We consider a reaction-diffusion system which describes the dynamics of the predator-prey interaction over a spatial bounded domain. We study the travelling wave solutions of the Lotka-Volterra equations with weak Allee effect type growth, in which the prey’s growth rate has an autocatalytic effect. It is known as Holling Type II predator functional response. The existence of travelling wave solutions is shown by means of bifurcation theorem. The stability of travelling wave solutions is established with singularly perturbed reaction-diffusion equations. The methods involve phase space investigation, invariant manifold and spectrum analysis. Numerical examples are provided to verify the main theoretical results.
Valproic acid enforces the priming effect of sphingosine-1 phosphate on human mesenchymal stem cells

<P>Engraftment and homing of mesenchymal stem cells (MSCs) are modulated by priming factors including the bioactive lipid sphingosine-1-phosphate (S1P), by stimulating CXCR4 receptor signaling cascades. However, limited <I>in vivo</I> efficacy and the remaining priming molecules prior to administration of MSCs can provoke concerns regarding the efficiency and safety of MSC priming. Here, we showed that valproic acid (VPA), a histone deacetylase inhibitor, enforced the priming effect of S1P at a low dosage for human umbilical cord-derived MSCs (UC-MSCs). A DNA-methylation inhibitor, 5-azacytidine (5-Aza), and VPA increased the expression of <I>CXCR4</I> in UC-MSCs. In particular, UC-MSCs primed with a suboptimal dose (50 nM) of S1P in combination with 0.5 mM VPA (VPA+S1P priming), but not 1 <I>µ</I>M 5-Aza, significantly improved the migration activity in response to stromal cell-derived factor 1 (SDF-1) concomitant with the activation of both MAPK<SUP>p42/44</SUP> and AKT signaling cascades. Both epigenetic regulatory compounds had little influence on cell surface marker phenotypes and the multi-potency of UC-MSCs. In contrast, VPA+S1P priming of UC-MSCs potentiated the proliferation, colony forming unit-fibroblast, and anti-inflammatory activities, which were severely inhibited in the case of 5-Aza treatment. Accordingly, the VPA+S1P-primed UC-MSCs exhibited upregulation of a subset of genes related to stem cell migration and anti-inflammation response. Thus, the present study demonstrated that VPA enables MSC priming with S1P at a low dosage by enhancing their migration and other therapeutic beneficial activities. This priming strategy for MSCs may provide a more efficient and safe application of MSCs for treating a variety of intractable disorders.</P>

<P><I>Context:</I> A growing body of evidence shows that compounds of plant origin have the ability to prevent cancer. The fruit of gardenia, <I>Gardenia jasminoides</I> Ellis (Rubiaceae), has long been used as a food additive and herbal medicine, and its pharmacological actions, such as protective activity against oxidative damage, cytotoxic effect, and anti-inflammatory and anti-tumor activity, have already been reported.</P><P><I>Objective:</I> The purpose of the present study was to investigate the presence of DNA topoisomerase 1 inhibitor in various solvent fractions of <I>Gardenia</I> extract and examine the induction of oral cancer cell death upon treatment with <I>Gardenia</I> extract.</P><P><I>Materials and methods:</I> The methanol extract of <I>Gardenia</I> was partitioned with <I>n-</I>hexane, dichloromethane, ethyl acetate, <I>n-</I>butanol, and water.</P><P><I>Results:</I> In the DNA topoisomerase 1 assay, <I>n-</I>hexane and dichloromethane fractions inhibited topoisomerase 1 and led to a decrease in the cell viability of KB cells. The dichloromethane fraction (0.1 mg/mL) also showed 77% inhibition of cell viability in KB cells compared with HaCaT cells. Treatment with dichloromethane fraction led to apoptotic cell death as evidenced by flow cytometric analysis and morphological changes. In addition, treatment with <I>Gardenia</I> extract dichloromethane fraction led to the partial increase of caspase-3, caspase-8 and caspase-9 activities and the cleavage of poly (ADP-ribose) polymerase.</P><P><I>Conclusion:</I> Taken together, these results suggest that the dichloromethane fraction from <I>Gardenia</I> extract induces apoptotic cell death by DNA topoisomerase 1 inhibition in KB cells. These findings suggest the possibility that <I>Gardenia</I> extract could be developed as an anticancer modality.</P>

