backscatter Assisted Wireless Powered Communications

        <P>Ambient backscatter communication technology has been introduced recently, and is quickly becoming a promising choice for self-sustainable communication systems, as an external power supply or a dedicated carrier emitter is not required. By leveraging existing RF signal resources, ambient backscatter technology can support sustainable and independent communications and consequently open up a whole new set of applications that facilitate Internet of things (IoT). In this article, we study an integration of ambient backscatter with wireless powered communication networks (WPCNs). We first present an overview of backscatter communication systems with an emphasis on the emerging ambient backscatter technology. Then we propose a novel hybrid transmitter design by combining the advantages of both ambient backscatter and wireless powered communications. Furthermore, in the cognitive radio environment, we introduce a multiple access scheme to coordinate hybrid data transmissions. The performance evaluation shows that the hybrid transmitter outperforms traditional designs. In addition, we discuss open issues related to ambient backscatter networking.</P>

        On the H∞ Deconvolution Fixed-lag Smoothing

        The paper deals with the H∞ deconvolution fixed-lag smoothing problem for a linear time-invariant discrete-time system with both known and unknown input series. The H∞ fixed-lag smoother is derived by proposing a new approach termed as re-organized innovation analysis in Krein space. Under the new approach, it is clearly shown that the central deconvolution smoother in an H∞ setting is the same as the one in an H2 setting associated with one self-constructed stochastic state-space model. This insight allows us to calculate the complicated H∞ deconvolution smoother in an intuitive and simple way. The deconvolution smoother is calculated by performing Riccati equation with the same order as the original system.

        Taxonomy of fungal complex causing red-skin root of Panax ginseng in China

        Background: Red-skin root of Asian ginseng (Panax ginseng) significantly reduces the quality and limits theproduction of ginseng in China. The disease has long been thought to be a noninfectious physiologicaldisease, except one report that proved itwas an infectious disease. However, the causal agents have not beensuccessfully determined. In the present study, we were to reveal the pathogens that cause red-skin disease. Methods: Ginseng roots with red-skin root symptoms were collected from commercial fields in NortheastChina. Fungi were isolated from the lesion and identified based on morphological characters alongwith multilocus sequence analyses on internal transcription spacer, b-tubulin (tub2), histone H3 (his3),and translation elongation factor 1a (tef-1a). Pathogens were confirmed by inoculating the isolates inginseng roots. Results: A total of 230 isolates were obtained from 209 disease samples. These isolates were classifiedinto 12 species, including Dactylonectria sp., D. hordeicola, Fusarium acuminatum, F. avenaceum, F. solani,F. torulosum, Ilyonectria mors-panacis, I. robusta, Rhexocercosporidium panacis, and three novel speciesI. changbaiensis, I. communis, and I. qitaiheensis. Among them, I. communis, I. robusta, and F. solani had thehighest isolation frequencies, being 36.1%, 20.9%, and 23.9%, respectively. All these species isolated werepathogenic to ginseng roots and caused red-skin root disease under appropriate condition. Conclusion: Fungal complex is the causal agent of red-skin root in P. ginseng.

        Structural and Optical Properties of RF Magnetron Reactively Sputtered Ag2O Film

        A series of <111> oriented Ag2O films was deposited onto glass substrates by using RF magnetron reactive sputtering at different flow ratios (FRs) of oxygen to argon. The Ag2O film deposited at FR = 0.667 was the best <111> oriented film due to dual contributions from both lattice strain and film stress. The films’ transmissivity of over 70% in the near-infrared region indicated that Ag2O films are not suitable for applications as transparent conductive films in the visible region. Ag2O films deposited at FRs from 0.467 to 0.800 had optical band gaps ranging from 3.266 eV to 3.107 eV. The redshift in the films’ absorption edge may be attributed to the decrease in the lattice strain with increasing in FR.         Wireless-Powered Device-to-Device Communications With Ambient Backscattering: Performance Modeling and Analysis

        <P>The recent advanced wireless energy harvesting technology has enabled wireless-powered communications to accommodate wireless data services in a self-sustainable manner. However, wireless-powered communications rely on active RF signals to communicate and result in high power consumption. On the other hand, ambient backscatter technology that passively reflects existing RF signal sources in the air to communicate has the potential to facilitate an implementation with ultra-low power consumption. In this paper, we introduce a hybrid device-to-device (D2D) communication paradigm by integrating ambient backscattering with wireless-powered communications. The hybrid D2D communications are self-sustainable, as no dedicated external power supply is required. However, since the radio signals for energy harvesting and for backscattering come from the ambient, the performance of the hybrid D2D communications depends largely on environment factors, e.g., distribution, spatial density, and transmission load of ambient energy sources. Therefore, we design two mode selection protocols for the hybrid D2D transmitter, allowing a more flexible adaptation to the environment. We then introduce analytical models to characterize the impacts of the considered environment factors on the hybrid D2D communication performance. Together with extensive simulations, our analysis shows that the communication performance benefits from larger repulsion, transmission load, and density of ambient energy sources. Furthermore, we investigate how different mode selection mechanisms affect the communication performance.</P>
