Therapeutic Benefits of Mesenchymal Stromal Cells in a Rat Model of Hemoglobin-Induced Hypertensive Intracerebral Hemorrhage

        Ding, Rui,Lin, Chunnan,Wei, ShanShan,Zhang, Naichong,Tang, Liangang,Lin, Yumao,Chen, Zhijun,Xie, Teng,Chen, XiaoWei,Feng, Yu,Wu, LiHua Korean Society for Molecular and Cellular Biology 2017 Molecules and cells Vol.40 No.2

Previous studies have shown that bone marrow mesenchymal stromal cell (MSC) transplantation significantly improves the recovery of neurological function in a rat model of intracerebral hemorrhage. Potential repair mechanisms involve anti-inflammation, anti-apoptosis and angiogenesis. However, few studies have focused on the effects of MSCs on inducible nitric oxide synthase (iNOS) expression and subsequent peroxynitrite formation after hypertensive intracerebral hemorrhage (HICH). In this study, MSCs were transplanted intracerebrally into rats 6 hours after HICH. The modified neurological severity score and the modified limb placing test were used to measure behavioral outcomes. Blood-brain barrier disruption and neuronal loss were measured by zonula occludens-1 (ZO-1) and neuronal nucleus (NeuN) expression, respectively. Concomitant edema formation was evaluated by H&E staining and brain water content. The effect of MSCs treatment on neuroinflammation was analyzed by immunohistochemical analysis or polymerase chain reaction of CD68, Iba1, iNOS expression and subsequent peroxynitrite formation, and by an enzyme-linked immunosorbent assay of pro-inflammatory factors (IL-$1{\beta}$ and TNF-${\alpha}$). The MSCs-treated HICH group showed better performance on behavioral scores and lower brain water content compared to controls. Moreover, the MSC injection increased NeuN and ZO-1 expression measured by immunochemistry/immunofluorescence. Furthermore, MSCs reduced not only levels of CD68, Iba1 and pro-inflammatory factors, but it also inhibited iNOS expression and peroxynitrite formation in perihematomal regions. The results suggest that intracerebral administration of MSCs accelerates neurological function recovery in HICH rats. This may result from the ability of MSCs to suppress inflammation, at least in part, by inhibiting iNOS expression and subsequent peroxynitrite formation.

Experimental Investigation on Damage Behavior of Polypropylene Fiber Reinforced Concrete under Compression

        Lihua Xu,Biao Li,Xiaoxiao Ding,Yin Chi,Changning Li,Biao Huang,Yuchuan Shi 한국콘크리트학회 2018 International Journal of Concrete Structures and M Vol.12 No.7

This paper presents an experimental investigation on the stress–strain behavior and the damage mechanism of polypropylene fiber reinforced concrete (PFRC) under monotonic and cyclic compression. Fifty-four specimens for different fiber volume fractions and aspect ratios were tested. Acoustic emission (AE) technique was used to monitor the damage progression. The damage mechanism of concrete was analyzed based on the AE parametric analysis. The results show that the incorporation of polypropylene fiber (PF) has a positive effect on the monotonic and cyclic behaviors of concrete, especially for the post-cracking branch. The toughness and ultimate strain are enhanced and the performance degradation in terms of elastic stiffness and strength is alleviated by the addition of PF. However, PF has little influences on the plastic strain, and the damage process of concrete is mainly driven by the envelope strain. The effect of fiber volume fraction on the cyclic behavior of concrete shows more pronounced than that of aspect ratio. In addition, it is found from AE results that the damage, closely related to AE events, has a quick evolution just after the peak stress, with the AE hits having a concentrated release. The total amount of AE hits increases with increasing fiber volume fraction due to fiber pullout and sliding, while the concrete with fiber aspect ratio of 280 reaches the largest amount. Meanwhile, as substantiated by AE, the failure of PFRC shows an obvious shear mode, with shear cracks dominating the damage progression. Finally, a damage elasto-plastic model is developed to predict the monotonic and cyclic responses of PFRC and the prediction yields a fairly close estimation with experimental results.