Transplantation of stem cells, such as mesenchymal stem cells (MSCs), is a promising strategy for treating several types of intractable disorders. Mechanistically, it could not only replace damaged cells by direct contribution, but also establish an anti-inflammatory or immunomodulatory microenvironment. However, the cellular mechanisms underlying molecular and biological properties of stem cells during ex vivo expansion and also after transplantation in pathological environments remain largely elusive. We recently developed the cyanoacrylamide-based coumarin derivatives (Fluorescent real-time thiol tracer; FreSHtracer*) reversibly react with glutathione for monitoring of glutathione levels in living stem cells. These probes revealed that glutathione levels are heterogeneous among subcellular organelles and among individual cells and show dynamic changes and heterogeneity in repopulating stem cells depending on oxidative-stress or culture conditions. Importantly, a subpopulation of stem cells with high-glutathione levels exhibited increased self-renewal and migration activities in vitro and showed improved therapeutic efficiency in treating asthma. Furthermore, employing a novel combination of longitudinal intravital confocal fluorescence imaging and microcystoscopy in living animals, we investigated the distributions and properties of transplanted multipotent MSCs derived from human embryonic stem cells at single-cell resolution in real-time by performing confocal imaging of bladder tissues in a rat model of IC/BPS for up to 6 months post-transplantation. These novel real-time monitoring strategies demonstrate the novel molecular insight for maintaining stem cell functions and also enhance understanding of the in vivo behaviors of the engrafted stem cells, which is crucial to determine the efficacy and safety of stem cell-based therapies. This strategy may facilitate the translation of various stem cell-based approaches into clinical practice.

<P><B>Summary</B></P> <P><B>Background</B></P> <P>This study aimed to assess trends of underweight, obesity, and abdominal obesity and related lifestyle factors in young Korean adults.</P> <P><B>Methods</B></P> <P>The analysis focused on adults aged 19–39 years using data from the Korea National Health and Nutrition Examination Survey (KNHANES) I–V (1998–2012). Underweight was defined as body mass index (BMI) <18.5kg/m<SUP>2</SUP>; obesity as BMI≥30kg/m<SUP>2</SUP>; and abdominal obesity as waist circumference ≥90cm in men and ≥85cm in women. We evaluated trends by analyzing the prevalence and odds ratios (ORs) of underweight, obesity, abdominal obesity, and the proportions and ORs of lifestyles.</P> <P><B>Results</B></P> <P>As compared with KNHANES I, the ORs for obesity were higher in III, IV, and V and the ORs for abdominal obesity were higher in IV and V in men, which showed an increasing trend (P for trend<0.001). In women, the prevalence of underweight and obesity showed a significant increasing tendency (P for trend<0.001). Lifestyle analysis revealed the overconsumption of energy intake proportion was increased in men and decreased in women. The lack of physical activity and high-risk alcohol consumption proportions showed increasing trends in both men and women. The current smoking proportion was decreased in men and increased in women. The lack of adequate sleep proportion showed a continuous increasing trend in men.</P> <P><B>Conclusion</B></P> <P>Prevention of obesity in young men and women, and abdominal obesity in young men is needed. Meanwhile, management of underweight in young women is simultaneously required.</P>
MODELING AN EXCITATORY SYNAPTIC TRANSMISSION IN THE CENTRAL NERVOUS SYSTEM

We have developed mathematical models of central excitatory synapse, including kinetics of neurotransmitter release in a hippocampal neurons in the mammalian central nervous system. Glutamate is a primary neurotransmitter in the CNS and it is important to understand the mechanism of glutamate activation and related regulation properties. Experimentally, it is impossible to have variation of released glutamate from nerve terminals. Aiming at investigation of fundamental issues unsolved, we discuss dynamics of glutamate release, involving with glutamate receptors, such as AMPARs, and glutamate optical sensor. Our mathematical formulation is shown to be effective for calculating certain kinetics related to glutamate release.
