        Integrin α5β1-Ang1/Tie2 receptor cross-talk regulates brain endothelial cell responses following cerebral ischemia

        We have previously demonstrated that in response to cerebral ischemia (CI), the growth factor angiopoietin-1 (Ang1) and α5β1 integrin are both induced in cerebral vessels, which likely provide positive signals driving the endogenous angiogenic response and vascular protection after CI. However, the precise relationship between endothelial Ang1 and α5β1 integrin after CI remains poorly understood. Here, we investigated the effects of the interaction between the Ang1/Tie2 system and α5β1 integrin on brain endothelial cells (BECs) under cerebral ischemic conditions in vivo and in vitro. Immunofluorescence analysis demonstrated that integrin α5β1 co-localized with Tie2/phosphorylated Tie2 on cerebral vessels in the penumbra. The in vitro study showed that oxygen–glucose deprivation/restoration (OGD/R) induced the expression of the Ang1 receptor Tie2 on BECs in a manner similar to that for integrin α5 and Ang1 in response to OGD/R, accompanied by increased activation of Tie2 and its downstream effectors focal adhesion kinase (FAK) and Akt. Knockdown of α5 integrin markedly suppressed OGD/R-induced Tie2 receptor activation in BECs, while in contrast, priming BECs with Ang1 promoted the expression of α5 integrin as well as the Tie2 downstream transcription factor Ets-1 in OGD-treated BECs. In line with this, Ets-1 knockdown significantly attenuated Ang1- mediated upregulation of α5 integrin. Functionally, Ang1 induced cell migration and tube formation of BECs after OGD, but this effect was inhibited by diminishment of the levels of α5 integrin in BECs. Taken together, our data indicate that the Ang1/Tie2 system cross-talks with integrin α5β1 in BECs after CI, which may contribute to the endogenous angiogenic vascular protective response following CI.

        Description of the whole mitochondrial genome of Bhatia longiradiata (Hemiptera, Cicadellidae, Deltocephalinae: Drabescini) and phylogenetic relationship

        Background Mitochondrial genomes are extremely conserved in genetic processes and valuable molecular indications for phylogenetic and evolutionary examination, but the mitochondrial genome of Bhatia has not yet been reported. Objective The target of this writing was to clarify the structural module of the mitochondrial genes of Bhatia longiradiata, verify the monophyletic of Drabescini, and explore the phylogenetic relationship between Drabescini with other leafhoppers. Methods We performed sequencing and explanatory note of the mitochondrion of Bhatia longiradiata. The phylogeny relation was created by ML and Bayesian approaches using three dissimilar datasets (PCG12, PCG12rRNA, and AA), which were constructed to discuss the phylogenetic status of Bhatia longiradiata. Results To report the architectural feature of the chondriosome of Bhatia longiradiata is a seal double-stranded annular molecule with 16,122 bp measurement and cover typically 37 genes. Several tandem repetitive units were observed in an AT enrichment area. The analysis showed that the branching relationships among the six trees were generally consistent, and each of the subfamilies was individually clustered into a monophyletic group within Cicadellidae. Bhatia longiradiata and other members of the Drabescini were aggregated into a clade that was situated within the Deltocephalinae. Conclusion The mitochondrial genome of Bhatia longiradiata covers 37 typical genes and a control region, which covers six tandem repeats. All species of Drabescini procedure a clade within Deltocephalinae. Drabescini and Scaphoideini form a branch and show a sister relationship with strong support. Therefore, we support the relegation of Selenocephalinae to a clan within Deltocephalinae.

        The physicochemical, antioxidant, hypoglycemic and prebiotic properties of γ-irradiated polysaccharides extracted from Lentinula edodes

        In this study, the influence of γ-irradiation with different dose (0, 4, 8, and 16 kGy) on chemical composition, physicochemical features and bioactivities of polysaccharides extracted from Lentinula edodes (LEP) were investigated. The carbohydrate content (from 59.47 to 70.96%), the solubility, the ⋅OH and DPPH scavenging ability of LEP increased with the increased γ-irradiation dose, while the protein content, the weight-average and number-average molecular weight of LEP were significantly decreased with the increased γ-irradiation dose. Moreover, γ-irradiation treatment caused LEP color changes and surface topography destroyed. γ-Irradiated LEP showed higher hypoglycemic activities in vitro than that of non-irradiated LEP. Moreover, γ-irradiated LEP had better proliferation promoting effects on Lactobacillus rhamnosus and L. plantarum. These results showed that γ-irradiation treatment changes the physicochemical features of LEP, thus affects its antioxidant, hypoglycemic and prebiotic properties, which suggests that γ-irradiated LEP has potential application in the pharmaceutical industries and functional foods.