Effects of Phytosterol Ester on the Fatty Acid Profiles in Rats with Nonalcoholic Fatty Liver Disease

        Lihua Song,Haiyue Zhou,Wenjuan Yu,Xinwen Ding,Li Yang,Jiayi Wu,Chenwei Song 한국식품영양과학회 2020 Journal of medicinal food Vol.23 No.2

Both serum and hepatic fatty acid (FA) compositions differ among nonalcoholic hepatic steatosis, nonalcoholic steatohepatitis, and healthy subjects. The severity of the above liver disease is closely associated with the concentration and composition of FAs. Our previous study found that phytosterol ester (PSE) could alleviate hepatic steatosis in nonalcoholic fatty liver disease rats. The aims of this work were to explore the effects of PSE (0.05/100 g·body weight) on FA profiles and the mRNA levels of FA metabolism-related genes. Compared with a high-fat diet alone group, PSE treatment significantly decreased hepatic saturated fatty acid levels (P < .05) and increased monounsaturated fatty acid (especially C16:1 n-7) levels in the liver, serum, and adipose tissue and polyunsaturated fatty acid levels in the serum and liver (P < .05) after 12 weeks of intervention. In particular, PSE treatment increased the level of C22:5 n-3, an FA that was negatively correlated with the degree of hepatic steatosis in the serum, liver, and adipose tissue. The increases in some unsaturated fatty acids are probably related to the upregulation of stearoyl-coenzyme A desaturase-1 and fatty acid desaturase-1.

Potential repair mechanisms involve anti-inflammation, anti-apoptosis and angiogenesis. However, few studies have focused on the effects of MSCs on inducible nitric oxide synthase (iNOS) expression and subsequent per-oxynitrite formation after hypertensive intracerebral hemorrhage (HICH). In this study, MSCs were transplanted intracerebrally into rats 6 hours after HICH. Blood–brain barrier disruption and neuronal loss were measured by zonula occludens-1 (ZO-1) and neuronal nucleus (NeuN) expression, respectively. The effect of MSCs treatment on neuroinflammation was analyzed by immunohistochemical analysis or polymerase chain reaction of CD68, Iba1, iNOS expression and subsequent peroxynitrite formation, and by an enzyme-linked immunosorbent assay of pro-inflammatory factors (IL-1 and TNF-). Microstructures and Thermal Properties of Polycaprolactone/Epoxy Resin/SiO2 Hybrids

He, Lihua,Liu, Pinggui,Ding, Heyan The Society of Adhesion and Interface 2006 접착 및 계면 Vol.7 No.4

A series of organic-inorganic hybrids, PCL/EP/$SiO_2$, involving epoxy resin and triethoxysilane-terminated polycaprolactone elastomer (PCL-TESi) were prepared via polymerization of diglycidyl ether of bisphenol A (DGEBA) with amine curing agent KB-2 and sol-gel process of PCL-TESi. The curing reactions were started from the initially homogeneous mixture of DGEBA, KB-2 and the PCL-TESi. The organicinorganic hybrids containing up to 4.95% (wt) of $SiO_2$ were obtained and characterized by FT-IR, transmission electron microscopy (TEM), differential scanning calorimetry (DSC) and thermogravimetry analysis (TGA). It was experimentally shown that the swelling property in toluene, morphologies and thermal properties of the resulting hybrids were quite dependent on the contents of $SiO_2$. The crosslink network density decreases with increasing of the PCL-TESi. And in TEM, the phase separated morphology of these hybrids was found, which resulted from the coagulation of Si-O-Si networks resulting from $-Si(OC_2H_5)_3$ of PCL-TESi self-curing by hydrolytic silanol condensation, with the advancement of the curing reaction in the modified epoxy resin systems. Meanwhile, the change of the $SiO_2$ content made the morphologies changed from aggregated particles of Si-O-Si in the hybrid to nanocluster of interconnected Si-O-Si particles, then to aggregated Si-O-Si dispersing in the continuous cured epoxy phase again, and last to co-continuous interpenetrating network. The glass transition behavior of the hybrid material was cooperative motion of large chain segments, which were hindered by the inorganic Si-O-Si network. And in TG analysis, the characteristic temperature at 5% of weight loss was evidently increased from $120.5^{\circ}C$ of pure cured epoxy to $277.6^{\circ}C$ of 3.84% (wt) of $SiO_2$ modified epoxy due to the existence of Si-O-Si when PCL-TESi was added in the hybrid.

