A sensitive silver nanoparticles (Ag NPs) enhanced chemiluminescence (CL) method is reported for the determination of mandelic acid (MA). This method is based on the luminol-KIO4 system catalyzed by Ag NPs to produce CL spectra. Prepared Ag NPs were characterized by UV-visible spectra and TEM image. Under optimal condition, CL spectra of the system were responded linearly with the concentration of MA in the range of 2.5×10(-9) to 2.0×10(-8) mol L(-1)(r=0.9989) with a detection limit of 1.2×10(-10) mol L(-1). The relative standard deviation of 1.0×10(-7) mol L(-1) MA was found 1.45 (n=9).

본 논문은 이미지 탐지 모델인 Faster R-CNN 을 통해 영유아가 존재하는 어린이 집, 공원, 놀이터, 거실 등의 2D 이미지를 읽어 영유아에게 위험이 되는 요소를 인식해 위험상황을 감지하는 시스템을 구현하였다. 실생활에서 쉽게 구할 수 있는 데이터를 바탕으로 탐지 모델을 구현 했으며 현재 머신 러닝 분야가 음성인식과 행위데이터를 기반으로 상용화 되어있는 반면 본 모델은 이미지를 데이터로 한 탐지 모델이 다양한 서비스 분야에서 활용 될 수 있음을 보여준다.

<P>A new cell culture technology enables highly tumorigenic 3D spheroids to be easily generated from various cancer cell sources in the common laboratory.</P><P><B></B></P><P>Although cancer stem cells (CSC) are thought to be responsible for tumor recurrence and resistance to chemotherapy, CSC-related research and drug development have been hampered by the limited supply of diverse, patient-derived CSC. Here, we present a functional polymer thin film (PTF) platform that promotes conversion of cancer cells to highly tumorigenic three-dimensional (3D) spheroids without the use of biochemical or genetic manipulations. Culturing various human cancer cells on the specific PTF, poly(2,4,6,8-tetravinyl-2,4,6,8-tetramethyl cyclotetrasiloxane) (pV4D4), gave rise to numerous multicellular tumor spheroids within 24 hours with high efficiency and reproducibility. Cancer cells in the resulting spheroids showed a significant increase in the expression of CSC-associated genes and acquired increased drug resistance compared with two-dimensional monolayer-cultured controls. These spheroids also exhibited enhanced xenograft tumor-forming ability and metastatic capacity in nude mice. By enabling the generation of tumorigenic spheroids from diverse cancer cells, the surface platform described here harbors the potential to contribute to CSC-related basic research and drug development.</P><P><B>Significance:</B></P><P>A new cell culture technology enables highly tumorigenic 3D spheroids to be easily generated from various cancer cell sources in the common laboratory.</P>

Surfactant enhanced fluorescence (FL) of europium-salicylic acid (SA) complex has been studied. It was observed that weak FL of Eu(III) at the wavelength of 589 nm and 612 nm was found to be enhanced after addition of salicylic acid and cetyltrimethylammonium bromide (CTAB) surfactant upon excitation at 395 nm. Under optimized condition, FL intensity of Eu(III) at 612 nm responded linearly with the concentration of SA in the range of 5.5×10(-9) to 1.5×10(-6) M of SA. The detection limit was calculated from the calibration curve(3Sb/m) as 9.27×10(-9)M.
HaCaT 세포에서 회향 열매의 피부장벽기능과 hyaluronic acid 생성에 미치는 영향

