      • Differential Role of VDR Coactivators SRC3 and MED1 in Regulating Skin Homeostasis

        The vitamin D receptor (VDR) participates in skin homeostasis maintenance by modulating sequential expression of a number of genes involved in proliferation, differentiation, barrier function, and immune response, etc. This process is under precise regulation of coactivators and corepressors, activating or inhibiting the transcription activity of VDR in specific spatio-temporal fashion. In keratinocytes, the major coactivator complexes include the vitamin D receptor interacting protein (DRIP, or Mediator) complex and steroid receptor coactivator (SRC) complex. MED1, a subunit of Mediator that that binds directly to VDR, is abundantly expressed in proliferating keratinocytes. We found that MED1 had a specific role in the regulation of epidermal stem cell self-renewal, keratinocyte proliferation and differentiation ability by modulating the activation of β-catenin signaling pathways. SRC3, on the other hand, regulates the ability of 1,25(OH)2D3 to induce functions in the more differentiated keratinocyte such as lipid synthesis and processing required for permeability barrier formation and innate immune response triggered by disruption of the barrier. Therefore, we provide evidence to show that alternative combination of MED1 or SRC3 with VDR could regulate the on and off of target genes expression, selectively control different VDR functions, and achieve the skin homeostasis maintenance in diverse layers of skin.

        Image Captioning with Synergy-Gated Attention and Recurrent Fusion LSTM

        Long Short-Term Memory (LSTM) combined with attention mechanism is extensively used to generate semantic sentences of images in image captioning models. However, features of salient regions and spatial information are not utilized sufficiently in most related works. Meanwhile, the LSTM also suffers from the problem of underutilized information in a single time step. In the paper, two innovative approaches are proposed to solve these problems. First, the Synergy-Gated Attention (SGA) method is proposed, which can process the spatial features and the salient region features of given images simultaneously. SGA establishes a gated mechanism through the global features to guide the interaction of information between these two features. Then, the Recurrent Fusion LSTM (RF-LSTM) mechanism is proposed, which can predict the next hidden vectors in one time step and improve linguistic coherence by fusing future information. Experimental results on the benchmark dataset of MSCOCO show that compared with the state-of-the-art methods, the proposed method can improve the performance of image captioning model, and achieve competitive performance on multiple evaluation indicators.

        Effect of hyperthermia on cell viability, amino acid transfer, and milk protein synthesis in bovine mammary epithelial cells

        The reduction of milk yield caused by heat stress in summer is the main condition restricting the economic benefits of dairy farms. To examine the impact of hyperthermia on bovine mammary epithelial (MAC-T) cells, we incubated the MAC-T cells at thermal-neutral (37℃, CON group) and hyperthermic (42℃, HS group) temperatures for 6 h. Subsequently, the cell viability and apoptotic rate of MAC-T cells, apoptosis-related genes expression, casein and amino acid transporter genes, and the expression of the apoptosis-related proteins were examined. Compared with the CON group, hyperthermia significantly decreased the cell viability (p < 0.05) and elevated the apoptotic rate (p < 0.05) of MAC-T cells. Moreover, the expression of heat shock protein (HSP)70, HSP90B1, Bcl-2-associated X protein (BAX), Caspase-9, and Caspase-3 genes was upregulated (p < 0.05). The expression of HSP70 and BAX (pro-apoptotic) proteins was upregulated (p < 0.05) while that of B-cell lymphoma (BCL)2 (antiapoptotic) protein was downregulated (p < 0.05) by hyperthermia. Decreased mRNA expression of mechanistic target of rapamycin (mTOR) signaling pathway-related genes, amino acid transporter genes (SLC7A5, SLC38A3, SLC38A2, and SLC38A9), and casein genes (CSNS1, CSN2, and CSN3) was found in the heat stress (HS) group (p < 0.05) in contrast with the CON group. These findings illustrated that hyperthermia promoted cell apoptosis and reduced the transport of amino acids into cells, which inhibited the milk proteins synthesis in MAC-T cells.

        Performance analysis of two-dimensional DOA estimation for uniform circular array

        Perturbation analysis of direction of arrival (DOA) estimation has been studied mostly on one-dimensional arrays rather than planar arrays, which remains room for exploration. For uniform circular arrays with various model errors, we establish the corresponding system model. Then a performance analysis of the two-dimensional MUSIC algorithm is proposed by first-order error analysis and verified by simulation. The results show that the root-mean-squared error of DOA estimation is proportional to the standard deviation of the model errors, including gain-phase errors, sensor position errors, and mutual coupling effect.