Microvascular Myocardial Ischemia in Patients With Diabetes Without Obstructive Coronary Stenosis and Its Association With Angina

        Yu Yarong,Yang Wenli,Dai Xu,Yu Lihua,Lan Ziting,Ding Xiaoying,Zhang Jiayin 대한영상의학회 2023 Korean Journal of Radiology Vol.24 No.11

Objective: To investigate the incidence of microvascular myocardial ischemia in diabetic patients without obstructive coronary artery disease (CAD) and its relationship with angina. Materials and Methods: Diabetic patients and an intermediate-to-high pretest probability of CAD were prospectively enrolled. Non-diabetic patients but with an intermediate-to-high pretest probability of CAD were retrospectively included as controls. The patients underwent dynamic computed tomography-myocardial perfusion imaging (CT-MPI) and coronary computed tomography angiography (CCTA) to quantify coronary stenosis, myocardial blood flow (MBF), and extracellular volume (ECV). The proportion of patients with microvascular myocardial ischemia, defined as any myocardial segment with a mean MBF ≤ of 100 mL/min/100 mL, in patients without obstructive CAD (Coronary Artery Disease–Reporting and Data System [CAD-RADS] grade 0–2 on CCTA) was determined. Various quantitative parameters of the patients with and without diabetes without obstructive CAD were compared. Multivariable analysis was used to determine the association between microvascular myocardial ischemia and angina symptoms in diabetic patients without obstructive CAD. Results: One hundred and fifty-two diabetic patients (mean age: 59.7 ± 10.7; 77 males) and 266 non-diabetic patients (62.0 ± 12.3; 167 males) were enrolled; CCTA revealed 113 and 155 patients without obstructive CAD, respectively. For patients without obstructive CAD, the mean global MBF was significantly lower for those with diabetes than for those without (152.8 mL/min/100 mL vs. 170.4 mL/min/100 mL, P < 0.001). The mean ECV was significantly higher for diabetic patients (27.2% vs. 25.8%, P = 0.009). Among the patients without obstructive CAD, the incidence of microvascular myocardial ischemia (36.3% [41/113] vs. 10.3% [16/155], P < 0.001) and interstitial fibrosis (69.9% [79/113] vs. 33.3% [8/24], P = 0.001) were significantly higher in diabetic patients than in the controls. The presence of microvascular myocardial ischemia was independently associated with angina symptoms (adjusted odds ratio = 3.439, P = 0.037) in diabetic patients but without obstructive CAD. Conclusion: Dynamic CT-MPI + CCTA revealed a high incidence of microvascular myocardial ischemia in diabetic patients without obstructive CAD. Microvascular myocardial ischemia is strongly associated with angina.

Intelligent On-demand Routing Protocol for Ad Hoc Network

        Yongfei Ye,Xinghua Sun,Minghe Liu,Jing Mi,Ting Yan,Lihua Ding 한국정보처리학회 2020 Journal of information processing systems Vol.16 No.5

Ad hoc networks play an important role in mobile communications, and the performance of nodes has asignificant impact on the choice of communication links. To ensure efficient and secure data forwarding anddelivery, an intelligent routing protocol (IAODV) based on learning method is constructed. Five attributes ofnode energy, rate, credit value, computing power and transmission distance are taken as the basis ofsegmentation. By learning the selected samples and calculating the information gain of each attribute, thedecision tree of routing node is constructed, and the rules of routing node selection are determined. IAODValgorithm realizes the adaptive evaluation and classification of network nodes, so as to determine the optimaltransmission path from the source node to the destination node. The simulation results verify the feasibility, effectiveness and security of IAODV.