회향은 미나리과에 속하는 다년생식물인 Foeniculum vulgare Mill.의 성숙한 과실로서 항염, 진통, 피부노화방지 등의 효과를 가지고 있어 다양한 질환에 우수한 치료효과를 나타내는 것으로 알려져 왔다. 따라서 본 연구에서는 기능성이 우수한 회향 열매 추출물을 이용하여 피부 질환 치료제 및 피부장벽 기능 개선 소재로서의 적용가능성을 확인하였다. 이를 위해 각질형성세포주인 HaCaT에 회향 열매 메탄올 추출물과 그 분획물(hexane, methyl chloride, ethyl acetate, butanol)을 처리한 후, involucrin, loricrin, filaggrin의 발현과 hyaluronic acid의 생성 및 β-defensin -1, -2, -3, LL-37의 mRNA 발현을 측정하였다. 그 결과butanol 분획물 50 μg/ml을 24시간 동안 처리시 involucrin과 filaggrin단백질 발현을 각각122.8%, 105%로 유의하게 증가시킴을 확인하였다. Elisa assay를 통해 분석한 결과, ethyl acetate와 butanol 분획물은 대조군에 비해 hyaluronic acid의 생성을 각각17%, 11% 증가시켰으며, 이는 hyaluronic acid synthesis-1의 mRNA 발현 증가에 의한 것임을 확인하였다. 그러나 회향 열매 메탄올추출물과 분획물 모두에서 β-defensin -1, -2, -3, LL-37의 mRNA 발현은 증가되지 않았다. 이러한 결과는 회향열매 butanol 분획물이 각질형성세포에서 물리적 장벽을 형성하고 보습인자 조절을 통해 피부장벽 기능을 강화하는데 효과적임을 의미한다. Foeniculum vulgare (FV) has long been used in traditional medicine for the treatment of inflammatory diseases. In addition, it is usually known as an important medicinal and aromatic plant widely used as a carminative, digestive, lactogogue, and diuretic, and for treating respiratory and gastrointestinal disorders. The skin barrier protects against the invasion of pathogens, fends off chemical and physical assaults, and protects against extensive water loss. In this study, the effects of solvent-fractionated FV fruits on strengthening the skin barrier and maintaining moisture, as well as their antifungal activity, were investigated in human keratinocyte (HaCaT) cells. The expression of involucrin, loricrin, filaggrin, hyaluronic acid synthase, human β defensin, and cathelicidin genes and proteins was measured by reverse transcription polymerase chain reaction (RT-PCR) and western blotting. The production of hyaluronic acid was determined by enzyme-linked immunosorbent assay (ELISA). The butanol fraction increased the expression of involucrin and filaggrin. Both the ethyl acetate and the butanol fractions increased hyaluronic acid production by promoting the expression of hyaluronic acid synthase-1. Although the antimicrobial peptides were increased by FV crude extract and its fractions, the samples did not show a significant effect compared to the normal group. These results suggest that the butanol fraction of FV could be very useful in cosmetics for the treatment of dermatological diseases.
Europium(3)와 EDTA의 3차 복합체 형성에 의한 Levodopa의 분광분석적 정량

A highly sensitive spectrofluorimetric method was developed for the determination of levodopa based on the formation of a ternary complex with Eu3+ in the presence of ethylenediaminetetraacetic acid. It was found that this complex manifests intense fluorescence at 591 and 613 nm with excitation at 372 nm and maximum intensity was obtained at 613nm. Under the optimum conditions, the enhanced fluorescence intensity was proportional to the concentration of levodopa over the range of 3×10(-9)-2.5×10(-7) mol L(-1) with limit of detection (LOD) of 4.37×10(-10) mol L(-1). The method offers higher sensitivity and selectivity which could be effectively applied for the determination of levodopa in pharmaceutical and biological samples.

A rapid flow injection chemiluminescence method (CL-FIA) is proposed for the determination of folic acid (FA). The method is based on the intensified CL intensity of Ce(IV)-Na2S2O4-FA system catalyzed by gold nanoparticles (Au NPs). The CL intensity of the Ce(IV)-Na2S2O4-FA-Au NPs system was increased with the concentration of FA in the linear range of 2.3×10(-9)-8.5×10(-7) mol L(-1) with correlation coefficient of 0.9992. The limit of detection was found to be 2.7×10(-10) mol L(-1). The method affords good accuracy with high sensitivity and selectivity which can be applied successfully for the determination of folic acid in pharmaceutical and biological samples.

Apolipophorin-III (apoLp-III) was isolated from the last instar larval hemolymph of Spodoptera litura by cation exchange chromatography and reverse phase chromatography. Molecular mass of apoLp-III was estimated to be 18,266 daltons. Presence and exact localization of apoLp-III in both ovary and testis were identified by immunological analysis. In the earliest stage of ovary, apoLp-III is confined to the electron dense granules in the sheath cells between oogonia. During vitellogenesis, apoLp-III in the hemolymph is actively taken up into ovary through intercellular spaces between follicle cells and then stored in protein bodies in the ooplasm. After apoLp-III is taken up from the hemolymph into the basal portion of the vas deference, it is localized to electron dense granules on the apical surface of the vas deference epithelium. In in vitro culture study, after apoLp-III is actively synthesized in adult fat body, most of the synthesized apoLp -III is secreted to the medium.
