        Statistical Optimization of Medium Components for Milk-Clotting Enzyme Production by Bacillus amyloliquefaciens D4 Using Wheat Bran-an Agro-Industry Waste

        In this paper, two statistical methods were applied to optimize medium components to improve the production of the milk-clotting enzyme by Bacillus amyloliquefaciens D4. First, wheat bran juice, skim milk powder, and Na2HPO4 were shown to have significant effects on D4 enzyme production using the Plackett?Burman experimental design. Subsequently, an optimal medium was obtained using the Box?Behnken method, which consisted of 3.31 g/l of skim milk powder, 5.0 g/l of sucrose, 0.1 g/l of FeSO4?7H2O, 0.1 g/l of MgSO4?7H2O, 0.1 g/l of MnSO4?2H2O, 0.1 g/l of ZnSO4?7H2O, 1.52 g/l of Na2HPO4, and 172.45 g/l of wheat bran juice. With this optimal medium, the milk-clotting enzyme production was remarkably enhanced. The milk-clotting enzyme activity reached 3,326.7 SU/ml after incubation of 48 h, which was 1.76-fold higher than that of the basic medium, showing that the Plackett?Burman design and Box?Behnken response surface method are effective to optimize medium components, and B. amyloliquefaciens D4 possessed a high rennet-producing capacity in the optimal medium.

        A large-area free-standing graphene oxide multilayer membrane with high stability for nanofiltration applications

        <P><B>Abstract</B></P> <P>A flexible and free-standing graphene oxide and nylon 6 (GO@nylon 6) multilayer nanofiltration membrane was prepared by a layer-by-layer assembly process. The combination of electrospinning and electrospraying technique was employed, which can facilely prepare large-area membrane with size of 20 × 30 cm. The mechanical stability of multilayer membrane has enhanced significantly due to the tightly locked structure achieved by nylon 6 nanofibers network. The novel GO@nylon 6–13 multilayer nanofiltration membrane demonstrated a pure water flux up to 11.15 L m<SUP>−2</SUP> h<SUP>−1</SUP> bar<SUP>−1</SUP>, while keeping high organic dye rejection rate (>95% for methylene blue, and >99% for methyl orange). The rejections rate of the Na<SUB>2</SUB>SO<SUB>4</SUB>, NaCl, CuSO<SUB>4</SUB>, and Pb(NO<SUB>3</SUB>)<SUB>2</SUB> were 56.5%, 27.6%, 36.7%, and 18.9%, respectively.         ATP6V0d2 Suppresses Alveoli Macrophage Alternative Polarization and Allergic Asthma via Degradation of PU.1

        Purpose Macrophages are important regulators of environmental allergen-induced airway inflammation and asthma. ATP6V0d2 is a subunit of vacuolar ATPase highly expressed in macrophages. However, the functions of ATP6V0d2 in the regulation of pathogenesis of allergic asthma remain unclear. The aim of this study is to determine the function and related molecular mechanisms of macrophage protein ATP6V0d2 in allergic asthma. Methods We compared the disease severity between female C57BL/6 wild-type and ATP6V0d2−/− mice in an ovalbumin (OVA)-induced asthma model. We also investigated the association of expression of ATP6V0d2, PU.1 and CCL17 with disease severity among asthmatic patients. Results The expression of ATP6V0d2 in sputum cells of asthmatic patients and in the lungs of OVA-challenged mice was enhanced compared to healthy subjects and their counterparts, respectively. However, ATP6V0d2-deficient mice exaggerated inflammatory cell infiltration as well as enhanced alternative activated macrophage (AAM) polarization and mucus production in an OVA-induced asthma model. Furthermore, we found that Atp6v0d2 promoted lysosomal degradation of Pu.1, which induced AAM polarization and Ccl17 production. Among asthma patients, ATP6V0d2 expression was inversely associated with disease severity, whereas PU.1 and CCL17 expression was positively associated with disease severity. Conclusions Our results identify macrophage Atp6v0d2, as an induced feedback inhibitor of asthma disease severity by promoting Pu.1 lysosomal degradation, which may in turn leads to reduced AAM polarization and Ccl17 production.

        A Duplex PCR Assay for Rapid Detection of Phytophthora nicotianae and Thielaviopsis basicola

        A duplex PCR method was developed for simultaneous detection and identification of tobacco root rot pathogens Phytophthora nicotianae and Thielaviopsis basicola. The specific primers for P. nicotianae were developed based on its internal transcribed spacer (ITS) regions of ribosomal gene, ras gene and hgd gene, while the specific primers for T. basicola were designed based on its ITS regions and β-tubulin gene. The specificity of the primers was determined using isolates of P. nicotianae, T. basicola and control samples. The results showed that the target pathogens could be detected from diseased tobacco plants by a combination of the specific primers. The sensitivity limitation was 100 fg/μl of pure genomic DNA of the pathogens. This new assay can be applied to screen out target pathogens rapidly and reliably in one PCR and will be an important tool for the identification and precise early prediction of these two destructive diseases of tobacco.

        Linear Optimal Estimation for Discrete-time and Continuous-time Systems with Multiple Measurement Delays

        In this paper, we investigate the linear optimal estimation problems of discrete-time and continuous-time systems with multiple state delays in measurements. For discrete-time systems, we obtain the linear optimal estimation of state by direct calculation of optimal gain in terms of the solution to a retarded Riccati-like difference equation instead of a group of Riccati difference equations. For continuous-time systems, we also obtain the analytical expression of linear optimal estimation without resorting to Riccati partial differential equations. All the Riccati equations are of the same dimension as the system to be estimated and the computational cost is much saved. Infinite horizon case is also studied by stability analysis. Kalman filter can be recovered from our result when delays disappear. A numerical example is provided to demonstrate the results.

