        Population genetics reveals new introgression in the nucleus herd of min pigs

        Objective Min pigs are a unique genetic resource among local pig breeds in China. They have more excellent characteristics in cold and stress resistance, good meat quality, and a high reproductive rate. However, the genetic structure and driving factors remain unclear in the nucleus herd. In this study, the genetic diversity of Min pigs was studied to reveal the formation mechanism of its unique genetic structure. We hope to protect and develop the genetic resources of Min pigs. Methods We analyzed different types of genes to identify the genetic structure and gene introgression pattern of Min pigs. The nuclear DNA dataset includes information on 21 microsatellite loci and 6 Y-chromosome genes, and the mitochondrial D-loop gene is selected to represent maternal lineages. The above genes are all from the nucleus herd of Min pigs. Results The results of genetic structure identification and analysis of potential exogenous gene introgression patterns indicate that the nucleus herd of Min pigs maintains a high level of genetic diversity (polymorphism information content = 0.713, expected heterozygosity = 0.662, observed heterozygosity = 0.612). Compared with other Asian pig breeds, the formation of Min pig breeds is more special. Gene introgression from European pig breeds to Min pigs has occurred, which is characterized by complete introgression of paternal genes and incomplete introgression of maternal genes. Conclusion Gene introgression caused by cross-breeding is not the main factor leading to the formation of the current genetic structure of Min pigs, but this process has increased the level of genetic diversity in the nucleus herd. Compared with the influence of gene introgression, our research suggest that artificial selection and environmental adaptive evolution make Min pigs form unique genetic characteristics.         Investigate the Effect of Potassium on Nodule Symbiosis and Uncover an HAK/KUP/KT Member, GmHAK5, Strongly Responsive to Root Nodulation in Soybean

        Leguminous plants form root nodule organs with soil rhizobia bacteria, which can fix atmospheric nitrogen (N2) and supply ammonia to the host plant. It is established that the symbiotic N fixation efficiency is substantially influenced by plant nutrients, such as molybdenum (Mo), phosphorus (P), copper (Cu), and zinc (Zn). Potassium (K+) is the most abundant cation in plant cells; however, little evidence regarding the potential link between K nutrition and symbiotic N fixation efficiency is available to date. Here, we showed that K+ deficiency reduced the efficiency of symbiotic nitrogen fixation, and inoculated with rhizobia strain USDA110 could improve plant K+ acquisition. Furthermore, we identified a potassium transporter gene (GmHAK5) that was highly expressed in the root steles and in the vasculature cells of nodules. The GmHAK5 protein was localized at the plasma membrane and could rescue the growth phenotype of yeast K+ uptake-defective mutant. The results obtained from this study provides new insights for the understanding of the potential role of K+ nutrition in modulating symbiotic N fixation in soybean.

        A Sensorless Rotor Position Estimation Scheme for IPMSM Using HF Signal Injection with Frequency and Amplitude Optimization

        High frequency signal injection (HFI) is an alternative method for estimating rotor position of interior permanent magnet synchronous motor (IPMSM). The general method of frequency and amplitude selection is based on error tolerance and experiments, and is usually set with only one group of HF parameters, which is not efficient for different working modes. This paper proposes a novel rotor position estimation scheme by HFI with optimized frequency and amplitude, based on the mathematic model of IPMSM. The requirements for standstill and low-speed operational modes are met by applying this novel scheme. Additionally, the effects of the frequency and amplitude of the injected HF signal on the position estimation results under different operating conditions are analyzed. Furthermore, an optimization method for HF parameter selection is proposed to make the estimation process more efficient under different working conditions according to error tolerance. The effectiveness of the propose scheme is verified by the experiments on an IPMSM motor prototype.

        A Novel Network Anomaly Detection Method based on Data Balancing and Recursive Feature Addition

        Network anomaly detection system plays an essential role in detecting network anomaly and ensuring network security. Anomaly detection system based machine learning has become an increasingly popular solution. However, due to the unbalance and high-dimension characteristics of network traffic, the existing methods unable to achieve the excellent performance of high accuracy and low false alarm rate. To address this problem, a new network anomaly detection method based on data balancing and recursive feature addition is proposed. Firstly, data balancing algorithm based on improved KNN outlier detection is designed to select part respective data on each category. Combination optimization about parameters of improved KNN outlier detection is implemented by genetic algorithm. Next, recursive feature addition algorithm based on correlation analysis is proposed to select effective features, in which a cross contingency test is utilized to analyze correlation and obtain a features subset with a strong correlation. Then, random forests model is as the classification model to detection anomaly. Finally, the proposed algorithm is evaluated on benchmark datasets KDD Cup 1999 and UNSW_NB15. The result illustrates the proposed strategies enhance accuracy and recall, and decrease the false alarm rate. Compared with other algorithms, this algorithm still achieves significant effects, especially recall in the small category.

      • Necessary Experimental Conditions for Single-Shot Diffraction Imaging of DNA-Based Structures with X-ray Free-Electron Lasers

        <P>It has been proposed that the radiation damage to biological particles and soft condensed matter can be overcome by ultrafast and ultraintense X-ray free-electron lasers (FELs) with short pulse durations. The successful demonstration of the “diffraction-before-destruction” concept has made single-shot diffraction imaging a promising tool to achieve high resolutions under the native states of samples. However, the resolution is still limited because of the low signal-to-noise ratio, especially for biological specimens such as cells, viruses, and macromolecular particles. Here, we present a demonstration single-shot diffraction imaging experiment of DNA-based structures at SPring-8 Angstrom Compact Free Electron Laser (SACLA), Japan. Through quantitative analysis of the reconstructed images, the scattering abilities of gold and DNA were demonstrated. Suggestions for extracting valid DNA signals from noisy diffraction patterns were also explained and outlined. To sketch out the necessary experimental conditions for the 3D imaging of DNA origami or DNA macromolecular particles, we carried out numerical simulations with practical detector noise and experimental geometry using the Linac Coherent Light Source (LCLS) at the SLAC National Accelerator Laboratory, USA. The simulated results demonstrate that it is possible to capture images of DNA-based structures at high resolutions with the technique development of current and next-generation X-ray FEL facilities.</P> [FIG OMISSION]</BR>