      • Clinicopathologic Characteristics and Prognostic Factors in Patients with Operable HER-2 Overexpressing Breast Cancer

        Objective: To study the relationship between clinical pathologic characteristics, treatment modalities and prognostic factors in HER-2 (Human Epidermal growth factor Receptor-2) overexpressed breast carcinoma. Materials and Methods: Major clinico-pathological factors including therapeutic modalities and survival status of 371 breast cancer patients with HER2 over-expression, teated at Yantai Yuhuangding Hospital from March of 2002 to December of 2010 were retrospectively studied, with special attention focused on survival-related factors. Results: The median age of the total 371 patients in this study was 48 years at time of diagnosis, among which, the leading pathological type was infiltrating ductal carcinoma (92.5%); 62.8% presented with a primary tomor larger than 2 cm in diameter at diagnosis, 51.0% had axillary lymph node (ALN) metastases; ER (Estrogen receptor)/PR (Progesterone receptor) double negative occured in 52.8% of cases, and PCNA (proliferation cell nuclear antigen) (+++) was found in 55.1%. HER-2 overexpressed patients were usually in advanced stage when the diagnosis was made (72.8% at stages IIA~IIIC). The prognosis and survival were assessed in 259 patients with complete follow-up data. 5-year DFS (disease-free survival) and OS (overall survival) rate was 68.0% and 78.0% respectively. Univariate analysis revealed that age, tumor size, ALN metastases, LVSI (lymph-vascular space involvement), PCNA status, hormonal therapy, chemotherapy cycles, and HER-2 overexpression, correlated closely with the prognosis. ALN metastases, LVSI, PCNA status and chemotherapy cycles were independent predictors of survival. Conclusions: HER-2 overexpressed breast cancer has special clinical and pathological characteristics, with advanced clinical stages and high rate of ER/PR double negative. Lymph node metastases, LVSI, PCNA and chemotherapy cycles are independent predictors of prognosis.

      • Improving the Stability of High-Performance Multilayer MoS<sub>2</sub> Field-Effect Transistors

        <P>In this study, we propose a method for improving the stability of multilayer MoS<SUB>2</SUB> field-effect transistors (FETs) by O<SUB>2</SUB> plasma treatment and Al<SUB>2</SUB>O<SUB>3</SUB> passivation while sustaining the high performance of bulk MoS<SUB>2</SUB> FET. The MoS<SUB>2</SUB> FETs were exposed to O<SUB>2</SUB> plasma for 30 s before Al<SUB>2</SUB>O<SUB>3</SUB> encapsulation to achieve a relatively small hysteresis and high electrical performance. A MoO<I><SUB>x</SUB></I> layer formed during the plasma treatment was found between MoS<SUB>2</SUB> and the top passivation layer. The MoO<I><SUB>x</SUB></I> interlayer prevents the generation of excess electron carriers in the channel, owing to Al<SUB>2</SUB>O<SUB>3</SUB> passivation, thereby minimizing the shift in the threshold voltage (<I>V</I><SUB>th</SUB>) and increase of the off-current leakage. However, prolonged exposure of the MoS<SUB>2</SUB> surface to O<SUB>2</SUB> plasma (90 and 120 s) was found to introduce excess oxygen into the MoO<I><SUB>x</SUB></I> interlayer, leading to more pronounced hysteresis and a high off-current. The stable MoS<SUB>2</SUB> FETs were also subjected to gate-bias stress tests under different conditions. The MoS<SUB>2</SUB> transistors exhibited negligible decline in performance under positive bias stress, positive bias illumination stress, and negative bias stress, but large negative shifts in <I>V</I><SUB>th</SUB> were observed under negative bias illumination stress, which is attributed to the presence of sulfur vacancies. This simple approach can be applied to other transition metal dichalcogenide materials to understand their FET properties and reliability, and the resulting high-performance hysteresis-free MoS<SUB>2</SUB> transistors are expected to open up new opportunities for the development of sophisticated electronic applications.</P> [FIG OMISSION]</BR>

      • p38 MAPK Signaling Mediates Mitochondrial Apoptosis in Cancer Cells Induced by Oleanolic Acid

        Oleanolic acid (OA) is a nutritional component widely distributed in various vegetables. Although it has been well recognized for decades that OA exerts certain anti-tumor activity by inducing mitochondria-dependent apoptosis, it is still unclear that what molecular signaling is responsible for this effect. In this study, we employed cancer cell lines, A549, BXPC-3, PANC-1 and U2OS to elucidate the molecular mechanisms underlying OA anti-tumor activity. We found that activation of MAPK pathways, including p-38 MAPK, JNK and ERK, was triggered by OA in both a dose and time-dependent fashion in all the tested cancer cells. Activation was accompanied by cleavage of caspases and PARP as well as cytochrome C release. SB203580 (p38 MAPK inhibitor), but not SP600125 (JNK inhibitor) and U0126 (ERK inhibitor), rescued the pro-apoptotic effect of OA on A549 and BXPC-3 cells. OA induced p38 MAPK activation promoted mitochondrial translocation of Bax and Bim, and inhibited Bcl-2 function by enhancing their phosphorylation. OA can induce reactive oxygen species (ROS)-dependent ASK1 activation, and this event was indispensable for p38 MAPK-dependent apoptosis in cancer cells. In vivo, p38 MAPK knockdown A549 tumors proved resistant to the growth-inhibitory effect of OA. Collectively, we elucidated that activation of ROS/ASK1/p38 MAPK pathways is responsible for the apoptosis stimulated by OA in cancer cells. Our finding can contribute to a better understanding of molecular mechanisms underlying the antitumor activity of nutritional components.
